Book excerpt: The past decades have seen major developments in the understanding of the cellular and molecular biology of cancer. Significant progress has been achieved regarding long-term survival for the patients of many cancers with the use of tamoxifen for treatment of breast cancer, treatment of chronic myeloid leukaemia with imatinib, and the success of biological drugs. The transition from cytotoxic chemotherapy to targeted cancer drug discovery and development has resulted in an increasing selection of tools available to oncologists. In this Special Issue of Pharmaceuticals, we highlight the opportunities and challenges in the discovery and design of innovative cancer therapies, novel small-molecule cancer drugs and antibody–drug conjugates, with articles covering a variety of anticancer therapies and potential relevant disease states and applications. Significant efforts are being made to develop and improve cancer treatments and to translate basic research findings into clinical use, resulting in improvements in survival rates and quality of life for cancer patients. We demonstrate the possibilities and scope for future research in these areas and also highlight the challenges faced by scientists in the area of anticancer drug development leading to improved targeted treatments and better survival rates for cancer patients.

Available in PDF, EPUB and Kindle. Book excerpt: Naturally occurring RNA always contains numerous biochemically altered nucleotides. They are formed by enzymatic modification of the primary transcripts during the complex RNA maturation process designated RNA modification. A large number of enzymes catalyzing the formation of these modified nucleosides or converting one canonical base into another at the posttranscriptional level have been studied for many years, but only recently have systematic and comparative studies begun. The functions of individual enzymes and/or the modified/edited nucleosides in RNA, however, have remained largely ignored. This book provides advance information on RNA modification, including the associated editing machinery, while offering the reader some perspective on the significance of such modifications in fine-tuning the structure and functions of mature RNA molecules and hence the ability to influence the efficiency and accuracy of genetic expression. Outstanding scientists who are actively working on RNA modification/editing processes have provided up-to-date information on these intriguing cellular processes that have been generated over the course of millions of years in all living organisms. Each review has been written and illustrated for a large audience of readers, not only specialists in the field, but also for advanced students or researchers who want to learn more about recent progress in RNA modification and editing.

Available in PDF, EPUB and Kindle. Book excerpt: Transfer RNAs (tRNAs) are one of the classical non-coding RNAs whose lengths are approximately 70–100 bases. The secondary structure of tRNAs can be represented as the cloverleaf with 4 stems, and the three dimensional structure as an “L” shape. Historically, the basic function of tRNA as an essential component of translation was established in 1960s, i.e., each tRNA is charged with a target amino acid and these are delivered to the ribosome during protein synthesis. However, recent data suggests that the role of tRNA in cellular regulation goes beyond this paradigm. In most Archaea and Eukarya, precursor tRNAs are often interrupted by a short intron inserted strictly between the first and second nucleotide downstream of the anticodon, known as canonical nucleotide position (37/38). Recently, a number of reports describe novel aspects of tRNAs in terms of gene diversity, for example, several types of disrupted tRNA genes have been reported in the Archaea and primitive Eukarya, including multiple-intron-containing tRNA genes, split tRNA genes, and permuted tRNA genes. Our understanding of the enzymes involved in tRNA functions (e.g., aminoacyl-tRNA synthetase, tRNA splicing endonuclease, tRNA ligase) has deepened. Moreover, it is well known that tRNA possesses many types of base modifications whose enzymatic regulations remain to be fully elucidated. It was reported that impaired tRNA nuclear-cytoplasmic export links DNA damage and cell-cycle checkpoint. Furthermore, a variety of additional functions of tRNA, beyond its translation of the genetic code, have emerged rapidly. For instance, tRNA cleavage is a conserved part of the responses to a variety of stresses in eukaryotic cells. Age-associated or tissue-specific tRNA fragmentation has also been observed. Several papers suggested that some of these tRNA fragments might be involve in the cellular RNA interference (RNAi) system. These exciting data, have lead to this call for a Research Topic, that plans to revisit and summarize the molecular biology of tRNA. Beyond the topics outlined above, we have highlighted recent developments in bioinformatics tools and databases for tRNA analyses.

Solar photons are the unique source of the large amounts of energy likely required to initiate the organisation of matter to produce biological life. The Miller–Urey experiment simulated the conditions thought to be present on the early earth and supported the hypothesis that under such conditions complex organic compounds could be synthesised from simpler inorganic precursors. The experiment inspired many others, including the production of various alcohols, aldehydes and organic acids through UV-photolysis of water vapour with carbon monoxide. This book covers the photochemical aspects of the study of prebiotic and origin of life chemistry an ideal companion for postgraduates and researchers in prebiotic chemistry, photochemistry, photobiology, chemical biology and astrochemistry.

The Combinatorial Index is an answer to that problem. This compendium of methods from the primary literature provides quick and convenient access to reliable synthetic transformations as well as information on linkers and analytical methods. Each synthetic procedure is preceded by a section entitled"Points of Interest,"which highlights the strengths and weaknesses of the various studies. The index also covers the use of solution-based synthesis for the generation of molecular diversity. - Organized for rapid retrieval of published information on classes of synthetic transformations, linkers, and analytical methods - Serves as a laboratory manual for bench chemists - Includes a chapter on linkers to assist in choice of linking strategy - Discusses strengths and limitations of the various methods - Contains a structural index showing functional group transformations in solid-phase synthesis
