Book excerpt: This book summarizes the NATO Advanced Research Workshop (ARW) on “Nanoengineered Systems for Regenerative Medicine” that was organized under the auspices of the NATO Security through Science Program. I would like to thank NATO for supporting this workshop via a grant to the co-directors. The objective of ARW was to explore the various facets of regenerative me- cine and to highlight role of the “the nano-length scale” and “nano-scale systems” in defining and controlling cell and tissue environments. The development of novel tissue regenerative strategies require the integration of new insights emerging from studies of cell-matrix interactions, cellular signalling processes, developmental and systems biology, into biomaterials design, via a systems approach. The chapters in the book, written by the leading experts in their respective disciplines, cover a wide spectrum of topics ranging from stem cell biology, developmental biology, ce- matrix interactions, and matrix biology to surface science, materials processing and drug delivery. We hope the contents of the book will provoke the readership into developing regenerative medicine paradigms that combine these facets into cli- cally translatable solutions. This NATO meeting would not have been successful without the timely help of Dr. Ulrike Shastri, Sanjeet Rangarajan and Ms. Sabine Benner, who assisted in the organization and implementation of various elements of this meeting. Thanks are also due Dr. Fausto Pedrazzini and Ms. Alison Trapp at NATO HQ (Brussels, Belgium). The commitment and persistence of Ms.

Book excerpt: Regenerative medicine is an interdisciplinary field which has advanced with the use of biotechnologies related to biomaterials, growth factors, and stem cells to replace or restore damaged cells, tissues, and organs. Among various therapeutic approaches, cell-based therapy is most challenging and exciting for both scientists and clinicians pursuing regenerative medicine. Specifically, stem cells, including mesenchymal stem cells and adipose-derived stem cells, are promising candidate cell types for cell-based therapy because they can differentiate into multiple cell types for tissue regeneration and stimulate other cells through neovascularization or paracrine signaling. Also, for effective treatment using stem cells, the tissue engineered constructs, such as bioactive degradable scaffolds, that provide the physical and chemical cues to guide their differentiation are incorporated with stem cells before implantation. Also, it was previously demonstrated that tissue-engineered matrices can promote tubulogenesis and differentiation of stem cells to vascular cell phenotypes. Hence, during tissue regeneration after stem cell therapy, there are numerous factors that need to be monitored. As a result, imaging-based stem cell tracking is essential to evaluate the distribution of stem cells as well as to monitor proliferation, differentiation, and interaction with the microenvironment. Therefore, there is a need for a stem cell imaging technique that is not only noninvasive, sensitive, and easy to operate, but also capable of quantitatively assessing stem cell behaviors in the long term with high spatial resolution. Therefore, the overall goal of this research is to demonstrate a novel imaging method capable of continuous in vitro assessment of stem cells as prepared with tissue engineered constructs and noninvasive longitudinal in vivo monitoring of stem cell behaviors and tissue regeneration after stem cell implantation. In order to accomplish this, gold nanoparticles are demonstrated as photoacoustic imaging contrasts to label stem cells. In addition, ultrasound and photoacoustic imaging was utilized to monitor stem cells and neovascularization in the injured rat tissue. Therefore, using these methods, tissue regeneration can be promoted and noninvasively monitored, resulting in a better understanding of the tissue repair mechanisms following tissue injury.

Book excerpt: In regenerative medicine, tissue engineers largely rely on destructive and time-consuming techniques that do not allow in situ and spatial monitoring of tissue growth. Furthermore, once the therapy is implanted in the patient, clinicians are often unable to monitor what is happening in the body. To tackle these barriers, optical techniques have bee

Book excerpt: The Tissue Engineering approach has major advantages over traditional organ transplantation and circumvents the problem of organ shortage. Tissues that closely match the patient’s needs can be reconstructed from readily available biopsies and subsequently be implanted with minimal or no immunogenicity. This eventually conquers several limitations encountered in tissue transplantation approaches. This book serves as a good starting point for anyone interested in the application of Tissue Engineering. It offers a colorful mix of topics, which explain the obstacles and possible solutions for TE applications.

Book excerpt: Over the past decade, significant advances in the fields of stem cell biology, bioengineering, and animal models have converged on the discipline of regenerative medicine. Significant progress has been made leading from pre-clinical studies through phase 3 clinical trials for some therapies. This volume provides a state-of-the-art report on tissue engineering toward the goals of tissue and organ restoration and regeneration. Examples from different organ systems illustrate progress with growth factors to assist in tissue remodeling; the capacity of stem cells for restoring damaged tissues; novel synthetic biomaterials to facilitate cell therapy; transplantable tissue patches that preserve three-dimensional structure; synthetic organs generated in culture; aspects of the immune response to transplanted cells and materials; and suitable animal models for non-human clinical trials. The chapters of this book are organized into six sections: Stem Cells, Biomaterials and the Extracellular Environment, Engineered Tissue, Synthetic Organs, Immune Response, and Animal Models. Each section is intended to build upon information presented in the previous chapters, and set the stage for subsequent sections. Throughout the chapters, the reader will observe a common theme of basic discovery informing clinical translation, and clinical studies in animals and humans guiding subsequent experiments at the bench.
