Metastasis is a multistep process during which cancer cells leave the primary tumor, invade and seed inside secondary tissues, leading to lethal organ failure. Currently, no efficient method exists for the elimination of invasive cancer cells, which poses a major clinical challenge. Indeed, while localized cancers are clinically manageable, invasive breast cancer is largely an uncurable disease. Understanding how cancer cells sustain an invasive behavior may lead to the development of novel potent therapies.A vast genetic and phenotypic heterogeneity exists among invasive cancer cells. This heterogeneity is important to investigate as, in breast cancer patients, most metastatic nodules are polyclonal and consist of cancer cells that invaded and disseminated collectively. While recent studies demonstrated that collective invasion can be cooperative, where leader cells enable the invasion of otherwise non-invasive follower cells, the mechanisms regulating their cooperativity have been poorly investigated. Additionally, it is not known whether cooperative invasion can lead to metastasis, where metastatic cells enable the dissemination of non-metastatic cells. Our laboratory previously showed that breast cancer cells can locally degrade the extracellular matrix using invadopodia, which are membrane protrusions enriched in matrix metalloproteinases. Importantly, invadopodia are required for metastasis of single cancer cells. However, whether invadopodia enable the collective invasion and metastasis of cancer cell clusters remains unknown. Further, the role of invadopodia-mediated matrix degradation in leader vs. follower cells is unclear. In this dissertation, we aim to understand how cancer clones with and without invadopodia may cooperate during invasion and metastasis. Our results suggest that a few cells with invadopodia can drive the metastasis of cell clusters from multiclonal tumors. From this work, we believe that a more detailed understanding of how cancer cells cooperate may help in identifying new targets for preventing metastasis.

Overexpression of the HER2/ErbB2 receptor tyrosine kinase is associated with metastatic disease and correlates with reduced patient survival. TGF[beta] can activate signaling pathways that cooperate with ErbB2 to enhance the migratory, invasive and metastatic behavior of breast cancer cells. Furthermore, TGF[beta] is a strong inducer of an epithelial-to-mesenchymal transition (EMT), and breast cancers with EMT features are associated with poor clinical outcomes.Using a panel of ErbB2-overexpressing breast cancer cells that undergo TGF[beta]-induced EMT, I identified Lipoma Preferred Partner (LPP) as an indispensable mediator of breast cancer cell migration and invasion in vitro. I show that LPP is recruited to focal adhesion (FA) complexes through its LIM1 domain and subsequently recruits [alpha]-actinin, an actin cross-linking protein, as a mechanism to facilitate FA dynamics, enhance cell migration, and increase cell invasion of breast cancer cells that can undergo an EMT. In addition, I have discovered a novel role for LPP in invadopodia formation, ECM degradation and breast cancer metastasis. Live-cell time-lapse intravital imaging in a chick chorioallantoic membrane (CAM) system reveals that LPP localizes to invadopodia during breast cancer extravasation. In mouse xenograft models, loss of LPP leads to a significant impairment in spontaneous lung metastasis formation, while primary mammary tumor growth is unaffected. Moreover, I demonstrate that LPP is necessary for the efficient dissemination of breast cancer cells from the primary tumor, as diminished LPP expression significantly reduces the number of detectable circulating tumor cells, and impairs the extravasation of breast cancer cells to establish pulmonary metastasis. Together, these data demonstrate that LPP promotes breast cancer metastasis through its role in mediating invadopodia formation to drive tumor cell invasion and colonization of secondary sites. Finally, I have identified LPP as a novel substrate of the c-Src tyrosine kinase in response to TGF[beta]-stimulation. In multiple murine and human breast cancer cell lines, LPP is tyrosine-phosphorylated following TGF[beta] treatment, and this phosphorylation can be blocked using pharmacological inhibitors against Src family kinases. ErbB2-expressing breast cancer cells specifically lacking Src also revealed impaired LPP phosphorylation in response to TGF[beta]. To elucidate the potential consequences of LPP phosphorylation, I constructed a panel of LPP phospho-mutants and demonstrated that phosphorylation of Y245, Y301 and Y302 within LPP is required for cellular invasion, invadopodia formation, ECM degradation and breast cancer metastasis.Collectively, the work described in this thesis is the first to define LPP as a novel pro-metastatic mediator that integrates TGF[beta] and ErbB2 signaling to promote the migration, invasion and metastasis of breast cancer cells." --

