Book excerpt: Excerpt from The Journal of Infectious Diseases: Founded by the Memorial Institute for Infectious Diseases My purpose in this address is to give briefly some observations upon the value of the agglutination test in establishing the identity or relationship of bacteria and in detecting the variety of bacteria exciting disease in cases of bacterial infection. During the past three years, in connection with Dr. Katharine R. Collins, I have been more or less occupied in the study of these questions, and in this time I have learned much concerning the difficulties of properly interpreting the results, and of the limitations to the value of the agglutination reaction. It is my hope that a review of some of these experiences may be of interest. Before taking up the discussion of the two topics I wish to touch on some of the points to be thought of in the technique of carrying on the tests. Some Important Points to Consider in Making an Agglutination Test. 1. The quantitative nature of the union between bacteria and agglutinin. This necessitates that with increase in the number of bacteria in the serum dilution there is more material to combine with the agglutinated. A thick emulsion of bacteria is therefore not agglutinated in as high dilutions as a thin emulsion. 2. The varying sensitiveness of the same variety of bacteria from day to day, even when grown from the same stock culture. Following on that comes the proof of their inoculability, that is, of the possibility of their transference from one individual to another previously healthy, this transference being attempted both on individuals belonging to the same species as those in whom the disease arose spontaneously or was artificially produced, and on individuals of other species. Next we have to trace out the mode in which the pathogenic organisms enter the animal body, to follow their behaviour out side the body in the air, water, and soil, and finally to determine what in uence reagents may exert upon them either in the way of destroying them or preventing their development. Hygiene is only interested in their existence in animal tissues in so far as an explanation of the mode of infection may thereby be rendered possible, whether, for example, the pathogenic organisms are localized in the alimentary canal, whether they pass into the blood, or whether they form resting spores in the interstices of the tissues. The experi mental 'proof, ' however, is so conclusive that it must be accepted as an axiom, that no putrefaction can occur without the presence of bacteria, and that bacteria are the sole and only cause of such decomposition. In order to demonstrate this truth, it has been found most convenient to make use of clear solutions. Of organic substances, for instance infusions of animal or vegetable tissues. All that has been learned in this manner applies equally to the decomposition of the original solid substances, but it is not so con venient to demonstrate it. For in transparent solutions. Bacteria can be detected more readily than on the surface of solids. Moreover the occurrence, or absence, of a turbidity in the previously clear ﬂuid, is so sug gestive of the presence of, or freedom from, bacteria, that the microscope need not be appealed to in every' case. The first and most demonstrative result of such observations has been that putrefaction is arrested by any and all means, which arrest the development of bacteria. For the present let us merely consider the employment of heat for that purpose. Like all other living beings bacteria are killed by a sufficient heat. Book excerpt: Excerpt from Microbes and Health In the following pages the author has endeavored to give a rational cause for disease, especially those diseases that are contagious, or those said to be caused by germs. He has endeavored to explain, First, what a germ is, and its relation to man. Second, the various germ theories are presented. These theories are released from all superfluous verbiage; stripped of their delusive coverings, and left to stand or fall according to their merits. Third, and last, there is presented the true medium by which disease is conveyed from one to another, a medium wholly independent of the germ. The author is a firm believer in germs, for germs are the means of carrying on the world's economy, the medium through which all material progress has been made; but it would be unreasonable to say that this medium is destructive to man. Yet, by a system of theorizing, the bacteriologists have actually succeeded in establishing in the minds of some a belief that germs are the cause of disease. The author believes that all such theories have emanated from a hypothesis, an assumed basis which fails to present even a comprehensive guess or a logical supposition.
