Available in PDF, EPUB and Kindle. Book excerpt: Over the last decade, high-sensitivity calorimetry has developed from a specialist method used mainly by dedicated experts to a major, commercially available tool in the arsenal directed at understanding molecular interactions and stability. Calorimeters have now become commonplace in bioscience laboratories. As a result, the number of those proficient in experimentation in this field has risen dramatically, as has the range of experiments to which these methods have been applied. Applications extend from studies in small molecule and solvent biophysics, through drug screening to whole cell assays. The technology has developed to include higher levels of sensitivity (and hence smaller sample size requirements) and a drive towards high-throughput technology, creating a very large user base in both academia and the pharmaceutical industry. This book is a fully revised and updated edition of the successful Biocalorimetry: Applications of Calorimetry in the Biological Sciences, published in 1998. Since then, there have been many advances in the instrumentation as well as in its applications and methodology. There are general chapters highlighting the usage of the isothermal titration calorimeter and the differential scanning calorimeter, more advanced chapters on specific applications and tutorials that cover the idiosyncrasies of experimental methods and data analysis. The book draws these together to create the definitive biological calorimetric text book. This book both explains the background to the method and describes novel, high-impact applications. It features works of interest to the experienced calorimetrist and the enthusiastic dilettante. The book should be of interest to all working in the field of biocalorimetry, from graduate students to researchers in academia and in industry.

I thought that the sections were well balanced, with rarely too much or too little on a given topic...This is a text to be welcomed by both teachers and students." BIOCHEMISTRY & MOLECULAR BIOLOGY EDUCATION (on the first edition) The second edition of this successful textbook explains the basic principles behind the key techniques currently used in the modern biochemical laboratory and describes the pros and cons of each technique and compares one to another. It is non-mathematical, comprehensive and approachable for students who are not physical chemists. A major update of this comprehensive, accessible introduction to physical biochemistry. Includes two new chapters on proteomics and bioinformatics. Introduces experimental approaches with a minimum of mathematics and numerous practical examples. Provides a bibliography at the end of each chapter. Written by an author with many years teaching and research experience, this text is a must-have for students of biochemistry, biophysics, molecular and life sciences and food science.

Book excerpt: Biocalorimetry Applications of Calorimetry in the Biological Sciences Edited by John E. Ladbury, University College London, UK and Babur Z. Chowdhry, University of Greenwich, London, UK Biocalorimetry: Applications of Calorimetry in the Biological Sciences is the first book to bring together an overview of the two key methods in calorimetry, (isothermal titration calorimetry (ITC) and differential scanning calorimetry (DSC)), with a wide variety of biological applications, such as DNA-drug/protein interactions, protein-protein (ligand) interactions and protein denaturation studies. Written by a team of international researchers, it discusses the principles and practical application of these methods, including their advantages and disadvantages. This book will be of interest to researchers working in biochemistry, biophysics and molecular biology, particularly those in the pharmaceutical, food and biotechnology industries. In addition, Biocalorimetry: Applications of Calorimetry in the Biological Sciences provides valuable insight for lecturers and students, (particularly final year undergraduates and postgraduates), for teaching purposes, as well as for understanding the theoretical basis and wider practical applications of calorimetry.

This book was released on 2018-10-31 with total page 55 pages. Available in PDF, EPUB and Kindle. Book excerpt: In the manufacture of bio-therapeutics the importance of a well-known process is key for a high product titer and low batch to batch variations. Soft sensors are based on the concept that online sensor signals can be used as inputs to mathematical models to derive new valuable process information. This information could then be used for better monitoring and control of the bioprocess. The aim of the present thesis has been to develop soft sensor solutions for upstream bioprocessing and demonstrate their usefulness in improving robustness and increase the batch-to-batch reproducibility in bioprocesses. The thesis reviews the potential and possibilities with soft sensors for use in production of bio-therapeutics to realize FDA´s process analytical technology (PAT) initiative. Modelling and hardware sensor alternatives which could be used in a soft sensor setup are described and critically analyzed. Different soft sensor approaches to control glucose feeding in fed-batch cultures of Escherichia coli are described. Measurements of metabolic fluxes and specific carbon dioxide production was used as control parameters to increase product yield and decrease the variability of produced recombinant proteins. Metabolic heat signals were used in uninduced cultures to estimate and control the specific growth rate at a desired level and thereby also estimate the biomass concentration online. The introduction of sequential filtering of the signal enabled this method to be used in a down-scaled system. The risk and high impact of contaminations in cell cultures are also described. An in situ microscope (ISM) was used as an online tool to estimate cell concentration and also to determine cell diameter size which enabled the detection of contaminant cells at an early stage. The work presented in this thesis supports the idea that soft sensors can be a useful tool in the strive towards robust and reliable bioprocesses, to ensure high product quality and increased economic profit.
