The effect of colonization by nonpathogenic P. syringae pv. syringae, was examined under field conditons. Strains of P. syringae nonpathogenic to beans originally obtained from bean leaf surface were introduced into field plots by seed treatment before planting and/or by spray application. Bacterial population sizes on bean leaflets and pods were estimated by dilution plating from bulked samples. The nonpathogenic strains of P. syringae colonized bean leaflets and pods, and reached populations for 10 [elevado a 3] to 10 [elevado a 5] CFU/ leaflet or pod. However, dry weather was apparently unfavorable for the establishment of the nonpathogenic strains on bean plants. The population sizes of the nonpathogenic strains remined below 10 [elevado a 3] CFU/ leaflet under these conditions. The presence of high epiphytic populations of P. syringae-like bacteria (PSLB) (10 [elevado a 6] CFU/leaflet) on bean plants, apparently prevented the establishment of large epiphytic populations (10 [elevado a 3] CFU/leaflet) of the nonpathogenic strains of P. syringae. The nonpathogenic strains failed to reduce the occurrence of bacterial brown spot, in these cases. Book excerpt: The in planta growth of three wild type strains of phytopathogenic bacteria and three nonpathogenic mutant strains was studied to determine if mutations eliminating the ability of the mutant strains to cause disease had affected their growth in leaf tissue. The mutant strains were obtained by Tn5 mutagenesis of Pseudomonas svringae pv. phaseolicola strain PP7010 and P. svringae pv. svringae strain PS9020, organisms which respectively cause halo blight of bean and bacterial brown spot of bean. Through cloning of Tn5-containing genomic DNA, bacterial conjugation, auxanography, and genetic marker exchange experiments, these studies demonstrated that the Tn5 mutation in strain PP7014 of P. svringae pv. phaseolicola had created an auxotrophic requirement for uracil and had caused an inability to produce typical necrotic and chlorotic disease symptoms in bean leaves. A simple and reproducible bioassay was developed to monitor bacterial growth in leaf tissue. Using this bioassay, mutant PP7014 was shown to be incapable of growth in planta, presumably due to its inability to obtain uracil for growth in the leaf. However, a similar Tn5 arginine auxotroph, PP7510, was capable of multiplication in bean leaves, demonstrating that prototrophy of P. svringae pv. phaseolicola is not a prerequisite for pathogenicity. Growth studies with pathogenic wild type P. syringae pv. syringae strains J900 and PS9020 revealed a correlation between growth in planta and the severity of symptom expression. Altered growth patterns and symptom expression were shown for two nonpathogenic strains derived by Tn5. mutagenesis of PS9020. Mutant strain PS9021 was incapable of in planta growth and disease symptom expression. Mutant strain PS9024, although initially capable of rapid multiplication in bean leaves, was unable to maintain the high population levels and produce the characteristic symptoms of the parental wild type strain. Further growth studies were conducted to investigate the complementation of the Tn5 mutation in PS9021 by cosmids containing wild type PS9020 DNA homologous to the mutated region. Although stable throughout multiple rounds of bacterial replication, the cosmids could partially, but not completely, restore the wild type in planta growth and symptom expression pattern to PS9021.

However, common bean is affected by numerous diseases that can negatively impact production. There are three major bacterial diseases of common bean: halo blight, caused by Pseudomonas syringae pv. phaseolicola (Psp), bacterial brown spot, caused by P. syringae pv. syringae (Pss), and common bacterial blight, caused by Xanthomonas axonopodis pv. phaseoli (Xap) and X. fuscans subsp. fuscans (Xff). Disease symptoms caused by Psp and Pss can be difficult to distinguish in certain common bean genotypes and under certain environmental conditions. Precise pathogen identification is important in terms of potential yield loss and the need for disease management. Symptoms with characteristics of both halo blight and bacterial brown spot were observed on a newly developed cultivar of dark red kidney beans in Wisconsin, and pseudomonads were recovered from all samples. To determine the etiology of this disease, a method of identification was developed that involved 1) PCR with primers specific for toxin genes utilized by Psp and Pss, 2) the ability to induce a hypersensitivity response in tobacco leaves, and 3) symptoms induced on common bean. When applied to the unusual disease phenotype on this dark red kidney variety most of these were atypical symptoms of halo blight. This method was then successfully applied to identify bacterial brown spot in disease samples from Oregon, and halo blight and bacterial brown spot in disease samples from Turkey. Xap and Xff strains induce identical symptoms on common bean but are genetically distinct species. Strains of Xap and Xff can be differentiated in culture by the production of a brown diffusible pigment by Xff strains. However, some Xap strains, particularly those that originate from East Africa, are more genetically related to Xff strains than to the globally predominant Xap strains, and show pathogenic variability on certain cultivars, e.g., Andean versus Middle American. To better understand the genetic diversity among Xap and Xff strains, the complete genome sequences of three globally predominant Xap strains, two East African Xap strains, three Xff strains, and one Xanthomonas spp. strain that is associated with common bean but not pathogenic were determined using long-read sequencing technology. Whole-genome alignment revealed differences in chromosome organization between Xap strains, East African Xap strains, and Xff strains; additionally, mobile element content is variable among these strains. Conserved genetic factors, e.g. conserved plasmids and type III secreted effector proteins, some of which may have been horizontally acquired by Xap and Xff strains, were identified. The brown diffusible pigment produced by Xff strains is the result of an oxidation product of homogentisic acid, which is referred to as pyomelanin. In the tyrosine catabolism pathway, homogentisic acid is produced by 4-hydroxyphenylpyruvate dioxygenase (HPPD) and converted to 4-maleylacetoacetate by homogentisate dioxygenase (HmgA). The complete genome sequences of Xap and Xff strains revealed that Xff strains have a conserved mutation in the hmgA gene, whereas Xap strains have an intact hmgA gene. HPPD deletion mutants in an Xff strain resulted in loss of pyomelanin (brown pigment) production, whereas deletion of hmgA in two Xap strains resulted in brown pigment production, making Xap colonies resemble those of wild-type Xff strains. Additionally, complementing a wild-type Xff strain with a functional hmgA gene eliminated visible pyomelanin accumulation. Finally, virulence of a Xff HPPD and a Xap hmgA deletion mutant was assessed on twelve bean cultivars, including both Andean and Middle American gene pools, with different levels of disease resistance to common bacterial blight. There was no significant differences in virulence between brown pigment mutants and wild-type strains.
