Feeding behavior in the mollusk Aplysia californica is a useful model system to analyze these two ubiquitous forms of associative learning and compare the underlying neuronal mechanisms. Here, we review studies that have analyzed and compared the mechanisms underlying classical and operant conditioning at the circuit, single-cell, and molecular levels. These analyses reveal similarities and intriguing differences. Both forms of learning lead to increased biting in vivo and fictive ingestion in vitro and also share a common reinforcement pathway, which uses dopamine as the reinforcement transmitter. Although the identified neuron B51 is a locus of plasticity common to both classical and operant conditioning, its activity is altered in opposite ways by these two forms of learning. B51 excitability is increased by operant conditioning, whereas it is decreased by classical conditioning.

In particular, the reflex can exhibit long-term sensitization (LTS), a form of nonassociative memory. LTS is mediated by long-term facilitation (LTF) of the monosynaptic connection between the sensory and motor neurons that mediate the withdrawal reflex. LTS and LTF represent one of the best understood model systems of long-term memory extent. Furthermore, discoveries from work on this system have provided fundamental insights into the cellular and molecular mechanisms that mediate the induction and maintenance of long-term memory. This chapter reviews this work; it concludes with a discussion of recent studies of the role of protein kinase M in the persistence of the long-term memory and of memory reconsolidation in Aplysia. It is suggested that the study of LTS and LTF can provide important mechanistic information on these two intriguing memory phenomena.

Differences from response to response, and from one individual to another, have been found in systems ranging from insects (Hooper et al. 2006) to humans (Nussbaum and Chaffin 1997, Borzelli et al. 1999). As a motor pattern progresses from ions to movement, there are many sources of variability. Sometimes variability is carried from one stage to the next, while in other instances it can be filtered out by the system. Aplysia feeding has been shown to be so highly variable (Horn et al. 2004) that Lum et al. 2005 were unable to make strong correlations between motor patterns and behavior. If sources of variability that do and do not influence behavior are mixed together, they may obscure the relationship between motor programs and behavior. We determined that is possible to parse different types of variability and determine which sources influence the final behavior. We determined that motor program variations that distinguish different behavior types are likely to be correlated with behavior, whereas individual variability is unlikely to be correlated with behavior. It appears that sensory feedback shifts intrinsically individual motor programs into a common motor solution to meet the demands of the behavior being performed.
