Recently, they have been described as playing a critical role in breast cancer metastasis. As all GPCRs interact with arresting, we have sought to better understand the role of arrestin interaction with GPCRs particularly in the process of apoptosis. Previously published data has described the initiation of apoptosis when GPCRs are stimulated in the absence of arrestins. It is therefore our goal to better understand this process and therefore better understand potential roles for GPCRs and arrestins in breast cancer development. From this work, we hypothesize that arrestin can be used as a novel chemotherapeutic target. If arrestins are inhibited, stimulation of GPCRs found on metastatic breast cancer cells may undergo apoptosis therefore providing a new treatment to eradicate breast cancer.

Book excerpt: Cardiovascular disease (CVD) is the most common cause of morbidity and mortality worldwide, putting a major burden on life quality and social health care systems. Type 2 diabetes mellitus (T2DM) and chronic kidney disease (CKD) have been identified as important risk factors for CVD, severely increasing the risk on e.g. myocardial infarction, and cardiovascular complications constitute the main cause of death in patients presenting with T2DM, CKD or a combination of both. As these pathologies are expected to rise alarmingly in the next decades, a better understanding of molecular and cellular mechanisms contributing to T2DM, CKD and CVD is required to improve prevention and treatment of these diseases. Furthermore, insight into the interplay between these pathologies and identification of molecular players interconnecting these comorbidities is of tremendous importance for optimal health management in the future. This Research Topic will focus on the chemokine receptor CXCR4 and its ligands CXCL12/SDF-1a and macrophage migration inhibitory factor (MIF) in the context of CVD and its link with T2DM and CKD, as well as address dipeptidyl peptidase-4 (DPP4) as an important protease destabilizing CXCL12. Chemokines and their receptors are important mediators of cell mobilization, recruitment and arrest, and also more broadly induce cell activation by triggering various intracellular signalling tracks. They control homeostatic conditions, but are also critically involved in inflammatory and pathological processes. Genome-wide association studies revealed single nucleotide polymorphisms connecting CXCL12 as well as MIF with CVD, and a role for both chemokines in T2DM and CKD has also been reported. In this review collection, current knowledge on molecular aspects of the CXCR4 ligand/receptor family and associated signalling pathways will be discussed. The physiological roles of CXCR4, CXCL12, MIF and DPP4 will be summarized, and recent findings on their function in pathological conditions of CVD, T2DM and CKD will be highlighted. This is combined with an extensive introduction providing insight into the pathologies of CVD, T2DM and CKD, discussing clinical features and common pathological aspects of these comorbidities on cellular and molecular level. Also, an overview of available animal models to study these diseases will be provided. This way, this Research Topic summarizes latest knowledge on this crucial molecular axis and its relationship with cardiovascular pathologies for both specialists and interested non-specialists and aims to stimulate further initiatives to unravel the mechanistic involvement of the CXCR4 ligand/receptor family in these morbidities, potentially paving the way for new therapeutical initiatives in the future.

Book excerpt: In 1996 a cDNA called KISS1 (KI in reference to the place of discovery-Hershey Pennsylvania, home of the famous Hershey Chocolate Kisses- and SS as suppressor sequence) was identified in non-metastatic melanoma cell lines. Its 54 amino-acid product, Kisspeptin-54 (Kp-54), was originally called metastin for its ability to inhibit cancer metastasis through the activation of a G coupled receptor, previously known as GPR54 and currently renamed the Kisspeptin receptor (KISS1R). Shorter active peptides all capable of binding to KISS1R have been identified and the physiological activities of these Kisspeptins are now known to extend not only to the suppression of metastasis. Kisspeptins currently represent evolutionarily conserved biological modulators, with a recognized role in the central control of sex maturation, reproduction and fertility. Consequently, the focus on the central role of the Kisspeptins has led to neglecting their possible activities in peripheral tissues. Increasing data reveals that Kisspeptins and KISS1R have a wider expression and possibly a broader spectrum of action in several peripheral tissues such as the gonads, adipose tissue, and liver with direct consequences on gamete quality and fertility rate, pregnancy, energy homeostasis and body weight control. In this respect, the Kisspeptin system may represent a promising prognostic/diagnostic biomarker and therapeutic target for cancer and other human diseases such as infertility and metabolic disorders. This Research Topic provides a comprehensive picture of the recognized and the emerging role of the Kisspeptin system. Targeted delivery relies on two basic approaches, passive and active targeting. While passive targeting approaches have shown great promise, the improved selectivity achieved with active targeting approaches has resulted in significantly higher efficacy. Interestingly there are numerous strategies for active targeting, many of which are already highlighted in , Targeted Drug Delivery: Concepts and Applications. Nevertheless an exciting and practical strategy for active targeting, which could enable high intracellular delivery, is through exploitation of RME. Cells in the body express receptors to enable various physiological and biochemical processes. As a result, many of these receptors are overexpressed in pathological conditions, or newer receptors expressed due to defective cellular functioning. RME is based on exploitation of such receptors to achieve intracellular delivery. While targeted delivery can have manifold applications, in this book we focus on two major and challenging therapeutic areas; i) Cancer and ii) Infectious Diseases. Targeted Intracellular Drug Delivery by Receptor Medicated Endocytosis discusses the major receptors that are useful for targeted delivery for these afflictions.
