Book excerpt: This thesis is focused on the design and investigation of ellipticine derivatives as potential anti-cancer agents. Using a modified synthetic route, the total synthesis of the natural product ellipticine was improved leading to overall yields of 14% compared with previous yields of 3%. Furthermore, of the twenty eight novel ellipticine derivatives synthesized for biological evaluation, ten were found to inhibit the topoisomerase II enzyme with a greater potency than that of the parent ellipticines. The inhibition of topoisomerase enzymes is a significant target for cancer chemotherapy. As part of this research, the complex mechanisms of topoisomerase I were investigated. Through the use of molecular modeling, the mechanistic duality surrounding certain benzo[c]phenanthridines is explained. Chapters one and two summarize the literature background regarding the total synthesis and derivatisation of ellipticine along with the anti-cancer mechanisms of action. The third chapter lists the aims and objectives regarding both the chemical synthesis and biological evaluation of this project. The fourth chapter provides a detailed discussion of the total synthesis of ellipticine, ellipticine derivatives and ellipticinium salts undertaken during the course of this research. The improvement of the Vilsmeier-Haack reaction of 3-formyl-1,4,-dimethylcarbazole is central in obtaining improved overall yields of ellipticine. Formation of ellipticinium salts is also discussed along with a comprehensive investigation of the 1H NMR spectra relating to these compounds. The ability of 9 formylellipticine to form adducts with biological nucleophiles was also studied by MS and LCMS analysis, leading to the identification of adduct formation with the amino acid cysteine. Chapter five discusses the biological evaluation of ellipticine derivatives synthesized during the course of this research. Dual topoisomerase inhibition was one of the key aims of this project. Investigation of these compounds was achieved using topoisomerase relaxation, cleavage and decatenation assays. Chapter six provides full experimental details relating to the total synthesis of these compounds, many of which have previously been unreported. Chapter seven describes the molecular modeling of the interaction of benzo[c]phenanthridines, fagaronine and ethoxidine, on topoisomerase I. The ternary complex of drug-DNA-enzyme is comprehensively investigated leading to a reasonable hypothesis as to why fagaronine acts as a DNA poison and ethoxidine is a DNA suppressor.
