Book excerpt: To study the scope of guests that can be recognized, several proteins were functionalized with R-NMe3+ binding handles and the recognition event between the functionalized proteins and cavitand:SLB system was monitored by Surface Plasmon Resonance (SPR) Spectroscopy. The binding was monovalent, and displayed a binding affinity of>105 M-1, showing that the cavitand is capable of recognizing suitably labeled large proteins via cavity-based recognition.

Book excerpt: Recognition receptors play a key role in the successful implementation of chemical and biosensors. Molecular recognition refers to non-covalent speci?c binding between molecules, one of which is typically a macromolecule or a molecular assembly, and the other is the target molecule (ligand or analyte). Biomolecular recognition is typically driven by many weak interactions such as hydrogen bo- ing, metal coordination, hydrophobic forces, van der Waals forces, pi-pi interactions and electrostatic interaction (due to permanent charges, dipoles, and quadrupoles) the polarization of charge distributions by the interaction partner leading to ind- tion and dispersion forces, and Pauli-exclusion-principle-derived inter-atomic repulsion, and a strong, “attractive” force arising largely from the entropy of the solvent and termed the hydrophobic effect. In recent years, there has been much progress in understanding the forces that drive the formation of such complexes, and how these forces are relate to the physical properties of the interacting molecules and their environment allows rational design of molecules and materials that interact in speci?c and desired ways. This book presents a signi?cant and up-to-date review of the various recognition elements, their immobilization, characterization techniques by a panel of dist- guished scientists. This work is a comprehensive approach to the recognition receptors area presenting a thorough knowledge of the subject and an effective integration of these receptors on sensor surfaces in order to appropriately convey the state-of the-art fundamentals and applications of the most innovative approaches.

Available in PDF, EPUB and Kindle. Book excerpt: Bioanalytical chemistry plays today a central role in various fields, from healthcare to food and environmental control. This book presents the main methodologies for analyzing biomacromolecules, with a focus on methods based on molecular recognition. The six chapters move from fundamentals to the most recent advances, achieved by a synergetic combination of bio and nanotechnologies. The need for rapid and reliable analytical tools able to perform a large number of quantitative analyses, not only in centralized laboratories and core facilities but also for point-of-care testing, has been dramatically stressed by the recent crisis caused by the COVID-19 pandemic. The aim of the authors is to provide graduate students and young researchers with the elements of interdisciplinary knowledge necessary not only to use the wide arsenal of bioanalytical tools available today but also to contribute to the development of even more effective devices and methods.

This new strategy emphasizes the rational complementation of functionality along extended patches of a protein surface with the aim of inhibiting protein/protein interactions. The successful development of compounds able to inhibit these interactions offers a unique chance to selectively intervene in a large number of key cellular processes related to human disease. Protein Surface Recognition presents a detailed treatment of this strategy, with topics including: an extended survey of protein-protein interactions that are key players in human disease and biology and the potential for therapeutics derived from this new perspective the fundamental physical issues that surround protein-protein interactions that must be considered when designing ligands for protein surfaces examples of protein surface-small molecule interactions, including treatments of protein-natural product interactions, protein-interface peptides, and rational approaches to protein surface recognition from model to biological systems a survey of techniques that will be integral to the discovery of new small molecule protein surface binders, from high throughput synthesis and screening techniques to in silico and in vitro methods for the discovery of novel protein ligands. Protein Surface Recognition provides an intellectual “tool-kit” for investigators in medicinal and bioorganic chemistry looking to exploit this emerging paradigm in drug discovery.

Available in PDF, EPUB and Kindle. Book excerpt: Molecular recognition, also known as biorecognition, is the heart of all biological interactions. Originating from protein stretching experiments, dynamic force spectroscopy (DFS) allows for the extraction of detailed information on the unbinding process of biomolecular complexes. It is becoming progressively more important in biochemical studies and is finding wider applications in areas such as biophysics and polymer science. In six chapters, Dynamic Force Spectroscopy and Biomolecular Recognition covers the most recent ideas and advances in the field of DFS applied to biorecognition: Chapter 1: Reviews the basic and novel aspects of biorecognition and discusses the emerging capabilities of single-molecule techniques to disclose kinetic properties and molecular mechanisms usually hidden in bulk measurements Chapter 2: Describes the basic principle of atomic force microsocopy (AFM) and DFS, with particular attention to instrumental and theoretical aspects more strictly related to the study of biomolecules Chapter 3: Overviews the theoretical background in which experimental data taken in nonequilibrum measurements of biomolecular unbinding forces are extrapolated to equilibrium conditions Chapter 4: Reviews the most common and efficient strategies adopted in DFS experiments to immobilize the interacting biomolecules to the AFM tip and to the substrate Chapter 5: Presents and discusses the most representative aspects related to the analysis of DFS data and the challenges of integrating well-defined criteria to calibrate data in automatic routinary procedures Chapter 6: Overviews the most relevant DFS applications to study biorecognition processes, including the biotin/avidin pair, and selected results on various biological complexes, including antigen/antibody, proteins/DNA, and complexes involved in adhesion processes Chapter 7: Summarizes the main results obtained by DFS applied to study biorecognition processes with forthcoming theoretical and experimental advances Although DFS is a widespread, worldwide technique, no books focused on this subject have been available until now. Dynamic Force Spectroscopy and Biomolecular Recognition provides the state of the art of experimental data analysis and theoretical procedures, making it a useful tool for researchers applying DFS to study biorecognition processes.
