However, for many of them efficient vaccines are not yet available. This will probably change dramatically with the current Covid-19 pandemic, as a vast variety of vaccinology approaches are being tested against it, with hundreds of candidates under development, dozens of them already in clinical trials, a fact that is breaking records in vaccine development and implementation. This is becoming possible thanks to the enormous work carried out during years to have the bases for a quick response, even against unknown pathogens, in an impressive short time. Here, results obtained with different vaccine´s methodological approaches against human (HIV, HCV, HRV) animal (PRRSV, PEDV, FMDV, VHSV) and zoonotic (RVF, WNV), RNA viruses are presented by field experts.

Book excerpt: Abstract : Conventional vaccinology uses live attenuated viruses as well as inactivated viruses as vaccines. Live attenuated vaccines are very immunogenic and offer long lasting immunity to infectious agent but there cannot be used for everyone especially in patients with a compromised immune system. Inactivated vaccines are very safe and can be used to immunize everyone regardless of their immune status. However, unlike attenuated vaccines, their immune response is not long lasting. Given the limitations of conventional vaccinology, reverse vaccinology has recently been explored as an alternative approach to design safe and immunogenic vaccines against viral infections. Some regions in the envelope or the capsid of viruses have been predicted/shown to elicit neutralizing/protective antibodies against viruses. Reverse vaccinology is aimed at developing safe vaccines targeting only these regions (epitopes) on the envelope or the capsid of viruses. Although this approach is very safe, the epitopes (peptide antigens) used in developing these vaccines are less immunogenic due to the size, morphology, and the geometry of the peptide antigen in question. In this dissertation, I assessed the immunogenicity of peptide antigens derived from two mosquito-borne RNA virus, Zika virus (ZIKV) and Chikungunya virus (CHIKV), on a highly immunogenic bacteriophage platform known as virus-like particles (VLPs). Currently, there are no approved vaccines against ZIKV and CHIKV. In this dissertation, I showed that ZIKV and CHIKV peptides displayed on bacteriophage VLPs elicited high antibody titers even at a dose of 5 mg of the VLPs displaying the peptides. Additionally, sera from mice immunized with bacteriophage VLPs displaying ZIKV peptides neutralized ZIKV from infecting monkey kidneys cells. Bacteriophage VLPs is an excellent approach to enhance the immunogenicity of ZIKV and CHIKV peptide antigens. Because VLPs resemble viruses in terms of shape, size, morphology exception that, they lack the viral genome (do not replicate), there are excellent platforms to develop safe and effective peptide vaccines.

Book excerpt: Zika virus (ZIKV) is a member of the family Flaviviridae which includes many other important mosquito-borne viruses such as dengue virus (DENV), West Nile virus (WNV), and yellow fever virus (YFV). Since it was discovered in 1947, sporadic outbreaks of ZIKV have been reported in Africa and Asia. Historically people infected with ZIKV often have no or mild symptoms. However, in 2015 a ZIKV pandemic began in South America, Central America, the Caribbean, and the USA, suddenly becoming a global public health issue. Importantly, ZIKV from these recent outbreaks can cause Congenital Zika Syndrome (including microcephaly), Guillain-Barre syndrome, and other severe neurological disorders. Thus, a safe and efficacious vaccine is urgently needed for ZIKV. Similar to other flaviviruses, the ZIKV genome encodes a single polyprotein that is cleaved into three structural proteins (capsid, C; premembrane, prM, and envelope, E) and seven nonstructural proteins. The E protein mediates cellular attachment and membrane fusion, and is the target for most neutralizing antibodies. Flavivirus prM protein typically associates with E to form heterodimers and is important for proper folding of E. Therefore, prM and E proteins have been the primary targets for subunit and recombinant ZIKV vaccine studies. Meanwhile, the nonstructural protein NS1 of several flaviviruses has been shown to confer some protection against flavivirus infection in animal models in the absence of detectable neutralizing antibody. Whether immunization with the ZIKV NS1 protein has certain protective capabilities is unclear. The goal of my research is to develop vesicular stomatitis virus (VSV) and measles virus (MeV)-based vaccine candidates for ZIKV. Both VSV and MeV are non-segmented negative-sense (NNS) RNA viruses. VSV is an animal virus and has been shown to be an excellent vaccine vector because of its high genetic stability and expression efficiency and no pre-existing immunity in the human population. In 2019, a VSV-based Ebola virus vaccine was approved by FDA for human uses. MeV is another excellent vector to deliver vaccines for human pathogens primarily because of its high safety and efficacy even in the presence of pre-existing anti-MeV vector immunity. Several MeV-based vaccine candidates are highly efficacious in human clinical trials. In Chapter 2, I developed a methyltransferase (MTase)-defective recombinant VSV (mtdVSV)-based ZIKV vaccine platform. The full-length E, E truncation mutants, prM-E, NS1, and prM-E-NS1 of ZIKV were inserted individually into a VSV vector that carrying an MTase mutation in the large polymerase protein.
