Book excerpt: The importance of chloride ions in cell physiology has not been fully recognized until recently, in spite of the fact that chloride (Cl-), together with bicarbonate, is the most abundant free anion in animal cells, and performs or determines fundamental biological functions in all tissues. For many years it was thought that Cl- was distributed in thermodynamic equilibrium across the plasma membrane of most cells. Research carried out during the last couple of decades has led to a dramatic change in this simplistic view. We now know that most animal cells, neurons included, exhibit a non-equilibrium distribution of Cl- across their plasma membranes. Over the last 10 to 15 years, with the growth of molecular biology and the advent of new optical methods, an enormous amount of exciting new information has become available on the molecular structure and function of Cl- channels and carriers. In nerve cells, Cl- channels and carriers play key functional roles in GABA- and glycine-mediated synaptic inhibition, neuronal growth and development, extracellular potassium scavenging, sensory-transduction, neurotransmitter uptake and cell volume control. Not so very many years ago, a pamphlet rather than book might have resulted from such an endeavor! One might ask why Cl-, the most abundant biological anion, attracted so little attention from investigators. The main reason was that the prevailing paradigm for cellular ion homeostasis in the 1950s and 1960s assigned Cl- a ther modynamically passive and unspecialized role. This view was particularly prominent among muscle and neuroscience investigators. In searching for reasons for such a negative (no pun intended) viewpoint, it seems to us that it stemmed from two key experimental observations. First, work on frog skeletal muscle showed that Cl- was passively distributed between the cytoplasm and the extracellular fluid. Second, work on Cl- transport in red blood cells confirmed that the Cl- transmembrane distribution was thermodynamically passive and, in addition, showed that Cl- crossed the mem brane extremely rapidly. This latter finding [for a long time interpreted as being the result of a high passive chloride electrical permeability(? CI)] made it quite likely that Cl- would remain at thermodynamic equilibrium. These two observations were gener alized and virtually all cells were thought to have a very high P Cl and a ther modynamically passive Cl- transmembrane distribution. These concepts can still be found in some physiology and neuroscience textbooks.

Book excerpt: Ion channels and transporters provide pathways through which otherwise impermeable ions and nutrients can cross cell membranes. Among these protein machines are a variety of chloride channels, which are activated by different stimuli and mechanisms, participating in a wide-spectrum of physiological processes. These Cl− channels are permeable to many anions although predominantly Cl− , due to its abundance, providing the substrate and electrical charge important for epithelial physiology as well as neuronal and muscular excitability. The function of these channels is tightly regulated in cells by a number of factors such as divalent cations, pH and even the permeable anions themselves. Studying the modulation mechanisms of these factors in channel function provides not only a tool for understanding the roles of these channels in the physiological and pathological conditions but also a structural mapping for channel activities. My dissertation examines the modulation of TMEM16A, a calcium-activated chloride channel, as well as that of CLC Cl− channels. When studying TMEM16A with excised inside-out patches, we find that neither the recombinant bovine calmodulin (CaM) nor the purified human brain CaM alters the permeability of HCO3− in mouse TMEM16A. We suspect that the contradictory studies, which report that CaM affects the anion permeability of TMEM16A using whole-cell recordings, may be compromised by a series resistance problem and/or ion accumulation effect in whole-cell recording when measuring the bi-ionic potentials. On the other hand, the function of CLC Cl− channels can be modulated in multiple ways. Apart from the well-characterized voltage-regulation, we find that beta-nicotinamide adenine dinucleotide (NAD), when applied intracellularly, inhibits WT CLC-1 mainly by altering its slow gating. This inhibitory effect of NAD(H) on CLC-1 is significantly enhanced by acidification, which has an important physiological relevance in maintaining muscle excitability during exercise. Intracellularly-applied Cd2+ can also inhibit the slow gating of CLC-0 by binding to a site at the dimer interface. The affinity of Cd2+ is greatly increased by mutations at the dimer-interface, which also reverse the voltage-dependence of channel opening and cause the channels to be activated only by hyperpolarization. These data suggest that altering the dimeric interaction, either by mutations or by binding of Cd2+, can affect the slow gating. We also find that similar reversal of voltage-dependence can be induced in WT CLC-0 simply by replacing extracellular Cl− with F− or by lowering intracellular pH to 5.5. Hyperpolarization-activation caused by mutations, F−ex, or pHin can be abolished by mutations of a conserved tyrosine residue at the center Cl−-binding site in the pore, suggesting a common mechanistic determinant that may be involved in the locking-down of the external glutamate gate.
