Book excerpt: Despite hypothyroxinemia, PFDA-treated rats are not functionally hypothyroid. Furthermore, any alteration in functional thyroid status can be dissociated from the overt toxicity (i.e., severe hypophagia and body weight loss). PFDA exerts effects on neutral lipid metabolism in both liver and carcass of the rat. At 7 days following a single administration of PFDA, hepatic esterification of free fatty acid into TG and CE was increased yet the expected augmentation in the export of these neutral lipids from liver into plasma was absent. An efficient chemical method was developed for the purification of commercially available PFDA that contains contaminants of mono- and diprotio-substituted materials. PFDA of high specific activity has been synthesized with 14 C-labeling in the C-1 position. In vivo experiments indicate that perfluorinated acid derived radioactivity found in rat tissues behaves similarly to PFDA or PFOA added directly to frozen rat tissue. Hepatic oxidation of long-chain medium-chain and short-chain fatty acids was unaffected by pretreatment with PFDA or PFOA. Similarly, esterification was not affected.

Experiments were conducted to determine the LD50 and to evaluate survival times of rats after single intraperitoneal (IP) injections of NDFDA or of Fluorooctanoic acid (PFOA), the shorter-chain analog, dissolved in propylene glycol/water (1:1, v:v). Ten male Fischer 344 rats were used in each dose group and an additional 10 rats were used in each dose group and an additional 10 rats were given 2 mL/kg propylene glycol/water, the same dosing volume used with all experimental animals. With NDFDA, the LD50/14 days was 63.6 mg/kg, and the LD50/30 days was 41.4 mg/kg. With PFOA, there was no mortality after the 5th day following injection, and both the LD50/14 and LD50/30 were 188.7 mg/kg.

Book excerpt: This AASERF grant has provided support for two graduate students in the Biomedical Sciences Ph. D. Program at Wright State University, Dayton, OH. A Ph. D. degree was conferred in 1994 to C. Goecke- Flora, while the other student, M. Adinehzadeh, currently continues in the program. These students were provided a stipend, a nominal budget for research supplies, and travel funds which enabled them to present their data at national scientific conferences. The students utilized nuclear magnetic resonance techniques in toxicology research concerning peroxisome proliferators (PPs). This work has culminated in five publications and eight published abstracts. PPs are a diverse series of chemicals with various commercial and Air Force applicabilities. Perfluorocarboxylic acids and various other halocarbons, for instance, are useful as nonflammable solvents, lubricants, and degreasers. Our research has shown that the berfluorocarboxylic acids are not metabolized, yet they produce significant effects on liver carbohydrate and phospholipid metabolism. Perfluorodecanoic acid, in particular, enhances the biosynthesis of liver phosphatidylcholine (PC), causes a significant increase in phosphocholine and diacylglycerol, and inhibits the major de novo pathway for the synthesis of PC. These data suggest that other pathways of phospholipid metabolism are activated following treatment with these PPs. Such effects may alter cellular signaling processes and help to explain the hepatotoxicity and carcinogenicity associated with these compounds.

Exposure to PFAS is suspected to increase the risk of adverse health effects, such as impacts on the thyroid gland, the liver, fat metabolism and the immune system. This study estimates the socioeconomic costs that may result from impacts on human health and the environment from the use of PFAS. Better awareness of the costs and problems associated with PFAS exposure will assist decision-makers and the general public to make more efficient and timely risk management decisions. Findings indicate that the costs are substantial, with annual health-related costs estimated to 2.8 – 4.6 billion EUR for the Nordic countries and 52 – 84 billion EUR for all EEA countries. Overall non-health costs are estimated at 46 million – 11 billion EUR for the Nordic countries.Upon request the excel spreadsheets used for the monetarisation and valuation in this report can also be provided along with a guidance on how to use the estimation of costs for value transfer.