However, defects in the regulation of this exchange appear to contribute to malignancy by allowing growth promoting, invasogenic, and angiogenic factors to accumulate within the microenvironment of the tumor. For example, recent studies suggest that abnormal interactions between tumor epithelial cells and stromal mesenchymal cells contribute to the overproduction and accumulation of scatter factor (hepatocyte growth factor), an invasogenic and angiogenic cytokine, in certain types of tumor. The production and and activation of type IV collagenase, a matrix-degrading enzyme required for tumor cell invasion, appears to require intimate cooperation between tumor and stromal cells. The material contained in this volume highlights the state-of-the-art of knowledge of the molecular mechanisms by which epithelia and mesenchyme collaborate, and the abnormalities in these mechanisms that may lead to the development of cancer.

Book excerpt: The tumor microenvironment (TME) is a heterogeneous region that is comprised of tumor cells, stromal cells, and secreted factors and creates an environment favorable for tumor cell invasion and metastasis. An important step in the shift to a pro-cancerous microenvironment is the transformation of normal stromal fibroblasts to carcinoma-associated fibroblasts (CAFs). CAFs are present in a majority of solid tumors and can directly promote tumor cell motility via cytokine, chemokine and growth factor secretion into the TME, making them a critical TME component to understand. The exact effects that the TME has upon cytoskeletal regulation in motile tumor cells remain enigmatic. The conserved formin family of cytoskeleton regulating proteins plays essential roles in the assembly and/or bundling of unbranched actin filaments. Mammalian Diaphanous-related formin-2 (mDia2/DIAPH3/Drf3/Dia) assembles a dynamic F-actin cytoskeleton that underlies tumor cell migration and invasion. We previously showed that the chemokine CXCL12 can influence breast tumor cell migration and invasion through modification of the mDia2-directed actin cytoskeleton. The endogenous source of CXCL12 was unexplored. Therefore, the objective of this study was to understand whether CAF-derived chemokines from the TME impact breast tumor cell motility through modification of the formin-assembled F-actin cytoskeleton. In this study, we used CAF-derived conditioned media (CM) from WS19T fibroblasts, a transformed patient-derived tumor-adjacent breast CAF cell line, and studied its cell-autonomous impact upon tumor-cell motility. In triple-negative MDA-MB-231 human breast cancer cells, WS19T CAF-CM significantly and robustly increased wound closure and invasion relative to normal human mammary fibroblast (HMF)-CM. Unexpectedly, western blot analysis of WS19T-CM-treated MDA-MB-231 cells revealed a significant loss of mDia2 protein expression. WS19T-CM also promoted proteasome-mediated mDia2 degradation in MDA-MB-231s relative to control HMF-CM and WS21T CAF-CM, a breast CAF cell line that failed to reduced mDia2 expression, but significantly increased MDA-MB-231 migration. Cytokine array analysis of CM identified upregulated secreted factors in WS19T-CM relative to control WS21T CM, including the chemokine CXCL12 (SDF1a). As CXCL12 was identified as a factor secreted by fibroblasts and heavily involved in cancer cell migration and invasion, we hypothesized that CXCL12 is a CM factor influencing mDia2 protein loss, while increasing MDA-MB-231 cell invasion and migration. Exogenous CXCL12 treatment resulted in increased wound closure and loss of mDia2 protein expression. Blocking CXCL12 signaling with AMD3100, a specific inhibitor of CXCR4, augmented motility and rescued mDia2 protein expression in the presence of WS19T-CM. Our data suggest a mechanism whereby CAFs promote tumor cell migration and invasion through CXCL12 secretion to regulate the mDia2-directed cytoskeleton in breast tumor cells, and highlights a novel therapeutic approach of treating highly invasive primary lesions through targeting the TME.
