Book excerpt: "Oxidative stress is caused by an imbalance between the production and removal of reactive oxygen species (ROS). Polyunsaturated fatty acids (PUFA), due to their multiple carbon-carbon double bonds, react readily with ROS in a process termed lipid peroxidation. Lipid peroxidation generates a number of potentially harmful secondary products, including the aldehydes ethanal, propanal, and hexanal. Indeed, raised aldehyde levels have been associated with various diseases, including cancer, leading researchers to consider them as potential diagnostic markers. However the abundance of their fatty acid precursors is dependent on dietary intake. As such, tissue aldehyde content may be diet-dependent, reducing their desirability as markers. To investigate this I fed 32 male Wistar rats diets containing 90% fat-free rat chow, 9% palm oil (mostly saturated fat), and 1% omega-3 fatty acid (EPA, DHA, ALA) for 8 weeks. The different diets resulted in changed fatty acid lipid composition in rat brain and liver. Compared to controls, DHA and EPA diets decreased liver arachidonic acid levels by 10%, while increasing levels of EPA and DHA by 7-11%. A similar effect was seen in brain lipid composition, although the changes, while statistically significant, weren't as pronounced. In brain lipids, feeding omega-3 PUFA did not lead to great changes in their concentrations, while DHA was found to be maintained at high levels even in the absence of any omega-3 PUFA in the diet. However, selected ion flow mass-spectrometry (SIFT-MS) analysis of the rat livers and brains suggests that diet does not significantly affect the concentrations of various aldehydes (P>0.05), or overall levels of oxidative stress (as measured by TBARS assays) in liver or brain (P>0.05). My results suggest that volatile aldehydes may represent a useful marker for oxidative stress with potential applications in a variety of common disorders."--Abstract.

However, non-surgical weight-loss treatments are ineffective. Treatments that can reduce cardiometabolic risk in the overweight and obese are urgently needed. Further, as the offspring of obese and/or diabetic women are programmed to become obese and insulin resistant with age, interventions in pregnancy could have substantial long-term impacts on public health. Supplemental omega-3 fatty acids appear promising as they modulate important metabolic pathways and are insulin-sensitising in rats. The central hypothesis in this thesis is that long chain omega-3 polyunsaturated fatty acids, can modify the effects of gene-environment interactions to improve aspects of glucose and lipid metabolism. Four predictions were tested 1) that an n-3 PUFA supplement would be insulin sensitising in overweight men and 2) that an n-3 PUFA supplement used in a model of insulin resistant pregnancy would have metabolic benefits to the offspring 3) that fish oils sold at retail would have excessive oxidation, and 4) that an oxidised fish oil supplement would not have benefits to the offspring when used during pregnancy. I used a randomised controlled crossover trial to investigate the metabolic effects of krill/salmon blended oil on insulin sensitivity in overweight men. At baseline, higher n-3 PUFA levels were associated with greater insulin sensitivity. However, supplementation with the marine oil led to reduced insulin sensitivity, suggesting increased cardiometabolic risk. In an animal model of insulin-resistant pregnancy (maternal high-fat diet supplementation with unoxidized fish oil prevented the development of insulin resistance in the adult offspring. I found that fish oil products purchased at retail were substantially oxidised and did not meet labelled n-3 PUFA content. I then assessed the effects of oxidised fish oil supplementation during rat pregnancy, and found a substantial increase in neonatal mortality and persistent maternal insulin resistance. I have shown that oxidation changes the metabolic effects of n-3 PUFA rich supplements. It is possible that there are negative consequences to consuming oxidised fish oil during human pregnancy, and unrecognised oxidation may have confounded the n-3 PUFA clinical trial literature. The present study examined the effect of dietary DHA or AA on hepatic fatty acid composition and triglycerides in rats. I hypothesized that DHA would increase the levels of 20:5n-3 and 22:5n-3 and suppress the levels of 20:4n-6 and 22:5n-6 and other n-6 fatty acids in both fed and fasted rats. Methods. Artificially reared rats were fed one of four experimental formulas: no AA + no DHA (control), no AA + DHA (2.5% of total fatty acids), AA (2.5% of total fatty acids) + no DHA, or both AA + DHA from postnatal days 6-18. On postnatal days 19-21 all rats were fed the same diet containing no AA + no DHA using a meal feeding regimen with overnight fasting. All of the diets contained marginal levels of linoleic acid and - linolenic acid to enhance incorporation of AA and DHA from the diet. Liver was isolated from fed and fasted rats (study 1 and 2, respectively). Results. The patterns of liver fatty acids were similar in both feeding states. The two groups fed diets containing DHA had higher levels of 20:5n-3 and 22:5n-3 and lower levels of 20:4n-6, 22:4n-6, and 22-5n-6 than the control. The AA (no DHA) group had higher levels of 22:4n-6 and 22:5n-6 than the control and the DHA (no AA) groups. The levels of 22:4n-6 and 22:5n-6 were lower when rats were fed diets with DI-JA.
