Book excerpt: Doctoral Thesis / Dissertation from the year 2004 in the subject Chemistry - Organic Chemistry, Bangalore University / Central College (Department of Studies in Chemistry), language: English, abstract: The importance of ß-amino acids has been focused, particularly in the past few decades. They are found as components of many biologically active peptidic and nonpeptidic natural products with antibiotic, antifungal, cytotoxic, and other pharmacological properties. ß-Amino acids are produced in humans, animals, microorganisms, marine organisms, and plants either in free state or as part of a peptide or depsipeptide. The importance of nonpeptidic ß-amino acids has been nfocused particularly on the ß-lactam antibiotics, since naturally occurring penicillin derivatives have been discovered as broad antibiotic active agents. Over the years, a large number of these compounds have been prepared and tested, and a variety of new ß-lactam ring systems have been introduced such as cephems, cephalosporins, oxacephems, penems, carbapenems, oxapenams as well as monocyclic and polycyclic ring systems. ß-Amino acids have been known to play an important role in primary and secondary metabolism also. Book excerpt: Organic chemists working on the synthesis of natural products have long found a special challenge in the preparation of peptides and proteins. However, more reliable, more efficient synthetic preparation methods have been developed in recent years. This reference evaluates the most important synthesis methods available today, and also considers methods that show promise for future applications. This text describes the state of the art in efficient synthetic methods for the synthesis of both natural and artificial large peptide and protein molecules. Subjects include an introduction to basic topics, linear solid-phase synthesis of peptides, peptide synthesis in solution, convergent solid-phase synthesis, methods for the synthesis of branched peptides, formation of disulfide bridges, and more. The book emphasizes strategies and tactics that must be considered for the successful synthesis of peptides.

Book excerpt: This volume contains the lectures presented at the second course of the International School of Space Chemistry held in Erice (Sicily) from October 20 - 30 1991 at the "E. Majorana Centre for Scientific Culture". The course was attended by 58 participants from 13 countries. The Chemistry of Life's Origins is well recognized as one of the most critical subjects of modem chemistry. Much progress has been made since the amazingly perceptive contributions by Oparin some 70 years ago when he first outlined a possible series of steps starting from simple molecules to basic building blocks and ultimate assembly into simple organisms capable of replicating, catalysis and evolution to higher organisms. The pioneering experiments of Stanley Miller demonstrated already forty years ago how easy it could have been to form the amino acids which are critical to living organisms. However we have since learned and are still learning a great deal more about the primitive conditions on earth which has led us to a rethinking of where and how the condition for prebiotic chemical processes occurred. We have also learned a great deal more about the molecular basis for life. For instance, the existence of DNA was just discovered forty years ago.

Available in PDF, EPUB and Kindle. Book excerpt: The human genome and other large-scale genome sequencing projects have inevitably led to a focus on the proteins encoded by genes. The field of proteomics has grown enormously as a result and a number of high-throughput technologies have now been developed allowing discovery-led investigations of protein populations rather than more traditional hypothesis-led studies on single proteins. These high-throughput techno- gies include gene and protein microarrays, the yeast two-hybrid system, and various mass spectrometry methodologies. However, despite developments and improvements in these technologies, two-dimensional electrophoresis (2DE) remains one of the most widely used approaches. This technique was revolutionised by the development of immobilised pH gradient strips which are now commercially available. This has made possible highly reproducible separations of matched samples. Developments in staining, mass spectr- etry, and bioinformatics supported these developments and have led to a measure of standardisation in design, execution, and analysis of proteomics experiments. This book began life as a proposed update of the excellent volume 2DE Protocols edited by Andrew Link of the University of Washington at Seattle. However, we re- ised that 2DE has undergone major development in aspects of its technology in recent years and we were anxious to reflect these in the present volume. We are also conscious that many researchers have now begun to apply proteomics methodologies to a growing range of biological material and we were anxious to include contributions to reflect the challenges posed in sample preparation in less widely used organisms.

It provides the antibiotic activities as well as chemical reactivities of beta-lactams, which are important because they are commonly applied as antibiotics in the treatment of bacterial infections. The volume provides comprehensive coverage of most of the known beta-lactam antibiotics with both structural and biological information, antibiotic mechanisms, self-defense mechanisms of bacteria, nearly all known synthetic methods for the preparation of beta-lactams, and possible reactions in which beta-lactams can participate. Key features: Provides the most comprehensive collection of beta-lactam antibiotics (up to 269 molecules) with chemical structures, CAS number, IUPAC names and associated biological activities attached Offers a comprehensive and detailed collection of beta-lactamase databases Explains the self-defense mechanisms of bacteria for surviving, including the formation of biofilm and conversion into L-form and secretion of beta-lactamase to deactivate the beta-lactam antibiotics Provides a comprehensive survey on the synthetic methods to make beta-lactams Gives all of the possible reactions involving beta-lactams as the starting materials Surveys over 1000 research works and includes all available DOI information The volume is a valuable resource on the antibiotic activities as well as chemical reactivities of beta-lactams for researchers and scientists, faculty, and advanced students, as well as for industry professionals working in medicinal and pharmaceutical chemistry, organic chemistry, organic synthesis, heterocycles, proteins and peptides.

Available in PDF, EPUB and Kindle. Book excerpt: Almost two centuries ago proteins were recognized as the primary materials (proteios = primary) oflife, but the significance and wide role of peptides (from pepsis = digestion) in practically all life pro cesses has only become apparent in the last few decades. Biologi cally active peptides are now being discovered at rapid intervals in the brain and in other organs including the heart, in the skin of amphibians and many other tissues. Peptides and peptide-like compounds are found among toxins and antibiotics. It is unlikely that this process, an almost explosive broadening of the field, will come to a sudden halt. By now it is obvious that Nature has used the combination of a small to moderate number of amino acids to generate a great variety of agonists with specific and often highly sophisticated functions. Thus, peptide chemistry must be regarded as a discipline in its own right, a major branch of biochemistry, fairly separate from the chemistry of proteins. Because of the important role played by synthesis both in the study and in the practical preparation of peptides, their area can be considered as belonging to bio-organic chemistry as well. The already overwhelming and still increasing body of know ledge renders an account of the history of peptide chemistry more and more difficult. It appears therefore timely to look back, to take stock and to recall the important stages in the development of a new discipline.
