Book excerpt: Studies toward the total synthesis of a highly potent cytotoxic marine natural product, phorboxazole A, were conducted and resulted in a route to an advanced intermediate, C4-C32, for this purpose. A key feature of our approach is the stereoselective synthesis of two cis-2,6-disubstituted tetrahydropyrans present in the macrolide portion of phorboxazole A by palladium (II) mediated intramolecular alkoxy carbonylation. This provided the C20-C32 and C9-C19 tetrahydropyran subunits of phorboxazole A. An attempt at diastereoselective formation of the third C5-C9 trans-2,6-disubstituted tetrahydropyran by hydride reduction of a C9 hemiketal was complicated by reduction of the C7 exocyclic olefin. However, the C5-C9 tetrahydropyran was constructed by an intramolecular etherification sequence using a novel allylsilane as the source of C4-C8 of the macrolactone. The studies carried out in the course of this thesis have set in place a major segment of the phorboxazole A structure; they require only the addition of the C1-C3 unit and minor functional group modifications to complete the macrolide portion of the molecule.

Book excerpt: The total synthesis of a highly potent cytotoxic marine natural product, phorboxazole A, is described. Both a palladium(II)-mediated and a palladium(II)-catalyzed intramolecular alkoxycarbonylation were used as key steps for the preparation of the two cis-2,6-disubstituted tetrahydropyran rings (C22-C26 and C11-C15) of phorboxazole A. The synthesis used a convergent approach involving five subunits, namely to prepare C3-C8 (195), C9-C19 (149), C20-C32 (131), C33-C41 (130) and C42-C46 (129). Assembly of two subunits, 130 and 131, employed Evans coupling protocol and provided 141 in excellent yield. The coupled product 141 was converted to aldehyde 215 in a six step sequence in which Julia-Kocienski olefination using a benzothiazole sulfone 129 was used to install the triene side chain. Allylsilylation of 149 with 195 afforded 202, which was linked by Wittig olefination with 215 to furnish 216. The C5-C9 trans-2,6-disubstituted tetrahydropyran ring was constructed from 216 in a three step sequence. Further transformations from 219 led to phorboxazole A (1) as an off-white solid.

Among the most powerful cytostatic agents people have discovered, phorboxazoles have attracted organic chemistry community to engage in the campaign to open a synthetic route in replace of isolation from their natural source since the first total synthesis of phorboxazole A was reported by Forsyth and co-workers in 1998. As the synthetic art advanced in long term exploration, many analogues derived from phorboxazoles were designed, synthesized and investigated for structure-activity relationship (SAR) related with further optimized biological activities. Described herein is an enhanced synthesis of phorboxazole C31-C46 tail domain based on stereoselective vinylogous Mukaiyama aldol reaction and modified Julia olefination. Two reliable and practical routes arose from different sources of chiral skeleton were developed respectively. Several novel phorboxazole tail domain oxazole or non-oxazole analogues were prepared from the advanced ester precursor, ready for further bioassays.

Book excerpt: This thesis presents our studies towards the first total synthesis of the novel anti- HIV agent lancifodilactone G, which has a highly unusual aliphatic enol. The first chapter provides a survey of architecturally diverse nortriterpenoids that were isolated from the Schisandraceae family. A proposed biosynthetic pathway for lancifodilactone G and closely related natural products provides a rationale for the formation of the consecutive 7/8/5 fused carbo cycles that are unique to Schisandra nortriterpenoids. Chapter 1 goes on to outline the reported strategies to access the core of lancifodilactone G and concludes with a retrosynthetic analysis proposed by the Evans group, which includes a biosynthetically inspired single-pot polycyclisation reaction. Chapter 2 describes the highly stereocontrolled synthesis of the eastern fragment (F-G rings) using transition metal-mediated Pauson-Khand reaction. This chapter also reviews the metal-mediated diastereoselective Pauson-Khand reaction directed by the stereogenic centre at C2, with the ample illustration to total synthesis. Attempted strategies for the assembly of the bicyclic cyclopentanone motif via a dienyl Pauson-Khand reaction of silicon- and oxygen- tethered diene-enes are presented. The failure of these strategies at different stages of the synthesis resulted in the exploration of a classical Pauson-Khand approach, which successfully furnished the eastern fragment. Finally, a second-generation synthesis is described which provided the fully functionalised eastern fragment with improved efficiency and overall yield. Chapter 3 discusses the successful synthesis of the western fragment (B-C rings) using a diastereoselective [4+3] cycloaddition strategy. Attempted strategies for the synthesis of the key 2,3-disubstituted furan derivative are presented, which was achieved via a hetero Pauson- Khand reaction. This chapter includes a brief account of the classical [4+3] cycloaddition reactions of furans using an in situ generated oxyallyl cation and also employing vinyl carbenoids in the metal-catalysed version. The review also highlights the application of the [4+ 3] cycloaddition reaction in the expeditious assembly of functionalised 7-membered rings that occur in a number of important biologically active natural products. The third chapter goes on to describe the application of these cycloaddition reactions in the synthesis of the fully functionalised western fragment of Lancifodilactone G. Chapter 4 describes a model study aimed at expediting the synthesis of the western fragment using a rhodium-catalysed allylic substitution reaction. A brief mechanistic discussion on unique aspects of the allylic alkylation reaction is illustrated. Chapter 4 concludes by outlining the coupling strategy for eastern and western fragments and the end game studies for the completion of the synthesis of lancifodilactone G.
