Screeninq of large numbers of natural and semisynthetic compounds resulted in the characterization of certain sub stances that had a limited efficiency as antiviral druqs. A few chemically synthesized compounds were also found to be effective as antiviral agents in the chemotherapy of human virus diseases. A major difficulty in the develop ment of effective antiviral agents has been the lack of selectivity. and toxicity for uninfected cells. of drugs that effectively inhibited virus replication in vitro. Further understanding of the molecular processes of virus replication in infected cells has resulted in the development of new antivirals directed at virus-coded enzymes or proteins. Recent studies on antivirals that are activated by the herpes simplex virus type l-coded thy midine kinase from a prod rug to an antiviral drug have opened new directions in the development of effective antiviral drugs. The present book deals with a number of antiviral drugs effective against herpes simplex viruses and provides some insight into the molecular aspects of virus replication. It also throws light on the new approaches to the development of antiviral drugs. The molecular basis of the antiviral activity of new and known drugs and their possible use in chemotherapy of viral disease are presented in this book.

It has led to major revisions in concepts of cellular immunity and theories on the recovery of multicellular systems from viral infection. Since its discovery, interferon has held the interest of the molecular biologist, and definitive answers as to its clinical value are close at hand. The following treatise is an attempt by the authors to pre sent a complete picture of the many aspects of interferon. Having recently had the privilege of visiting the laboratories of the au thors and several others of our Soviet colleagues working in the interferon field, I was most impressed with the amount of research being done in this area. Since a great deal of this work is published in the Russian language, there is unfortunately a time lag until it can be received and translated. We are therefore most grateful to Drs. It succeeded a similar Institute held at Porte Conte, Sardinia in 1975 on "Specificity in Plant Diseases. " What are active defence mechanisms in the context of plant disease in which a plant, the host, may be damaged by a pathogen? Defence mechanisms comprise properties of the host that decrease this damage. The mechanisms are passive when they are independent of the pathogen. They are active when they follow changes in the host caused by the pathogen. Thus for a fungal pathogen, cell walls of a higher plant which are lignified before infection would be a passive defence mechanism if they decreased damage by impeding growth of the fungus. Cell walls known to become lignified as a response to the pathogen would be an active defence mechanism if it were established that this response decreased damage. The papers and discussions at this Advanced Study Institute were about active defence mechanisms in higher plants, mainly econo mically important crop plants, against fungi, bacteria and viruses as pathogens. Taking the microorganisms first it is a truism but one that bears repeating that although plants almost always grow in close association with a wide variety of fungi and bacteria, often of types that can be pathogens, they rarely become diseased, at least not sufficiently so as to attract notice.
