Understanding of the regulation of brown and beige adipocytes will provide novel strategies to reach the goal. Pattern recognition receptors (PRR) are responsible for inflammation in adipose tissue, which leads to adipose dysfunction and obesity associated chronic diseases. It has been shown that PRR activation induces inflammation, leading to insulin resistance in white adipocytes and white adipose tissue (WAT). However, the roles of PRR activation in brown adipocytes and brown adipose tissue (BAT) have not been studied. In addition, in vitro and in vivo studies have shown that bioactive food components can contribute to obesity prevention; however, very few bioactive food components have been studied for their beneficial effects in promoting development and function of brown and beige adipocytes. This dissertation reports that (1) mRNA of PRRs and inflammatory cytokines/chemokines are upregulated in the BAT of both diet induced obesity mice model and ob/ob mice model; (2) activation of PRRs induces activation of NF-kB and MAPK signaling pathways, leading to upregulation of the proinflammatory genes, MCP- 1, IL-6, RANTES, and TNF-[alpha], in mature brown adipocytes; (3) activation of PRRs suppresses UCP-1 expression, leading to decreased mitochondrial respiration and thermogenesis in mature brown adipocytes; (4) chronic activation of PRRs reduces adipogenesis and suppresses expression of brown-specific genes in both classic brown adipocytes and multipotent stem cells. This dissertation further demonstrates that naringenin, a flavanone mainly found in citrus fruits, enhances thermogenic activation in isoproterenol-stimulated 3T3-L1 adipocytes through PKA/p38 MAPK pathways. The results suggest that PRR-mediated inflammation in brown adipocytes may be a potential target for regulating BAT development and function for obesity treatment and prevention. Dietary bioactive compounds, such as naringenin, could be beneficial in promoting functional BAT, thereby contributing to energy expenditure.

In mice, brown adipocytes like cells (beige adipocytes) appear in subcutaneous fat depot after prolonged cold exposure. Fully-functioning BAT and beige adipocytes in subcutaneous fat depot drive a sharp increase in energy expenditure. In fact, cold-elicited BAT activation and beige cell recruitment in subcutaneous adipose tissue have shown anti-obesity effect, making them a promising target for obesity treatment. Due to the practice reasons, intermittent cold exposure may be an effective approach to activate BAT and to treat obesity and type 2 diabetes. However, cold exposure is known to induce various physiological changes and therefore warrants a comprehensive study looking at the effects of cold exposure on the whole body energy homeostasis. We found that intermittent cold exposure (ICE) leads to mild adaptive hyperphagia and fat accumulation in mice. Fat accumulation was mostly observed in subcutaneous adipose tissue with increased lipogenesis genes expression. ICE also increased fat accumulation in liver. Interestingly, pair-feeding and restrain studies ruled out the causal role of hyperphagia and stress on ICE-induced fat accumulation. In conclusion, intermittent cold exposure shifts energy homeostasis favoring fat accumulation in mice, although ICE stimulates BAT activation and beige cell recruitment in inguinal fat.

Book excerpt: Abstract: Obesity is associated with an imbalance resulting from increased caloric intake and decreased energy expenditure. Humans have two types of adipose tissue: white adipose tissue (WAT), specialized for the storage of excess energy as lipid; and brown adipose tissue (BAT), which catabolizes lipid, releasing heat. Enhancing the development and/or activity of brown adipose tissue (BAT) or brown-like (beige) adipocytes within WAT is proposed as a means to enhance energy expenditure in obese individuals. Brown adipocyte progenitors share common origins with vascular cells; however, the mechanisms regulating commitment to these lineages are not understood. Bone Morphogenic Protein 7 (BMP7), a member of the Transforming Growth Factor B (TGFB) superfamily, promotes the development of brown adipocytes. The goal of this study was to identify novel mechanisms regulating the commitment of mesenchymal stem cells (MSCs) to the brown adipocyte lineage, and to characterize the contrasting effects of BMP7 and TGFB1 on cell fate.To address these questions, we used MSC culture models and identified several genes that are selectively regulated by BMP7 during BAT lineage commitment. These include the transcription factor Zinc Finger Protein of the Cerebellum 1 (Zic1 ), Gremlin1, a secreted BMP antagonist, and regulators of cell shape, the Rho-associated protein kinases, Rock1/2. Repression of Gremlin1 and Zic1 expression are necessary for MSCs to undergo brown adipogenesis. It is well-established that TGFB1 activates ROCK, and induces elongated, myofibroblast morphology in MSCs. We demonstrate that BMP7, in contrast, represses ROCK activity, altering actin dynamics and promoting a broadened morphology. Consistent with these findings, inhibition of ROCK activity or the downstream transcription factor Serum Response Factor (SRF), promotes brown adipocyte development. We conclude from these results that changes in cell shape and gene expression programs that are responsive to the status of the actin cytoskeleton are critical mediators of BAT lineage commitment.Our study has identified multiple genes involved in a brown fat/myofibroblast phenotypic switch. Modulation of ROCK or SRF activity may provide a novel means of promoting the development of brown/beige adipocytes in obese individuals. Further delineation of mechanisms regulating BAT development will lead to the identification of novel targets for anti-obesity therapeutics.
