Over the last decade this group has, in close collaboration with the departments of neurosurgery and urology of the Academic Hospital of Leiden, studied development and regeneration of the spinal cord and its peripheral nerves both from a neuroanatomical and a clinical perspective. During this period the development of brainstem projections to the spinal cord of the rat was studied with a retrograde tracing technique. Horseradish peroxidase was injected into the spinal cord of rat fetuses, both at different ages, and at different levels of the spinal cord. These studies aimed to discover regularities in the behavior of descending fiber systems that could yield insight into the logic that the nervous system must employ to structure its connectional pattern during development. Such insight might then be applied to improve regeneration of the nervous system.

The studies on morphoregulators, in particular, appeared to benefit two different scientific areas. Firstly, the correlation between morphoregulator expression patterns and known anatomy contributed to our knowledge about spinal cord development. Secondly, the correlation between morpho regulator expression patterns and known developmental processes may help to understand their precise function(s). This volume of Advances in Anatomy, Embryology and Cell Biology presents these particular studies on the development of the rat spinal cord performed over the last decade. The spinal cord was singled out as a favorable site when cytological techniques were first applied to the study of the embryonic development of the nervous system. Bidder and Kupffer (1857), using the new procedure of hardening nerve tissue with chromic acid (Hannover 1844), made an investigation of spinal cord development in fetal sheep. They reported that the cellular central mass of the spinal cord develops before its fibrous envelope, deducing from this that the fibers of the white matter of the embryonic spinal cord were outgrowths of cells in the gray matter. Bidder and Kupffer also noted that in the spinal ganglia fibers grew out from cells in both directions, peripherally and centrally. Their report was one of the earliest ontogenetic lines of evidence in support of the later-formulated neuron doctrine (Waldeyer 1891). The spinal cord re mained a favorite topic of morphogenetic studies of the nervous system through out the last quarter of the nineteenth century, with seminal contributions made by His (1886, 1889), von Lenhossek (1889), Retzius (1898), and Ramon y Cajal (1960). Indeed, the preoccupation with the spinal cord in the early investigations of neural development had a lasting, and to some extent regrettable, influence on ideas about the ontogeny of the brain and on the terminology adopted by anatomists.

The rat brain is analyzed through stereotaxic localization of discrete brain areas and the subdivisions of many areas of rat brain are mapped using plates and diagrams. Photographs of sufficient magnification are included to permit investigators to judge for themselves the veracity of the atlas delineations. This atlas is based on the study of 130 adult male Wistar rats (with a weight range of 270-310 g). It is suitable for brains of 250-350 g male and female rats and represents all areas of the brain and spinal cord. Brain areas are shown in coronal, sagittal, and horizontal planes. The brain sections shown were taken on average at 0.25 mm intervals and were stained with either cresyl violet or for the demonstration of acetylcholinesterase (AChE). The atlas is based on fresh brains frozen in the skull (using deeply anesthetized rats) in order to overcome distortion produced by fixation and to enhance staining contrast. Structures are delineated on the basis of data on cytoarchitecture, chemoarchitecture, and connectivity. The procedures used for surgery, histology, and photography are also explained. This atlas is intended for use by researchers and graduate students in the neuroscience, as well as senior undergraduates interested in brain anatomy and function.
