Book excerpt: The Macrophage in Neoplasia is a compilation of papers presented at a workshop held at the Marine Biological Laboratory, Woods Hole, Massachusetts, on October 8-11, 1975. The book presents the many faceted activities of macrophage. This book is divided according to the five sessions of the workshop. First session talks about the role of tumor macrophages in vivo. It then elucidates the macrophage function and indicates how the interaction of macrophages with other cells can alter the host-tumor balance. The remaining sessions, as presented in this book, explore the mechanism of macrophage mediated cytotoxicity, functional expression of macrophages and neoplasia, and the stimulation of macrophage function and applied therapy.

Book excerpt: Fresh living Bacillus Calmette-Guerin (BeG), injected i.v. into (C57BI/ 6xDBA/2)FI mice, activated peritoneal macrophages rendering them highly cytotoxic for tumor cells in vitro. This cytotoxic activity w~s already maximal 14 days after injection of 1 mg of BCG and remained stable when 3 or 5 mg of BCG were given. At the same time spleen cells of the BCG-treat~d mice showed strongly depressed responses to the T-cell mitogens,PHA and Con A, irrespective of the dose of BCG injected. The inhibitory effect was shown to be mediated by suppressor cells which had characteristics of macrophages since they could be removed by carbonyl iron and magnet treatment and were adherent to plastic. In contrast to it was observed after injec tion of 1 mg of BCG, these suppressor cells alone did not account for the depression of T-cell re.sponses induced by higher doses of BCG. Nylon-nonadherent cell populations obtained from spleen cells treated with 3 or 5 mg BCG partially retained the inhibitory activity suggesting that suppressor T cells were also induced after injection of high doses of BCG. In contrast, the responses to the B-cell mitogen LPS of unfractionated and macrophage-depleted spleen cells were not affected or significantly enhanced depending on the dose of BCG injected.

Available in PDF, EPUB and Kindle. Book excerpt: Macrophages are the sentinels of the immune system whose role has evolved beyond providing aseptic conditions to homeostasis, immune regulation, development, and behaviour. These cells have varied ontogenetic origins which reflects in their phenotypic and functional heterogeneity. Macrophage functions are fine-tuned by exogenous and endogenous signals and once tweaked, the information is included in their genetic makeup, albeit not indefinitely. Subversion of the macrophage functions is the hallmark of many pathogenic organisms and modulation of macrophage activity is pivotal to many therapeutic strategies. Fascinating and rapid developments in this field have necessitated the maintenance of currency of knowledge. This book provides a current account of information on varied topics in macrophage biology. Literature surveys have been presented in a captivating and lucid language. The contributing authors have also provided brief accounts of their own research. Every chapter provides a future perspective of what more could be achieved in the context of the current knowledge. The book will be of interest to students and researchers in microbiology, immunobiology, translational research, pathology, and related fields.

Book excerpt: Provides a review of current knowledge of macrophage biology relating to cancer. Each separately authored chapter highlights a particular aspect of macrophage biology and describes the current studies of each contributor. Chapters describe the nature and possible origin of the phenotypic heterogeneity of macrophages, the mechanisms of macrophage effector function as well as the consequent tumor cell reaction to these activities. The tumor cell-macrophage interaction is a complex process influenced by many factors. This monograph will make a measurable contribution to our grip on these phenomena. Book excerpt: Immunobiology of the Macrophage presents an account of the state of knowledge of the immunobiology of the macrophage. The book's contributors-immunologists of diverse scientific and geographic backgrounds-have been encouraged to give personal accounts of developments in their special fields of interest as well as critical surveys of the backgrounds leading to these developments. The book begins with a study on the functions of macrophages in the initiation and regulation of antibody responses in vitro. This is followed by separate chapters on topics such as the role of macrophages in making an ...

Available in PDF, EPUB and Kindle. Book excerpt: This volume – for pharmacologists, systems biologists, philosophers and historians of medicine – points to investigate new avenues in pharmacology research, by providing a full assessment of the premises underlying a radical shift in the pharmacology paradigm. The pharmaceutical industry is currently facing unparalleled challenges in developing innovative drugs. While drug-developing scientists in the 1990s mostly welcomed the transformation into a target-based approach, two decades of experience shows that this model is failing to boost both drug discovery and efficiency. Selected targets were often not druggable and with poor disease linkage, leading to either high toxicity or poor efficacy. Therefore, a profound rethinking of the current paradigm is needed. Advances in systems biology are revealing a phenotypic robustness and a network structure that strongly suggest that exquisitely selective compounds, compared with multitarget drugs, may exhibit lower than desired clinical efficacy. This appreciation of the role of polypharmacology has significant implications for tackling the two major sources of attrition in drug development, efficacy and toxicity. Integrating network biology and polypharmacology holds the promise of expanding the current opportunity space for druggable targets.

Book excerpt: Chemokines and Cancer synthesizes a state-of-the-art understanding of the role that chemokines and their receptors play in the pathophysiology of malignancy. It examines the influence of chemokines on a broad array of malignant cells, including their effects on such intrinsic properties as growth and movement, as well as exploring their influence on the host's response to a growing tumor. The authors also demonstrate the physiological consequences of chemokine expression and suggest how chemokines can be used to regulate tumor growth in vivo, including their direct effects on tumor growth, on tumor destruction by host inflammatory cells, and on tumor angiogenesis. The only book available that relates chemokines to cancer, Chemokines and Cancer holds out the promise of novel therapeutic approaches to malignancy through the manipulation of chemokines and/or their receptors.
