Book excerpt: Microparticles are one of the key novel drug delivery systems has been widely used to precisely modulate release rate. Microparticles based polymeric systems fabricated using suitable carrier has been extensively explored as an effective matrix for controlled and sustained release delivery of many drugs. With the controlled release systems, the rate of drug release matches the rate of drug elimination, and therefore the drug concentration is within the therapeutic window for the majority of the 24-hr period. The aim of this study was to prepare Eudragit microspheres containing Tramadol HCl by solvent evaporation method to achieve a controlled drug release profile. Investigation of the effect of various processing and formulation factors such as polymer type, drug: polymer ratio, stirring speed to obtain spherical particles. Then yield of production, shape, and mean particle size, particle size distribution, encapsulation efficiency, surface properties and release rate of drug from the microspheres were performed.

Book excerpt: This book will describe current research on drug delivery systems that encompass four broad categories, namely: routes of delivery, delivery vehicles, payload, and targeting strategies. Where appropriate delivery vehicles and relevant release of specific agents in any of these categories in clinical application will be discussed. All chapters will highlight the translational aspects of the various technologies discussed and will provide insights into the advantages of such delivery systems over current ones in clinical or research use. Each technology reviewed in this book will have significant potential to improve patients' lives by enhancing the therapeutic efficacy of drugs. This book: Discusses the various factors that mitigate effective oral insulin delivery and the current status of research efforts to overcome these barriers along with recent clinical projections Examines the advantages and disadvantages of each drug delivery system Examines the standard method of accomplishing controlled drug release through the incorporation of the drugs within polymeric biomaterials such as capsules and microcapsules as well as other vehicles such as liposomes Discusses various controlled drug delivery systems, including sustained release delivery systems and pulse or delayed release, e.g. to target different regions of the gastrointestinal tract. In view of these wide-ranging technological areas, and the up-to-date discussions of opportunities and challenges associated with these applications, the book should provide readers from technology, materials science, pharmacology and clinical disciplines with very valuable information.

Book excerpt: This book approaches the subject from a mechanistic perspective that pitches the language at a level that is understandable to those entering the field and who are not familiar with its common phrases or complex terms. It provides a simple encapsulation of concepts and expands on them. In each chapter the basic concept is explained as simply and clearly as possible without a great deal of detail, then in subsequent sections additional material, exceptions to the general rule, examples, etc., is introduced and built up. Such material was generously supplemented with diagrams; conceptually elegant line diagrams in two or three colors. The artwork was well thought out and able to condense the scientific principles into a novel and visually exciting form. The diagrams encourage browsing or draw the reader to salient points. In addition, the technique of highlighting key concepts in a separate box is used throughout each chapter.

Book excerpt: Any drug given in the form of parenteral dosage form like emulsion, suspension, solution etc. it is not best suited for the long time therapy. Long time drug release profile in controlled manner achieved by nano particle which is difficult by parenteral suspension or parenteral solution. Nano particulate drug delivery is the novel concept now a days. Nano particulate drug delivery is used to get maximum bio-availability for the potent drug with minimal side effect. Nano particulate drug delivery is also useful for maintaining the drug concentration in therapeutic window for the drug having narrow therapeutic window. Nano particulate drug delivery having disadvantage like cost effective even though it is widely useful because of its ease of preparation and suitability with most of drugs and excipients.

Book excerpt: Abstract: We report a novel soft lithography based technique to fabricate non-spherical biodegradable polymeric microparticles of different sizes and shapes as drug delivery systems. Geometrical control over the shape and size of these microparticles renders them different aerodynamic and fluidic dynamic properties when compared to conventional spherical microparticles and may prove to be beneficial in certain drug delivery strategies, such as pulmonary and intravenous routes. The surface morphology of the particles was studied using a scanning electron microscope and size distribution of the particles was determined using a Coulter counter. The process is reproducible and millions of uniform biodegradable particles of various sizes and shapes with dimensions ranging from 2-30 um have been fabricated and were collected by a simple vacuum filtration apparatus. In addition, we demonstrated encapsulation of a model drug (FITC), and FITC distributions within the particles were studied by confocal microscopy. Particle size is a critical parameter for several aspects of controlled drug delivery including control of drug-release kinetics, passive targeting to specific cell or tissue types, biodistribution upon administration and available routes of administration. In conclusion, the uniform, biodegradable polymeric microparticles produced have potential to be used in a variety of drug delivery applications and polymer-based microfabrication technology holds promise to produce sophisticated, multi-functional drug delivery devices.

By endowing drugs with new and innovative therapeutic benefits, drug delivery systems extend products’ profitable lifecycle, giving pharmaceutical companies competitive and financial advantages, and providing patients with improved medications. Formulation development is now being used to create new dosage forms for existing products, which not only reduces the time and expense involved in new drug development, but also helps with regard to patent protection and bypassing existing patents. Today’s culture demands convenience, a major factor determining adherence to drug therapy. Over the past few years, patient convenience-oriented research in the field of drug delivery has yielded a range of innovative drug-delivery options. As a result, various drug-delivery systems, including medicated chewing gums, oral dispersible tablets, medicated lozenges and lollipops, have now hit the market and are very popular. These dosage forms offer a highly convenient way to dose medications, not only for special population groups with swallowing difficulties, such as children and the elderly, but for the general populace as well. This book provides valuable insights into a number of formulation design approaches that are currently being used, or could be used, to provide new benefits from existing drug molecules.

Book excerpt: This volume is a thorough presentation of the state-of-the-art research and developments in drug delivery systems using nanotechnology and its applications. The second of this two-volume set, it addresses the applications of nanotechnology or nano-sized materials in the medical field and the real-world challenges and complexities of current drug delivery methodologies and techniques. This volume includes 11 chapters that focus on the targeting facet of drug delivery systems. Targeting is a focused maneuver to achieve the specified goals, including achieving the desired result and reaching the specific location. Targeting has now been successfully achieved for several diseases/disorders; however, its role is noteworthy in cancer treatment where chemotherapy is a main course of approach. Nanotechnology-based products have great potential by virtue of their inherent features. This edited book provides a detailed application of nanotechnology in drug delivery systems in health care. The book discusses general principles of drug targeting, material of construction and technological concerns of nanoparticles, and different drug delivery systems and their preparation. Taken together, the informative chapters will provide researchers and scientists as well as faculty and students with valuable research on the effective use of new approaches in advanced drug delivery nanosystems. Volume 1 of the two-volume series is subtitled Novel Drug Carriers. The volumes are available separately or as a set.
