The cerebellar cortex was from the first an object of close study in the electron microscope, repeating in modern cytology and neuroanatomy the role it had in the hands of RAMON y CAJAL at the end of the nineteenth century. The senior author vividly remembers a day early in 1953 when GEORGE PALADE, with whom he was then working, showed him an electron micrograph of a cerebellar glomerulus, saying "That is what the synapse should look like. " It is true that the tissue was swollen and the mitochondria were exploded, but all of the essentials of synaptic structure were visible. At that time small fragments of tissue, fixed by immersion in osmium tetroxide and embedded in methacrylate, were laboriously sectioned with glass knives without any predetermined orientation and then examined in the electron microscope. After much searching, favorably preserved areas' were studied at the cytological level in order to recognize the parts of neurons and characterize them. Such procedures, dependent upon random sections and uncontrollable selection by a highly erratic technique of preservation, precluded any systematic investigation of the organization of a particular nucleus or region of the central nervous system. It was difficult enough to distinguish neurons from the neuroglia.

The concept gradually achieved general acceptance, and by 1971 the amount of information that could profit by being examined from the point of view of metabolic compartmentation had increased sufficiently to warrant a symposium on the subject ("Metabolic Compartmentation in the Brain," Balazs and Cremer, eds. , MacMillan, 1973). Almost all the participants at that initial symposium were present at the second, on which this volume is based.

Book excerpt: 1. 1 Brief History The diversity of cells constituting the central nervous system did not deceive last century neurohistologists in recognizing that this organ contained essentially two cell types: the nerve cells, or as termed according to the emerging concept of neural contiguity, the neurons, and the neuroglial cells. Neurons were clearly shown to be the means of excitability, impulse generation, impulse transmission, and connectivity in the neural tissue. The neuroglia, as indicated by its name (YAloc=cement or glue) given by Virchow (1860), was thought to be the cement ing material ensuring the coherence of the nervous tissue, filling in the spaces of the neuropil, and isolating neuronal cell bodies. While this supposedly passive role did not attract multidisciplinary research on the neuroglia, successful efforts were made to extend our knowledge of the physiology, morphology, and bio chemistry of neurons. As a result of this, the investigation of the neuroglia carried out in the first half of this century was mainly confined to morphology, often as a by-product of comprehensive analyses of neuronal systems. At any rate, the histological classification of the neuroglia was accomplished, laying a framework which has been used to the present day. Accordingly, the glia was divided into two major groups: the macro- and microglia. The former comprises two further subclasses, the astroglia and oligodendroglia.

The subject matter is divided into three main categories: 1. The "acquired" lesions dating to the fetal, perinatal or early postnatal periods, 2. the malformations, and 3. the heritable metabolic defects. The first 6 chapters (2-7) are reserved to the lesions most intimately linked to the circumstances of birth. There is some inherent ambiguity in distinguishing between "acquired" lesions and malformations, as, indeed, no sharp distinction can be made between one and the other. Many malformations result from diseases acquired during fetal life and their peculiarity resides in the fact that the organ becomes affected before its development terminates and in such a way that its subsequent development becomes deranged or partly abrogated. A variety of causes acting at the same developmental period or over a common pathogenetic mechanism may produce the same type of derangement, including chemical, physical, infectious or genetic factors, as pointed out repeatedly in the text. Consequently, the definition of a malformation, as differing from an "acquired" residual lesion was made dependant on evidence for the derangement of developmental pro cesses subsequent to the acquisition of the disease.

This stain targeted intermediate filaments that we now know consist mainly of glial fibrillary acidic protein, a protein used today as an astrocytic marker. Cajal described the morphological diversity of these cells with some ast- cytes surrounding neurons, while the others are intimately associated with vasculature. We start the book by discussing the heterogeneity of astrocytes using contemporary tools and by calling into question the assumption by classical neuroscience that neurons and glia are derived from distinct pools of progenitor cells. Astrocytes have long been neglected as active participants in intercellular communication and information processing in the central nervous system, in part due to their lack of electrical excitability. The follow up chapters review the “nuts and bolts” of ast- cytic physiology; astrocytes possess a diverse assortment of ion channels, neu- transmitter receptors, and transport mechanisms that enable the astrocytes to respond to many of the same signals that act on neurons. Since astrocytes can detect chemical transmitters that are released from neurons and can release their own extracellular signals there is an increasing awareness that they play physiological roles in regulating neuronal activity and synaptic transmission. In addition to these physiological roles, it is becoming increasingly recognized that astrocytes play critical roles during pathophysiological states of the nervous system; these states include gliomas, Alexander disease, and epilepsy to mention a few.
