Book excerpt: Higher eukaryotes are characterized by the allocation of distinct functions to numerous types of differentiated cells. Whereas in animals the well-defined, protected cells of the germ line separate early, germ cells in plants differentiate from somatic cells only after many cycles of mitotic division. Therefore somatic mutations in plants can be transmitted via the germ cells to the progeny. There is thus a clear need for somatic tissues to maintain their genetic integrity in the face of environmental challenges, and two types of interactions have been shown to play important roles in the conservation as well as flexibility of plant genomes: homologous recombination of repeated sequences and silencing of multiplied genes. Sensitive methods have been developed that allow greater insights into the dynamics of the genome. This book summarizes current knowledge and working hypotheses about the frequencies and mechanisms of mitochondrial, plastid, nuclear and viral recombination and the inactivation of repeated genes in plants. Despite rapid developments in the field, it is often not possible to provide final answers. Thus, it is an additional task of this book to define the open questions and future challenges. The book is addressed to scientists working on plant biology and recombination, to newcomers in the field and to advanced biology students.

Thus, one should consider two possible situations, which are by no means mutually exclusive, to account for immunoƯ logic unresponsiveness: (a) one or more of the cell populations composing the synergistic unit is absent or immature, and (b) an antagonistic unit which interferes with the response is dominating. In view of this, an approach to development of immune reactivity necessitates parallel surveys of development of cells with the potential to perform, as well as of cells which can suppress the response. Classification of the various cell types has been based so far on their phenotypic properties (e. g., membrane antigen markers, cell receptors, proƯ duction and secretion of immunoglobulins, etc.). Genotypically, T and B cells may represent either separate, independent cell lines, or different stages of development within the same cell lineage.

It was long felt that the ability to generate mutants and conduct genetic studies in mammalian systems would prove to be equally useful. However, genetic analysis based on sexual systems is difficult in mammals because of the long generation times and the inability to perform controlled matings. As a result, genetic analysis of mam malian systems had to await the development of parasexual systems. This book is an attempt to bring together descriptions of a number of these parasexual systems. A common theme of all the parasexual systems is the transfer of genetic information from a defined source into a specific cell type. This volume deals with a number of methods of gene transfer into mammalian cells. The early methods of gene transfer involved transfer of relatively large amounts of genetic information. These include somatic cell hybridization, microcell fusion, and chromosome transfer, which constitute the first part of this book. Each of these methods has already proven to be of enormous value in arriving at a genetic understanding of the mammalian genome. Development of recombinant DNA methods, and the ability to introduce purified DNA into mammalian cells, has had a significant impact on our ability to dissect important aspects of mammalian gene expression and regulation. The second part of this book deals with gene transfer systems involving defined nucleic acid sequences.
