Book excerpt: [Phytophthora infestans], the oomycete potato and tomato pathogen which has caused substantial crop losses since the Irish Potato Famine, has high genomic variability and is acutely responsive to external environmental signals. These signals drive adaptation and changes in virulence which are related to gene expression. In my dissertation, I addressed 4 areas of research with unique features, but with the aforementioned overarching theme. i) I assessed the sensitivity of [P. infestans] isolates from naturally-occurring infections in Wisconsin fields to the fungicide mefenoxam. Isolates of clonal lineages US-8 and US-23 both demonstrated resistance and susceptibility to the fungicide. All isolates with a moderate level of tolerance had greater variability among replicates than those with high or low levels of tolerance. ii) In a collaborative project, contact reflectance spectroscopy was used to discriminate between infections of US-8 and US-23. We found that during early infection, US-8 has a significantly higher expression of the [Avr3a] effector gene. iii) I investigated the impact of continuous sub-culturing of [P. infestans] on Rye Agar A on virulence of isolates of [P. infestans] US-8.
