Beijerinck in Delft and Friedrich Loeffler on Riems Island discovered a new class of infectious agents in plants and animals, a new discipline was born. This book, a compilation of papers written by well-recognized scientists, gives an impression of the early days, the pioneer period and the current state of virology. Recent developments and future perspectives of this discipline are sketched against a historic background. With contributions by A. Alcami, D. Baulcombe, F. Brown, L. W. Enquist, H. Feldmann, A. This group of viruses includes human simplex type 1 and 2, Epstein–Barr virus, Kaposi's Sarcoma-associated herpesvirus, cytomegalovirus, HHV6A, 6B and 7, and varicella-zoster virus. The viral diseases and cancers they cause are significant and often recurrent. Their prevalence in the developed world accounts for a major burden of disease, and as a result there is a great deal of research into the pathophysiology of infection and immunobiology. Another important area covered within this volume concerns antiviral therapy and the development of vaccines. All these aspects are covered in depth, both scientifically and in terms of clinical guidelines for patient care. The text is illustrated generously throughout and is fully referenced to the latest research and developments.

Available in PDF, EPUB and Kindle. Book excerpt: This book will contain a series of solicited chapters that concern with the molecular machines required by viruses to perform various essential functions of virus life cycle. The first three chapters (Introduction, Molecular Machines and Virus Architecture) introduce the reader to the best known molecular machines and to the structure of viruses. The remainder of the book will examine in detail various stages of the viral life cycle. Beginning with the viral entry into a host cell, the book takes the reader through replication of the genome, synthesis and assembly of viral structural components, genome packaging and maturation into an infectious virion. Each chapter will describe the components of the respective machine in molecular or atomic detail, genetic and biochemical analyses, and mechanism. Topics are carefully selected so that the reader is exposed to systems where there is a substantial infusion of new knowledge in recent years, which greatly elevated the fundamental mechanistic understanding of the respective molecular machine. The authors will be encouraged to simplify the detailed knowledge to basic concepts, include provocative new ideas, as well as design colorful graphics, thus making the cutting-edge information accessible to broad audience.

Available in PDF, EPUB and Kindle. Book excerpt: Immunoregulation is one of the areas which has witnessed the most explosive advances of immunology during the past decade. It is in this area that the current view of the immune system has arisen and developed. There is indeed little doubt that immune reactions are primarily determined by messages which are genera ted within the immune system and passed among different types of immunologie cells. This cell communication not only determines the type, intensity and duration of the response after perturbation of the immune system by exogenous antigens, but it is also essential for preventing autoimmune reactions and their clinical conse quences. In order to assure aperfect balance within the enormous com plexity of the immune system, it is not surprising that multiple self-regulatory mechanisms are organized at different levels, such as antibody feedback, idiotypic-anti-idiotypic responses, suppres sor and helper T cells, lymphokine signals and genetic require ments. A nu mb er of observations in recent years have, however, demonstrated that consistent contributions to the immunological homeostasis are given also by signals generated outside of the immune system, namely,in the central and autonomous nervous system as weIl as in the endocrine apparatus. Furthermore, the interactions between the immune system and the other body homestatic mechanisms seem to be bidirectional: if immunological cells may be targets of neuroendocrinological factors, immunological products seem in turn to contribute to the neuro endocrine homeostasis.

Available in PDF, EPUB and Kindle. Book excerpt: The endothelium enables communication between blood and tissues and is actively involved in cardiovascular homeostasis. Endothelial dysfunction has been recognized as an early step in the development of cardiovascular diseases: respectively, endothelium represents a potential therapeutic niche with multiple targets. The purpose of the book is to point out some recent findings of endothelial physiology and pathophysiology emphasizing various aspects of endothelial dysfunction connected to the body's internal and external environment. While basic features of the endothelium are presented in an introductory chapter, the authors of the following 17 chapters have provided extensive insight into some selected topics of endothelial (dys)function. The book would hopefully be useful for anyone interested in recapitulating endothelial (patho)physiology and expanding knowledge of molecular mechanisms involved in endothelial dysfunction, relevant also for further clinical investigations.

Book excerpt: Although respiratory syncytial virus (RSV) has been a high priority for vaccine development for over 50 years now, still no vaccine is available and none has yet demonstrated sufficient promise to move to licensure. The success of RSV immune prophylaxis and the availability of ever more powerful tools to study the immune response and pathogenesis of disease, combined with the ability to construct a wide variety of vaccines using different vaccine platforms, give us grounds to believe that an RSV vaccine is within reach. This book brings together in one source what is currently known about the virus: its clinical and epidemiologic features; the host response and pathogenesis of the disease; vaccines, vaccine platforms, and treatment; and animal and tissue culture models of RSV infection. It is designed to organize the critical information relevant to RSV vaccine development, facilitate the assimilation of data, and speed progress toward producing a safe and effective vaccine.

Book excerpt: The technique of microinjection along with viral genetics and molecular biology has proven useful in the correlation of retroviral polynucleotide structure with function. The advantage of this technique is the involvement of living cells where rare activities may be observed and where properties of living cells can be assayed. Future studies involving recombinant DNA molecules and the asso ciation of proteins with nucleic acids promise to yield additional insight into the nucleotide sequences involved in the expression of viral activities.
