But when the cancer cells have to spread to form secondary colonies, the prognosis for the patient is worse. If meaningful improvements in survival are to occur, then control of metastasis will be a foundation. Relatively little is known about the control of the metastatic process at the molecular level. This volume begins to explore our current knowledge regarding the underlying molecular and biochemical mechanisms controlling the metastatic phenotype. While all of the authors attempted to put their findings into a context for translation to the clinical situation, the state-of-the-art does not fully allow this. Nonetheless, we write these summaries of our work as an early effort toward that end. I am grateful to all of the authors who have contributed generously of their time and energies to make this volume a reality. To metastasize, neoplastic cells dissociate from the primary tumor, enter a circulatory compartment (typically lymphatics or blood vasculature), survive transport, arrest, exit the circulation and finally proliferate at a discontinuous site in response to local growth factors. To study carbohydrate-mediated interactions in these processes, four closely-related murine mammary carcinoma cell lines were used: metastatic 4T1 and 4T1 altered-properties, and non-metastatic 4T07 and 67NR. The Consortium for Functional Glycomics (CFG) used RNA from each cell line to compare carbohydrate-related gene expression on a Glyco-gene chip. The Glyco-gene chip is a microarray capable of detecting over 1,800 different carbohydrate-related genes. Thirteen differentially-expressed genes on the Glyco-gene chip were validated by quantitative RT-PCR. Flow cytometry was used to detect cell surface expression of differentially-expressed gene products including: VCAM-1, CD34, TF-antigen, and linkages of sialic acid and carbohydrates. The CFG performed glycan profiling on each cell line. Cell surface carbohydrates were identified and quantified using a technique similar to mass-spectrometry. This analysis can be utilized to further understand, and to develop therapy for the prevention of metastasis.

Book excerpt: We propose to identify the functional characterization of two novel cancer-specific, metastasis-related genes whose constitutive expression may be pivotal for prostate cancer progression. Work accomplished was performed based on the proposed statement of work. We have characterized the full-length cDNAs of the Seq1 and Seq2 genes using at least three 5' and '3 rapid amplifications of cDNA ends (RACE) commercial kits (Invitrogen Carlsbad, CA, BD Bioscience (Clontech Inc), and Seegene, Rockville, MD). To optimize the PCR conditions for each kit, we had designed several sets of gene-specific primers (GSP;23-28 nt long) with 50-70% GC and Tm of 55 to 75 degrees C for each gene. We have also designed several sets of nested GSPs to verify our cloned genes. Because of unique secondary structures, high GC content, short SSH sequences, and low levels of expression of these genes in prostate cancer cell lines, we had great deal of difficulty in accomplishing this task in a timely fashion. As such, we devised different strategies for the first-strand synthesis using a modified oligo(dT) primers (5'-CDS primer or 3'-CDS primer), and Smart oligo II primer under various conditions. The full-length cDNA sequences were subcloned into mammalian expression vectors (Invitrogen) and ready to be used for generation of recombinant proteins and antibody production.

Cell migration is the basic and essential step in cancer metastasis process. To systematically investigate functional genes regulating cell migration and cancer metastasis on large scale, we developed a novel on-chip method, SAMcell (self-assembled cell microarray). This method was demonstrated to be particularly suitable for loss-of-function high-throughput screening because of its unique advantages. The first application of SAMcell was to screen human genome miRNAs, considering that more and more miRNAs had been proved to govern cancer metastasis. We found that over 20 % of miRNAs have migratory regulation activity in diverse cell types, indicating a general involvement of miRNAs in migratory regulation. Through triple-round screenings, we discovered miR-23b, which is down-regulated in human colon cancer samples, potently mediates the multiple steps of metastasis, including cell motility, cell growth and cell survival. In parallel, the second application of SAMcell was to screen human genome kinase genes, considering that more and more kinase genes had become successful diagnostic marker or drug targets. We found over 11% migratory kinase genes, suggesting the important role of kinase group in metastasis regulation. Through both functional screening and bioinformatics analysis, we discovered and validated 6 prospective metastasis-related kinase genes, which can be new potential targets in cancer therapy. These findings allow the understanding of regulation mechanism in human cancer progression, especially metastasis and provide the new insight into the biological and therapeutical importance of miRNAs or kinases in cancer.
