Book excerpt: Insects as a group occupy a middle ground in the biosphere between bacteria and viruses at one extreme, amphibians and mammals at the other. The size and general nature of insects present special problems to the study of ento mology. For example, many commercially available instruments are geared to measure in grams, while the forces commonly encountered in studying insects are in the milligram range. Therefore, techniques developed in the study of insects or in those fields concerned with the control of insect pests are often unique. Methods for measuring things are common to all sciences. Advances some times depend more on how something was done than on what was measured; indeed a given field often progresses from one technique to another as new methods are discovered, developed, and modified. Just as often, some of these techniques find their way into the classroom when the problems involved have been sufficiently ironed out to permit students to master the manipulations in a few laboratory periods. Many specialized techniques are confined to one specific research labora tory. Although methods may be considered commonplace where they are used, in another context even the simplest procedures may save considerable time. It is the purpose of this series (1) to report new developments in method ology, (2) to reveal sources of groups who have dealt with and solved particular entomological problems, and (3) to describe experiments which may be appli cable for use in biology laboratory courses.

It only scratches the surface of the exploding repertoire of general neuro chemical techniques that can be applied to insect research in 1985. But it al so presents the advantages of using insects for studying certain biological questions that are approachable by neurochemical techniques. Even more so, it summarizes the long list of unique problems encountered in attempting to study insects by neurochemical techniques. As in other volumes of this series, the contributors to this volume are the authorities in the field. They themselves have developed much of the material presented. Thus the sum effort provides a true description of the state of the art; and, pleasantly, it does so in a very complete and clear manner. Readers of this series will not need to be reminded that, despite the fact that vertebrates make up only about 3% of all animal species, research in in vertebrates such as insects has lagged behind that on vertebrates, at least in the neurochemistry area; the relative simplicity and large cell size of the in sect nervous system has always provided incentive for work in neurophysiol ogy and neuroanatomy. Toxicology interests will always stimulate a certain amount of work on insect neuropharmacology, and insects are extremely suitable for several areas of toxin research. Last but not least, the insects are beautiful organisms for which the applications of genetics can be made to the study of nervous system function.

Book excerpt: In insect and other arthropod immune systems, discrimination between self and nonself tissues is accomplished through the combined actions of two immunocytes and several humoral factors. Immunology of Insects and Other Arthropods presents a comprehensive look at this and other important topics in arthropod immunology. Issues discussed include insect immunocytes and other hemocytes, including computer image analysis of immunocyte serial sections; the two basic cellular immune reactions (phagocytosis and encapsulation), including the molecular basis and roles of gap junctions in encapsulation; how encapsulation is affected by polydnavirus and encapsulation-promoting factors; why insect cells are immune to HIV; humoral factors; and antibacterial factors in Lepidoptera, Diptera, and other insect orders. Other topics include hemolymph proteins interacting with mammalian complement cascade; adaptive humoral response in the American cockroach; antigenic stimulation of hemaglutinin production in insects; and the applications of the Limulus Amebocyte Lysate (LAL) in detecting endotoxins in pharmaceuticals, medical devices, clinical diagnosis, and hygienic control. This book represents an important reference source for hematologists, pathologists, immunologists, AIDS researchers, comparative immunologists, and pharmaceutical companies.

In Comstock Hall, the building of the Department of Entomology at Cornell University where I was first introduced to experimental science in the laboratory of Tom Eisner, those of us interested in the chemistry of life felt like interlopers. In the 35 years that have elapsed since then, all of biology has changed, and entomology with it. Arrogant molecular biologists and resentful classical biologists might think that what has happened is a hostile take-over of biology by molecular biology. But they are wrong. More and more we now understand that the events were happier and much more exciting, amounting to a new synthesis. Molecular Biology, which was initially focused on the simplest of organisms, bacteria and viruses, broke out of its confines after the initial fundamental questions were answered - the structure of DNA, the genetic code, the nature of regulatory genes - and, importantly, as its methods became more and more generally applicable. The recombinant DNA revo lution of the 1970s, the development of techniques for sequencing macromolecules, the polymerase chain reaction, new molecular methods of genetic analysis, all brought molecular biology face to face with the infinite complexity and the exuber ant diversity of life. Molecular biology itself stopped being an isolated diScipline, pre occupied with the universal laws of life, and became an approach to addressing fas cinating specific problems from every field of biology.

Book excerpt: This volume surveys the chemistry, biochemistry, biosynthesis, metabolism and pharmacological properties of lectins. Lectins, which are most commonly found in plants, are widespread natural products with striking biological activities. Their specific ability to recognise and bind to simple or complex saccharides facilitates their role as effective information protein molecules. As agents of cell-to-cell recognition, lectins promote symbiosis between plants and specific nitrogen-fixing soil bacteria. As natural defensive molecules, they can protect plants against predators such as bacteria, fungi and insects. As part of our diet, lectins are powerful exogenous growth factors in the small intestine and influence our health, the digestive function and the bacterial ecology of the alimentary tract. Lectins are also important research tools in preparative biochemistry and cell science.

Book excerpt: Manual of Techniques in Invertebrate Pathology, Second Edition, describes a wide range of techniques used in the identification, isolation, propagation/cultivation, bioassay, quantification, preservation, and storage of the major groups of entomopathogens, including entomophthorales, entomopathogenic fungi, entomopathogenic bacteria of the Bacilli, Nematode parasites, and pathogens and parasites of terrestrial molluscs. The book presents the perspectives of an international group of experts in the fields of invertebrate pathology, including microbiology, mycology, virology, nematology, biological control, and integrated pest management. Organized into 15 chapters, the book covers methods for the study of virtually every major group of entomopathogen, as well as methods for discovery and diagnosis of entomopathogens and the use of complementary methods for microscopy. It discusses the use of molecular techniques for identifying and determining phylogeny, factors that contribute to resistance to entomopathogens, and several other aspects of the science of invertebrate pathology. It also explains initial handling and diagnosis of diseased invertebrates, basic techniques in insect virology, and bioassay of bacterial entomopathogens against insect larvae. In addition, the reader is introduced to the use of bacteria against soil-inhabiting insects and preservation of entomopathogenic fungal cultures. The remaining chapters focus on research methods for entomopathogenic microsporidia and other protists, how the pathogenicity and infectivity of entomopathogens to mammals are tested, and preparations of entomopathogens and diseased specimens for more detailed study using microscopy. Experienced insect pathologists, biologists, entomologists, students, biotechnology personnel, technicians, those working in the biopesticide industry, and government regulators will find this manual extremely helpful. Step-by-step instructions for the latest techniques on how to isolate, identify, culture, bioassay and store the major groups of entomopathogens New edition fully updated to address changes in the taxonomy of the vast majority of taxa Discussion of safety testing of entomopathogens in mammals and also broader methods such as microscopy and molecular techniques Provides extensive supplemental literature and recipes for media, fixatives and stains
