Book excerpt: It is commonly acknowledged that the nervous system and the immune system, those most complex of networks, share attributes beyond their intricacy. Elements common to the two systems include memory, connectivity, flexibility and developmental selection of cellular composition by a rigorous process involving widespread programmed cell death. There is one salient difference: the cells of the immune system are predominantly in constant motion, while post-mitotic neurons and glia are largely fixed in place. Therefore, chemokines, initially characterized as leukocyte chemoattractants, have for the last one and one-half decades been intensely and productively studied in the contexts of inflammation, immunity and hematopoietic development. Only recently have the two fields, neurobiology and immunology, displayed mutual interests in chemokines. This convergence of the two tribes of investigators was catalyzed by the finding that SDF-1 (now known as CXCL12) and its receptor, CXCR4, exerted significant and similar functions in development of both nervous and immune systems. Indeed CXCL12 and CXCR4 were required, in an uncannily similar fashion, for retention of pre-B lymphocytes at sites of maturation in the bone marrow and of neuronal progenitors in the external granule cell layer of the developing cerebellum. Recent reports indicate that chemoattraction of cerebellar granule cells through CXCR4 can be suppressed by reverse signaling initiated by binding of soluble eph receptors to transmembrane ephrin B, thereby establishing a link between chemokine action and a cardinal patterning system of the developing nervous system. As may be anticipated when a dam breaks, a massive influx of correlative observations in the nervous and immune systems is likely to ensue.This volume represents the state of current knowledge. To this end, introductory material for both systems is provided. Basic and advanced 'chemokinology' are presented. The recipe for making a nervous system (both ingredients and instructions for preparation) is described, as are the roles of chemokines and their receptors in making an immune system. Given their importance and complexity, CXCL12/CXCR4 interactions are separately treated in varying contexts.The field of 'neurobiology of chemokines' has not lain fallow during the last ten years. During much of this time the principal focus has been on neuroinflammation. Linking the immune and nervous systems are explanations of the functions of chemokines and their receptors for resident brain macrophages, the microglia, the unique cerebrovascular endothelium and angiogenesis.Understanding human disease is the goal of much of this research. New discoveries are being made and reported at a gratifying rate. It is expected that this volume will promote the steady production and application of useful new knowledge in this developing field. When initially identified close to 30 years ago, these molecules were associated with various human inflammatory diseases and it was recognized that expression may be integral in leukocyte recruitment to inflamed tissue. Within a relatively short period of time, early participants within the field determined that these proteins displayed distinct and conserved structural features and exerted potent chemotactic effects on defined lymphocyte subsets. There are now four sub-families of chemokines identified based on defined structural criteria relating to the positional location of conserved cysteine residues within the amino-terminus of the protein. Chemokines are now recognized as important in numerous biological processes ranging from maintaining the organizational integrity of secondary lymphoid tissue to participating in various aspects of both innate and adaptive immune responses following microbial infection.

Book excerpt: The first symposium in this series was held at the Royal College of Surgeons of England in December 1988 and was entitled "Novel Neutrophil Stimulating Peptides". That symposium successfully brought together the majority of laboratories working in the area of interleukin-8 and related peptides; see Immunology Today 10: 146-147 (1989). The Second International Symposium on Chemotactic Cytokines was held at the same venue in June 1990, and a much-increased attendance reflected the accelerating pace of work in the area of these chemotactic cytokines. The proceedings of that meeting were published in Advances in Experimental Medicine and Biology, vol. 305 (1991). The rapid advances made in the field of chemotactic cytokines over the last 18 months necessitated a third Symposium in this series to collate and place in perspective an explosion of new data. The Third International Symposium on Chemotactic Cytokines was held between August 31 and September 1, 1992 in Baden-bei-Wien, Austria. However, the lack of a clear nomenclature system was creating some confusion in the area, especially as new factors continue to be discovered and classified as family members. In the past, these inflammatory mediators had been placed arbitrarily under the broad heading of "intercrines" or "chemotactic cytokines" with no clear classification guidelines to follow. This nomenclature issue was addressed at the Symposium, where investigators in the field were invited to reach a consensus regarding a collective name for these mediators. The resulting decision was to identify the major family as chemokines, to replace all previous terms.

Whether soluble chemokines can promote integrin activation and arrest is controversial (Alon-Gerszten). Many aspects of the signaling pathway from the GPCR chemokine receptor to integrin activation are the subject of active investigation. Leukocyte adhesion deficiency III is a human disease in which chemokine-triggered integrin activation is defective because of a mutation in the cytoskeletal protein kindlin-3. About 10 different such mutations have been described. The defects seen in patients with LAD-III elucidate the importance of rapid integrin activation for host defense in humans. We welcome reports that help clarifying this crucial first step in the process of leukocyte transendothelial migration.
