The availability of genetic linkage maps, constructed by positioning molecular markers throughout the apple genome, enables the detection and analysis of major genes and quantitative trait loci (QTLs) contributing to the quality traits of a given genotype. herefore, the primary aim of this study was to construct a genetic linkage map of the Golden Delicious x Dietrich population for the identification of QTLs associated with fruit quality traits and then to examine the apple fruit pulp proteome with a specific focus on fruit firmness. In this regard, genomic DNA was extracted from leaves of the Golden Delicious x Dietrich population and used in megaplex PCR reactions. The PCR products were analysed prior to scoring of alleles. The theme of the meeting was ‘Breeding strategies for sustainable forage and turf grass improvement’. Grasslands cover a significant proportion of the land mass of the world, and play a pivotal role in global food production. At the same time we are faced with several challenges that affect the way in which we think about this valuable set of resources. The population of the world is expected to exceed 9 billion by 2050, and increase of about one third relative to today’s levels. This population increase will be focused in urban areas, and in what are currently viewed as “developing” countries, meaning that the buying power of this increased population will be greater – shifting the balance of demand from staple crops to high value items such as meat and dairy products. Overall that the world will have to approximately double agricultural output across all categories of food to meet the demands of this larger, urbanised population. This is occurring against a backdrop of equally large challenges in terms of global climate change. Agriculture is already a significant contributor to e.g. greenhouse gas emissions, deforestation and soil erosion. The situation is made more complex by an increased emphasis on biofuels as a solution for our imminent oil shortage, resulting in increased competition between land utilised for food and fuel. In short, agriculture must continue to feed the world, whilst not contributing to damaging it further. It must be sustainable. Plant breeding plays a significant but frequently understated role in meeting the challenges presented by this complex and changing scenario. However, plant breeding and improvement is itself undergoing radical change driven by technology. This book explores how forage and turf breeding is changing and adapting to meet these challenges using the technological advances being experienced in plant breeding as a whole.

However, this process has led to an inevitable narrowing of the cultivated plant's germplasm, resulting in the domesticated species only possessing a subset of the genetic diversity of its progenitor species. The vulnerability of the plant that results from the decrease in genetic diversity has been a topic of concern over the last two centuries and wild germplasm has emerged as a reservoir of alleles that may improve the agricultural traits of a plant in productivity, quality and resistance to disease and pest. The domesticated apple, Malus x domestica, is one of the most important fruits in the temperate region and are widely consumed around the world because of their high nutrition and year-round availability. However, domestication and vegetative propagation of the domesticated apple has also resulted in a decrease in genetic diversity due to only a small number of elite cultivars being used for breeding purposes. Various studies have suggested Malus sieversii, the wild apple species found in Central Asia to be the progenitor species of the domesticated apple. Investigating this wild germplasm has already identified many important traits such as resistance to diseases, pests as well as biotic and abiotic stress. With the recent developments in molecular genetic techniques such as RNA-sequencing and the completed genome sequencing of the domesticated apple cultivar, 'Golden Delicious', a detailed study comparing the genetic diversity between wild and domesticated apple seemed necessary. A comparative transcriptomic and non-targeted metabolomic study was undertaken on M. domestica, M. sieversii and M. sylvestris, the European wild crabapple. The metabolomic profiles of these three species showed how each species are characterized by a different group of metabolites. Also, ascorbyl glucoside, a stable form of vitamin C, was isolated for the first time in apple to the best of my knowledge and was found to be present in high concentrations in two M. sylvestris genotypes. Various possibilities of ascorbyl glucoside synthesis in apple are explored.
