Book excerpt: Autoradiography in Biology and Medicine focuses on the applications of autoradiography in medicine and biology, including photographic processes, emulsions, and response of films to isotopes and x-rays. The book first offers information on the meaning of autoradiography, including the etymology of autoradiography, photographic emulsion as a scientific instrument, and the relationship of biologists and autoradiography. The publication then examines the photographic process and comparative response of commercial and scientific emulsions. Discussions focus on fixation, washing, emulsion, response of films to electrons, x-rays, and gamma rays, and response of films to beta ray spectra of isotopes. The text takes a look at commercial photographic materials for autoradiography, autoradiographic image, and resolution. The manuscript then reviews the estimation of dose and exposure time, sources of error, and techniques. Topics include chemical variables, estimation of exposure time, errors occurring during the preparation of the autoradiogram for exposure, and contour autoradiography. The book is a valuable reference for readers interested in autoradiography.

Available in PDF, EPUB and Kindle. Book excerpt: This volume was originally intended to be an English translation of the book MetllOden in der medizinischen Cytogenetik, published in 1970. Just about then, however, a number of new techniques were introduced in human cytogenetics and soon acquired the utmost importance, parti cularly in clinical diagnosis, so that the English edition had to be con siderably enlarged. As a result, there are now twelve chapters instead of eight, and two additional authors have been called upon, Dr. KRONE and Dr. SCHNEDL. In addition to the up-to-date presentation of con ventional methods of cell culture and techniques for the preparation and identification of human chromosomes, this text covers the various tech niques of producing banding patterns and applying them in chromo some identification. Further, it deals with the culture of amniotic fluid cells and gives instructions for handling tissue-culture cells for bio chemical analysis; it thus meets the ever-increasing requirements of a modern cell-culture laboratory. To paraphrase the aims of this book, we quote part of the preface to the German edition: "It was intended to collect the various methods so as to make them accessible for laboratory use. Furthermore, it is hoped that the reader faced with current research problems will be stimulated to modify and supplement the techniques described, instead of merely applying them automatically. In a rapidly developing field, some methods are still preliminary, and no final presentation seems possible.

Book excerpt: In many aspects hematopoiesis in newborn rodents, especially in rats, resembles hema topoiesis in the human fetus in the 6th-7th month of gestation. In man the transition from the stage of liver to bone marrow erythropoiesis takes place at this time (Bessis, 1973). In rodents, however, the liver is almost the only place where hematopoiesis occurs until birth. Thereafter it is replaced to a growing extent by the bone marrow, which so far consists mainly of immature mesenchymal cells (Maximow, 1910; Cuda, 1970). Thus hematopoietic precursor cells appear in the sternum only around 30 h after birth. Just as in premature human infants, a macrocytic anemia can be demonstrat ed in normal neonatal rats (Lucarelli et aI., 1964, 1968). Beside liver (fetal) and bone marrow, the spleen is involved in hematopoiesis. In rodents like rats and mice, splenic hematopoiesis persists more or less markedly until adulthood; in man, however, it ceases after birth and reappears only under certain pathological conditions (Fischer et aI., 1970; Hennekeuser et aI., 1967; Fresen, 1960).

Book excerpt: This text is a summary of basic principles and techniques and is dedicated to all those students who have been told by their mentors, "Go forth and do two-dimensional gels and have the results on my desk tomorrow. " No attempt has been made in this text to provide exhaustive lists of references related to basic principles or techniques or to list every com pany or supplier involved in this area of research. Nevertheless, it is hoped that sufficient information is given to help a new investigator or student appreciate the complexities but develop sufficient expertise to carry out these techniques successfully. The discussions are designed to instill in basic science and clinical investigators of all levels of expertise an appreciation of the power of combining a variety of techniques as well as to provide basic insight into the theories, complexities, and problems frequently encountered with electrophoretic and immunochemical meth ods. Bonnie S. Dunbar Houston v Acknowledgments I wish to thank my students and staff for their patience and support throughout the preparation of this text. I would like to acknowledge my appreciation for my extensive discussions with Dr. David Sammons (Uni versity of Arizona) and to Dr. N. L. and Dr. N. G. Book excerpt: The past decade has seen a remarkable increase in the use of electron microscopy as a researm tool in biology and medicine. Thus, most institu tions of higher learning now boast several electron optical laboratories having various levels of sophistication. Training in the routine use of elec tron optical equipment and interpretation of results is no longer restricted to a few prestigious centers. On the other hand, temniques utilized by researm workers in the ultrastructural domain have become extremely diverse and complex. Although a large number of quite excellent volumes of electron microscopic temnique are now dedicated to the basic elements available whim allow the novice to acquire a reasonable introduction to the field, relatively few books have been devoted to a discussion of more ad vanced temnical aspects of the art. It was with this view that the present volume was conceived as a handy reference for workers already having some background in the field, as an information source for those wishing to shift efforts into more promising temniques, or for use as an advanced course or seminar guide. Subject matter has been mosen particularly on the basis of pertinence to present researm activities in biological electron microscopy and emphasis has been given those areas whim seem destined to greatly expand in useful ness in the near future.

Available in PDF, EPUB and Kindle. Book excerpt: Mark Chase There are many literature resources available to molecular biologists wishing to assess genetic variation, but the myriad of techniques and approaches potentially available to the plant breeder and the evolutionary biologist is truly bewildering, and most have never been evaluated side-by-side on the same sets of samples. Additionally, it is often not recognized that tools that are useful for breeders can often be adapted for use in evolutionary studies and vice versa, but this is generally the case. The borderline between population genetics and phylogenetics is vague and difficult to assess, and a combination of both types of tools is best when it is not clear with which area one is dealing. Furthermore, it is not now appropriate to use just one type of marker in any kind of study; most markers have the potential to misinform under certain conditions, so it is always wise to incorporate at least two different types of assessments into any project. This volume is designed to facilitate this sort of multiple approach and provides comparative data on most currently available methods so that researchers can more intelligently select those appropriate to their area of interest, regardless of whether it is in the realm of breeding or evolutionary biology.
