Book excerpt: The B lymphocyte lineage represents an important paradigm for exploring the molecular mechanisms underlying cell fate specification, differentiation and cellular activation. In the past five years, major advances have been achieved in our understanding of the transcriptional control of early B cell development and terminal plasma cell differentiation. In addition new insights became available for the processes of B cell activation, class switch recombination and somatic hypermutation. Many of the new findings and their implications for a molecular understanding of B cell biology in particular and cell differentiation in general are covered in this volume.

Available in PDF, EPUB and Kindle. Book excerpt: Transcription Factors Normal and Malignant Development of Blood Cells Katya Ravid and Jonathan Licht The role of transcription factors in activating specific genes in blood cells is an important facet of hematopoiesis. Equally important, however, is the pursuit of genes rearranged and aberrantly activated in leukemias (blood malignancies). Transcription Factors: Normal and Malignant Development of Blood Cells focuses on those major transcription factors involved in activation of lineage-specific gene expression during normal versus malignant development of specific blood lineages, as revealed from gene promoter studies, knockout of transcription factors in mice models, and the identification and characterization of chromosomal rearrangement in human blood leukemias. This complete digest of current transcription factor data offers comprehensive coverage of the myriad of transcription factors in blood cell development, composed by established experts in the field. In addition to updating the reader on the connection between chromosomal translocations involving transcription factors and cellular transformation leading to leukemia, Transcription Factors also reviews such subjects as: * Transcription factors and the megakaryocytic, myeloid, and erythroid lineages * Leukemias due to chromosomal translocations involving gene encoding transcription factors * Oncogenesis and hematopoiesis * In vivo studies of transcription factors implicated in hematopoiesis * And much more Appealing to both the researcher and the clinician in the field of hematology, Transcription Factors is a timely presentation of cell lineage development and sheds light on the processes involved in the development of specific leukemias. Providing insight into the study of transcription factors, readers will gain an understanding of mechanisms that lead to normal lineage commitment and terminal differentiation.

Book excerpt: Despite the tremendous diversity of the cells of the hematopoietic system, they are all derived from common precursor cells that are generated in the fetus and persist into adult life. In this regard, Band T lymphocytes, which comprise the two arms of the antigen-specific and inducible immune system, though functionally very different, are descendants of the same stem cell precursor. In the past several years, we have witnessed an explosion of information regarding the process by which differentiation of B-and T-cells from stem cells occurs. This information, like the answers to most important biological questions, has come from multiple and diverse directions. Because all hematopoietic cells arise from common precursors, complex regulatory processes must be involved in determining commitment to various lineages. Understanding commitment to the B- or T-cell lineage remains incomplete; however, identification of transcription factors necessary for progression along specific B-and T-cell pathways suggests that we are on the verge of understanding the molecules involved in the initial fate-determining steps. Studies of this type previously could be accomplished only in nonmammalian systems that are more amenable to genetic approaches. However, new technologies allow increasingly elegant and informative studies in mammalian systems, particularly for cells of the hematopoietic system.

Book excerpt: Molecular Biology of B Cells, Second Edition is a comprehensive reference to how B cells are generated, selected, activated and engaged in antibody production. All of these developmental and stimulatory processes are described in molecular, immunological, and genetic terms to give a clear understanding of complex phenotypes. Molecular Biology of B Cells, Second Edition offers an integrated view of all aspects of B cells to produce a normal immune response as a constant, and the molecular basis of numerous diseases due to B cell abnormality. The new edition continues its success with updated research on microRNAs in B cell development and immunity, new developments in understanding lymphoma biology, and therapeutic targeting of B cells for clinical application. With updated research and continued comprehensive coverage of all aspects of B cell biology, Molecular Biology of B Cells, Second Edition is the definitive resource, vital for researchers across molecular biology, immunology and genetics. - Covers signaling mechanisms regulating B cell differentiation - Provides information on the development of therapeutics using monoclonal antibodies and clinical application of Ab - Contains studies on B cell tumors from various stages of B lymphocytes - Offers an integrated view of all aspects of B cells to produce a normal immune response

Available in PDF, EPUB and Kindle. Book excerpt: This book gives an in-depth overview on nuclear structure and function. It clearly shows that the epigenome and the three-dimensional organization of the nucleus are not independent properties. The intimate relationship between the location and the epigenetic modifications of gene loci is highlighted. Finally, it shows that the complex three-dimensional organization of the nucleus is not just of academic interest: The structure, composition and function of virtually all of the sub-nuclear compartments identified so far can be implicated to a list of human genetic diseases. Hence, a detailed elucidation of how these domains are assembled and function will provide new opportunities for therapeutic intervention in clinical practice.

Book excerpt: The comparative approach to immunology can be traced to the era of Pasteur and Metchnikov in which observations regarding foreign recognition in invertebrates was a factor in the develop ment of the principal concepts that created the foundation of what now is the broad field of immunology. With each major experimental and conceptual breakthrough, the classical, albeit essential, question has been asked "are the immune systems of phylogenetically primitive vertebrates and invertebrates similar to that of mammals?" Somewhat surprisingly for the jawed verte brates, the general answer has been a qualified form of "yes", whereas for agnathans and invertebrate phyla it has been "no" so far. The apparent abruptness in the appearance of the immune system of vertebrates is linked to the introduction of the somatic generation of the diversity of its antigen specific receptors. Therefore the questions regarding the origin and evolution of the specific immune system revolve around this phenomenon. With respect to the origin of the system (aside from the or igin of the rearranging machinery itself, the study of which is still in its infancy) one can ask questions about the cellular and mo lecular contexts in which the mechanism was introduced.
