Affecting more than 2.4 million worldwide in the second and third decades of life, it is the primary cause of non-traumatic disability in the United States. Sensory and motor neuron loss follows the demyelination of axons in the brain and spinal cord after autoreactive T lymphocytes gain access to the central nervous system and mediate an inflammatory reaction. Thus, most current therapies in multiple sclerosis seek to suppress the immune system in order to slow progression. Several cytokines have been shown to be involved in the pathogenesis of autoimmune diseases, including multiple sclerosis. We have focused on macrophage migration inhibitory factor, a ubiquitously expressed proinflammatory cytokine that has been described in a number of syndromes. Utilizing an MIF knockout model of EAE in the C57Bl/6 strain of mice, we show that MIF is required for susceptibility to EAE. MIF knockout mice are protected with a decreased incidence of disease and lower clinical scores. Additionally, we show that an inhibitor of MIF is therapeutic during ongoing disease. Administration of an MIF inhibitor reduced clinical scores. We propose several mechanisms of MIF that mediate inflammation. First, MIF inhibits the expansion of CD4+CD25+Foxp3+ regulatory T lymphocytes. This population of cells is found in higher numbers after immunization in MIF knockout and inhibitor-treated mice. CD4+CD25+Foxp+ regulatory T lymphocytes are protective during EAE through contact inhibition with autoreactive lymphocytes and the production of IL-10. Second, we propose that MIF mediates leukocyte migration into the brain and spinal cord. We show that MIF knockout mice have profoundly reduced inflammation and the administration of an inhibitor of MIF prevents new migration into the brain. Furthermore, MIF knockout recipient mice were universally protected following adoptive transfer of autoreactive lymphocytes. Finally, MIF regulates the synthesis of testosterone. MIF knockout mice have a four fold higher level of testosterone than wild type mice. Testosterone is an immunosuppressive hormone, and gonadectomy of MIF knockout mice greatly increased the incidence of EAE. We propose that MIF mediates inflammation via different mechanisms depending on the timing of disease. Inhibition of testosterone in naive mice enhances susceptibility to disease. Later changes during inflammation, including inhibition of regulatory T cell differentiation and facilitation of migration into peripheral tissues allows the progression of disease. We propose that targeting MIF during multiple sclerosis could be therapeutic through novel regulation of multiple aspects of inflammation.

This leads to clinical symptoms of decreased motor and sensory functions. Macrophage migration inhibitory factor (MIF) has been shown to be critical for the progression of experimental autoimmune encephalomyelitis (EAE), an animal model for MS. When immunized with the encephalitogenic peptide of myelin oligodendrocyte glycoprotein (MOG) 35-55, mice deficient in the pro-inflammatory cytokine MIF are protected from EAE. . We show the clinical signs of EAE can be reversed by the use of small molecule inhibitors of MIF. Yet, MIF is constitutively expressed in all cell types and important in innate immune function, thus global inhibition of MIF could potentially cause adverse side effects. Thus, we investigate the cellular source of the biologically relevant MIF for EAE. Through a series of bone marrow chimera and adoptive transfer experiments, we concluded the necessary MIF is bone marrow derived. These experiments eliminated T lymphocytes as the source of the biologically relevant MIF. Recently, MIF has been linked to B lymphocyte maturation. Although the role of B lymphocytes in MS pathology remains unclear, the recent success of Rituximab, a monoclonal antibody against human B lymphocytes, argues for a strong role of B lymphocytes in MS. The purpose of the latter part of this study is to investigate the autocrine role of MIF production by B lymphocytes in EAE as a future specific therapeutic target. We observed significantly fewer mature B lymphocytes in the immune compartments of MIF deficient mice when compared to wild type counterparts. Yet, significantly more progenitor B lymphocytes were observed in an MIF deficient mice, leading to the hypothesis that MIF may be crucial for B lymphocyte survival. We also observed impaired proliferation of B lymphocytes in the bone marrow of a MIF knockout mouse. Next, we performed a series of transfer experiments where the only source of MIF is B lymphocytes to determine if this source alone could induce EAE; therefore providing a specific cell type to target MIF production in EAE. We observed protection of MIF deficient recipients regardless of MIF production by the B lymphocytes. Our data suggests that although MIF is important for B cell survival, it does not act in an autocrine manner.

This stain targeted intermediate filaments that we now know consist mainly of glial fibrillary acidic protein, a protein used today as an astrocytic marker. Cajal described the morphological diversity of these cells with some ast- cytes surrounding neurons, while the others are intimately associated with vasculature. We start the book by discussing the heterogeneity of astrocytes using contemporary tools and by calling into question the assumption by classical neuroscience that neurons and glia are derived from distinct pools of progenitor cells. Astrocytes have long been neglected as active participants in intercellular communication and information processing in the central nervous system, in part due to their lack of electrical excitability. The follow up chapters review the “nuts and bolts” of ast- cytic physiology; astrocytes possess a diverse assortment of ion channels, neu- transmitter receptors, and transport mechanisms that enable the astrocytes to respond to many of the same signals that act on neurons. Since astrocytes can detect chemical transmitters that are released from neurons and can release their own extracellular signals there is an increasing awareness that they play physiological roles in regulating neuronal activity and synaptic transmission. In addition to these physiological roles, it is becoming increasingly recognized that astrocytes play critical roles during pathophysiological states of the nervous system; these states include gliomas, Alexander disease, and epilepsy to mention a few.
