In "radical relay" C-H functionalization reactions, a C(sp3)-H bond can be abstracted by an H atom transfer (HAT) reagent to form an alkyl radical. It has been demonstrated that alkyl radicals can be cross coupled with nucleophilic coupling partners in the presence of transition metal catalysts or reacted directly with compatible radical traps to form new bonds. Such oxidative coupling reactions allow bond construction from otherwise unreactive C-H sites enabling conversion of simple molecular building blocks into value added product libraries and generation of structural analogues of important molecules that can be tested for improved properties. This thesis discloses the development, mechanistic understanding, and application of several oxidative cross coupling methods including C(sp3)-H arylation, fluorination, and methylation reactions as well as an aerobic oxidative amidation reaction between alcohols and amines. Collectively, these methods contribute to the breadth of oxidation reactions available for the synthesis and functionalization of diverse molecules. The C-H arylation, fluorination/fluoride displacement, and amide coupling all are opportunities to apply oxidative coupling directly to the synthesis of important molecules. Complementarily, the fluorination and methylation reactions present opportunities to functionalize existing molecules to allow derivatized compositions of matter to be accessed for testing. Both forms of methodology are appealing for application in synthesis-driven specialties, where rapid and direct access to new structures is essential to solving complex challenges

Consequently, the development of new methods to rapidly construct diverse, complex molecules has become of increasing importance in the field of synthetic organic chemistry in recent decades. To address these needs, our group leverages the merger of transition metal catalysis and photocatalysis (i.e., metallaphotocatalysis) toward the development of new coupling and activation platforms. The ability of photocatalysis to selectively activate organic substrates via single electron transfer pathways to generate open-shell species under mild conditions, coupled with transition metal cross-coupling, has led to the development of a diverse and powerful array of synthetic manifolds that enable previously elusive bond disconnections and streamline the synthesis of molecules. Although this field has advanced rapidly in the past decade, the development of novel metallaphotocatalytic methods that broadly enable the coupling of diverse fragments continues to be in high demand. This thesis details several efforts that employ metallaphotocatalysis in the development of novel cross-coupling methods and activation modes. First, Chapter 2 describes a new approach for nickel/photoredox-catalyzed cross-electrophile coupling of activated alkyl chlorides and aryl halides, facilitated by silyl radical-mediated halogen atom abstraction as a key design element. Chapter 3 extends this design element to the activation of C(sp2)-Br bonds which, in combination with dual copper/photoredox catalysis, enables a general protocol for trifluoromethylation of aryl bromides. Finally, Chapter 4 describes a novel ligand-to-copper charge transfer platform that promotes the decarboxylative functionalization of aryl carboxylic acids (a modular and abundant yet recalcitrant class of substrates), rendering a previously inaccessible range of aryl carboxylic acids broadly accessible as adaptive functional handles.

During the past several decades, phosphorus containing ligands have been extensively used in transition metal catalyzed C-C and C-H bond forming reactions. Development of new phosphine ligands for palladium cross coupling and also methodology for C-H activation strategies will be the focus of this dissertation. A variety of triazole containing monophosphine ligands have been prepared via efficient 1,3-dipolar cycloaddition of readily available azides and acetylenes. Their palladium complexes provided excellent yields in the amination reactions (up to 98% yield) and Suzuki-Miyaura coupling reactions (up to 99% yield) of unactivated aryl chlorides. A CAChe model for one of the Pd-complexes shows that the likelihood of a Pd-arene interaction might be a rationale for its high catalytic reactivity. A main goal for Organic chemists is to develop and utilize efficient and atom-economical methods for the elaboration of complex structures from simple and readily available starting materials. C-H bonds are the most fundamental linkage in organic chemistry and recently tremendous strides have been have been made in the functionalization of C-H bonds. A central goal in the development of any new methodology is synthetic utility, which has been difficult to achieve with C-H activation strategies because of the inherent stability of C-H bond. Aryl carboxylic acid derivatives are very prevalent in industrial and pharmaceuticals and thus a direct C-H activation approach would be very desirable. A general protocol for the rhodium-catalyzed oxidative carbonylation of arenes to form esters has been developed. A broad substrate scope has been demonstrated allowing carbonylation of electron rich, electron-poor, and heterocyclic arenes, and the reaction shows wide functional group tolerance and excellent regioselectivities. Up to 96% yield of ortho-substituted aryl or heteroaryl carboxylic esters were obtained with this methodology. The possible mechanism for the rhodium-catalyzed oxidative carbonylation reaction was proposed in this article. Studies show that Oxone play an important role in the transformation. We have developed a new C2-symmetric monophosphine ligand based upon a C3* tunephos backbone. The ligand was available in several steps from commercially available starting materials. In future studies this ligand was be tested for its use in chiral cross coupling reactions.

Over the past 30 years, the protocol has been substantially improved and expanded to other coupling reactions of Li,B,N,O,Al,Si,P,S,Cu,Mn,Zn,In,Sn, and Hg compounds. These reactions provided an indispensable and simple methodology for preparative organic chemists. Due to the simplicity and rel- bility in the carbon-carbon, carbon-heteroatom, and carbon-metalloid bo- formations,as well as high efficiency of the catalytic process,the reactions have been widely employed by organic chemists in various fields. Application of the protocol ranges from various syntheses of complex natural products to the preparation of biologically relevant molecules including drugs, and of sup- molecules, and to functional materials. The reactions on solid surfaces allow robot synthesis and combinatorial synthesis. Now, many organic chemists do not hesitate to use transition metal complexes for the transformation of org- ic molecules. Indeed, innumerable organic syntheses have been realized by the catalyzed reactions of transition metal complexes that are not achievable by t- ditional synthetic methods. Among these, the metal-catalyzed cross-coupling reactions have undoubtedly contributed greatly to the development of such a new area of “metal-catalyzed organic syntheses”. An excellent monograph for the cross-coupling reactions and other met- catalyzed C-C bond-forming reactions recently appeared in Metal-catalyzed Cross-coupling Reactions (Wiley-VCH,1998).
