Book excerpt: The existence of a new family of chemotactic cytokines was realised in 1987 following the isolation and structural determination by several groups of a peptide consisting of 72 amino acids which was a potent activator of neutrophils and a chemotactic agent for lymphocytes. The first symposium of this series was held at the Royal College of Surgeons of England in December 1988, entitled Novel Neutrophil Stimulating Peptides, and brought together the majority of the laboratories which had published in this area, see Immunology Today 10: 146-147(1989). Since the first symposium there has been a dramatic increase in our knowledge of the biology of this family of structurally related peptides. The Second International Symposium on Chemotactic Cytokines was held at the Royal College of Surgeons of England in June 1990. The aim of this symposium was to provide both a forum for discussion and to determine whether this knowledge can be utilised in the design of novel therapeutic strategies for the treatment of inflammatory disorders. Although the majority of studies have been concerned with the regulation of these peptides at the molecular and cellular level, there is now evidence to suggest that specific members of this superfamily have a role in the pathogenesis of a number of diverse diseases including arthritis, psoriasis, atherosclerosis, wound repair, inflammatory lung diseases and glomerulonephritis.

That symposium successfully brought together the majority of laboratories working in the area of interleukin-8 and related peptides; see Immunology Today 10: 146-147 (1989). The Second International Symposium on Chemotactic Cytokines was held at the same venue in June 1990, and a much-increased attendance reflected the accelerating pace of work in the area of these chemotactic cytokines. The proceedings of that meeting were published in Advances in Experimental Medicine and Biology, vol. 305 (1991). The rapid advances made in the field of chemotactic cytokines over the last 18 months necessitated a third Symposium in this series to collate and place in perspective an explosion of new data. The Third International Symposium on Chemotactic Cytokines was held between August 31 and September 1, 1992 in Baden-bei-Wien, Austria. However, the lack of a clear nomenclature system was creating some confusion in the area, especially as new factors continue to be discovered and classified as family members. In the past, these inflammatory mediators had been placed arbitrarily under the broad heading of "intercrines" or "chemotactic cytokines" with no clear classification guidelines to follow. This nomenclature issue was addressed at the Symposium, where investigators in the field were invited to reach a consensus regarding a collective name for these mediators. The resulting decision was to identify the major family as chemokines, to replace all previous terms.

Severe malnutrition can cause significant alteration in immune response, but even subclinical deficits may be associated with an impaired immune response, and an increased risk of infection. Infectious diseases have accounted for more off-duty days during major wars than combat wounds or nonbattle injuries. Combined stressors may reduce the normal ability of soldiers to resist pathogens, increase their susceptibility to biological warfare agents, and reduce the effectiveness of vaccines intended to protect them. There is also a concern with the inappropriate use of dietary supplements. This book, one of a series, examines the impact of various types of stressors and the role of specific dietary nutrients in maintaining immune function of military personnel in the field. It reviews the impact of compromised nutrition status on immune function; the interaction of health, exercise, and stress (both physical and psychological) in immune function; and the role of nutritional supplements and newer biotechnology methods reported to enhance immune function. The first part of the book contains the committee's workshop summary and evaluation of ongoing research by Army scientists on immune status in special forces troops, responses to the Army's questions, conclusions, and recommendations. The rest of the book contains papers contributed by workshop speakers, grouped under such broad topics as an introduction to what is known about immune function, the assessment of immune function, the effect of nutrition, and the relation between the many and varied stresses encountered by military personnel and their effect on health.

Substances released from activated immune cells (cytokines) stimulate peripheral nerves, thereby signaling the brain and spinal cord that infection/inflammation has occurred. Additionally, peripheral infection/inflammation leads to de novo synthesis and release of cytokines within the brain and spinal cord. Thus, cytokines effect neural activation both peripherally and centrally. Through this communication pathway, cytokines such as interleukin-1, interleukin-6 and tumor necrosis factor markedly alter brain function, physiology and behavior. One important but underrecognized aspect of this communication is the dramatic impact that immune activation has on pain modulation. The purpose of this book is to examine, for the first time, immune-to-brain communication from the viewpoint of its effect on pain processing. It is aimed both at the basic scientist and health care providers, in order to clarify the major role that substances released by immune cells play in pain modulation. About 50 distinct chemokines produced by a variety cell types and tissues either c- stitutively or in response to inflammatory stimuli are involved in a plethora of biological processes. These small secreted proteins exert their exquisitely variegated functions upon binding to a family of seven-transmembrane spanning G-protein coupled receptors (GPCRs) composed of almost 20 distinct entities. The biological activities of chemokines range from the control of leukocyte trafficking in basal and inflammatory conditions to the regulation of hema- poiesis, angiogenesis, tissue architecture, and organogenesis. The basis for such diversified activities rests, on one hand, upon the ubiquitous nature of chemokine production and chemokine receptor expression. Virtually every cell type can produce chemokines and expresses a unique combination of chemokine receptors. On the other hand, chemokine receptors make use of a flexible and complex network of intracellular signaling machineries that can regulate a variety of cellular functions ranging from cell migration, growth, and differentiation to death. As knowledge of the size of chemokine and chemokine receptor families rapidly reaches completeness, much is still to be uncovered in terms of fu- tional architecture of the chemokine system. The disparity between the large number of chemokines and that smaller number of receptors is balanced by the promiscuity in ligand–receptor interactions, with multiple chemokines binding to the same receptor and several chemokines binding to more than one receptor.
