Book excerpt: "Lignans encompass a structurally diverse array of highly oxygenated, polyaromatic natural products with important biological activities. Despite their structural diversity, lignan biosynthesis begins by the oxidative coupling of only a small number of relatively simple propenyl phenols. Previous attempts to mimic this oxidative coupling have suffered from poor regio- and chemoselectivity. Other approaches to these compounds have proven efficient and relatively diversifiable but they have often been designed to access a single structural class and do not capitalize on the divergency of the biosynthesis. This thesis details a bio-inspired method that aims to solve the challenge of selectivity in the oxidative coupling. By coupling two propenyl phenols through a [2+2]-cycloaddition, the key carbon-carbon bond is formed prior to the oxidation. A subsequent oxidative fragmentation of the resulting cyclobutane allows us to intercept the proposed biosynthetic intermediate with high levels of control. We have demonstrated the utility of this method in the synthesis of several natural and non-natural 2-aryl-4-benzyltetrahydrofuran and furofuran-based lignans. To expand this strategy to access other structural classes of lignans, we developed a complementary approach. The corresponding dibenzylbutane was obtained by cleaving the diarylcyclobutane bond through a reductive ring opening. Through selective functionalization and oxidative cyclization, several aryltetralins and dibenzocyclooctadienes were afforded. This provided a unified approach to this diverse class of natural products, in which molecular complexity is rapidly increased." --

Book excerpt: Lignans are a large class of natural products defined as the products of an oxidative dimerisation of two or more phenylpropanoid units. 1,4-Benzodioxane neolignans are a significant subtype of lignan natural products that showcase a range of biological activities. Previous syntheses of 1,4-benzodioxane lignans are predominantly racemic and do not easily allow for the main points of variation between related and analogous products. This thesis focuses on the development of a flexible asymmetric synthesis of 1,4-benzodioxane compounds and its subsequent application to natural product synthesis. Initially, a model study was undertaken to investigate the key steps required in this synthesis. Following the model study, the developed synthesis was then applied to the first asymmetric synthesis of the rodgersinine family, successfully yielding all rodgersinine natural products 21, 83, 84 and 87 and their enantiomers. Through chiroptical analysis, it was found that the original assignment of 83 and 87 was incorrect, and the ECD comparison of 1,4-benzodioxane natural products and model compounds is not reliable. Members of the eusiderin family, eusiderins A (19), B (243), C (194), G (37), L (260) and M (245) and analogues were synthesised in a highly divergent manner from chiral aldehyde 259. An ECD study also confirmed the absolute stereochemistry of 19 and 194 from different natural sources, and provided a reference for stereochemical assignment of future eusiderins. The adaptation of the synthesis to 2-hydroxymethyl-1,4-benzodioxane neolignans was shown with the successful synthesis of (-)-isoamericanin A (24) and (+)-isoamericanol A (20). Further application of this synthesis to complex trilignan magnolianin 278 was unsuccessful following the failure to synthesise ether 351 because of steric restrictions. The formal asymmetric synthesis of silybin B (358) and isosilandrin B (364); 1,4-benzodioxane flavonolignans well-regarded for their potent activities, was achieved, demonstrating the applicability of the developed synthesis to this notable group of compounds. Subsequent testing by Eli Lilly and a leading anti-HCV expert, of selected synthesised 1,4-benzodioxane compounds identified them to exhibit activities with applications in areas of cancer and neurological treatments, as well as hepaprotective properties that are currently undergoing further exploration.

Over the course of my PhD studies, I have attempted to mimic certain enzymatic mechanisms of aerobic oxidation, in order to develop new synthetic tools for the preparation of complex, organic molecules. These studies can be divided into two phases, representative of the respective enzymes under consideration. The first focused on the development of Cu-catalysts based upon the Type III, di-copper enzyme tyrosinase. Tyrosinase is an essential enzyme that is part of the melanogenic pathway. At its center are two Cu-atoms that activate molecular oxygen (O2) as a characteristic Cu2-peroxo, in order to de-aromatize L-tyrosine. Inspired by this mechanism of O2-activation and oxygen atom transfer, I developed a series of catalytic aerobic processes for the functionalization of phenols. This included the synthesis of diaryl ethers by dehydrogenative C-O bond formation, as well as a direct synthesis of polyfunctional indoles starting from commercially available derivatives of L-tyrosine.The second phase of my studies focused on aerobic oxidations catalyzed by Fe-containing enzymes in the late stages of lignan biosynthesis. Here, our focus shifted towards the substrate, and we developed conditions to explore the fates of proposed biosynthetic radicals that are implicated in the biosynthesis of lignan-derived dibenzocyclooctadiene (DBCOD) natural products. DBCODs are secondary metabolites produced by medicinal plants of Schisandra and Kadsura that display a range of health benefits. In the final stages of their biosynthesis, certain family members are believed to be oxidized by Fe-containing oxygenases that leads to dramatic structural changes following radical cyclization. Typically, radical cyclizations are much slower than the more typical hydroxylation pathway, raising interesting mechanistic questions about how these enzymes delay or divert more traditional pathways of C-H oxidation. My work in this area led to mild, photoredox mediated conditions for late-stage radical cyclizations that could be applied to a number of highly oxidized and structurally distinct family members"--
