Book excerpt: Abstract: A series of dinitrosyl iron complexes with the formula [(L)Fe(NO)2], {L = ), 2,2'- bis(diphenylphosphino)-1,1'-binaphthyl (1), 1,1’-bis(diphenylphosphino)ferrocene (2B), and 1,4’-bis(diphenylphosphino)butane (3B)} has been prepared and characterized through a combination of FT-IR, NMR, UV-Vis, X-ray crystallography, CV and EPR techniques. IR spectroscopy showed NO shifts to the ranges of 1708 cm-1 – 1702 cm-1 and 1661 cm-1 – 1654 cm-1 for the chelated products 1, 2B and 3B. Molecular structures products 1 and 2B were confirmed by X-ray crystallography and confirmed to be chelated structures and in the “attracto” conformation. CV study of product 2B displayed two quasi-reversible processes with E1/2 values of -0.091 V and 0.451 V at the dinitrosyl iron center. The EPR spectroscopy confirmed that product 2B is EPR active with an EPR peak of a triplet of a quintet with a gfactor calculated of 2.033. calculated g-factor aligns with the signature non-heme dinitrosyl iron complexes with g-factor of “2.03”.

It is hoped that the compounds may be used for nitric oxide releasing agents in the biological system. Three novel dinitrosyl-iron complexes of bridged bis-phophine ligands were synthesized. Syntheses of the compounds were performed in the glove box under a nitrogen environment. The synthesized compound has general molecular formula of Fe2(NO)4(CO)2(L) where (L = ( - )-1,2-Bis[(2S,5S)-2,5-dimethylphospholano]ethane, ( - )-1,2-Bis((2R,5R)-2,5-diphenylphospholano)ethane, Bis(dicyclohexylphosphino)methane). These compounds were characterized using X-ray crystallography and spectroscopic methods of infrared (IR), ultraviolet-visible (UV-Vis), and nuclear magnetic resonance (NMR). FT-IR spectrum showed the formation of these complexes, UV-Vis analyses demonstrated the compounds characteristics, and the presence of compounds were detected by 1H and 13C NMR.

Infrared spectroscopy showed NO shifts to the region of 1723 and 1674 for complexes 4b and 8b, and 1708 and 1660 for 5b, indicating that ligand 5 acts as stronger [Sigma]-donor. The X-ray crystallographic data showed that 4b and 8b possess the rare repulso conformation while 5b has the attracto conformation. CV studies on compounds 4b, 5b and 8b display two quasi-reversible oxidations with the E[subscript 1/2] values at 0.101 and 0.186 V, 0.121 and 0.184, and 0.019 and 0.342V, respectively. The larger [Delta]E value for compound 5b compared with that of 4b and 8b is attributed to the lack of [Pi]-bonds between the two phosphorus atoms. Complexes 4b, 5b and 8b exhibit weak, yet broad absorbance bands, assigned to d-d transitions in the visible region from 475 to 600 nm with molar absorptivity coefficients of 184.6, 166.4 and 241.4 M−1 cm−1, respectively.

Book excerpt: Abstract: Three new [Fe-NO]1U+007b complexes coordinated with nitrogen chelating ligands have been synthesized and characterized. The new synthesized complexes are dinitrosyl(2-2'-biquinoline)iron(O), dinitrosyl(2,2' -bipyridine 4,4' -dimethyl)iron(0), and dinitrosy 1(2,2'-bipyridine 4,4' -diphenyl)iron(0). Each complex is synthesized utilizing a common starting material, dinitrosyldicarbonyliron(0), (Fe(NO)2(CO)2). Reaction conditions are in an inert atmosphere at ambient temperature and pressure. The characterizations of these molecules are done with ultraviolet visible spectroscopy, infrared spectroscopy, and single crystal X-ray diffraction. Ultraviolet visible spectroscopy display metal ligand charge transfers between 250-550 nm. Peaks below this are due to intraligand transitions from the chelating ligands. Infrared spectroscopy data reveal that NO stretching frequencies shift to lower wavenumbers when compared to Fe(NO)2(CO)2 due to its better U+0076-donating abilities of the nitrogen chelating ligands. Single crystal X-ray diffraction data show that all complexes are pseudo tetrahedral in an attracto conformation.

Book excerpt: Dinitrosyl iron complexes (DNICs) are organometallic-like compounds formed endogenously as products of degradation of iron-sulfur clusters by NO or its interaction with the cellular chelatable iron pool. Substantial evidence has accrued on the role of DNICs as endogenous transport and storage agents for the highly reactive NO radical. Such biological significance has led to biomimetic work on such entities in synthetic inorganic chemistry. Recently, such synthetic models have emerged as a new class of potential NO releasing agents. A series of N-heterocyclic carbene (NHC)-stabilized oxidized and reduced DNICs ({Fe(NO)2}^9 /{Fe(NO)2}^10: Enemark-Feltham notation) appeared promising candidates for further investigations on the reactivity of DNICs and on synthetic improvements for possible therapeutic applications. Planar NHC ligands mimic ligands such as imidazole, itself a surrogate for histidine. A base-promoted conversion was observed of an N-alkyl imidazole bearing, neutral {Fe(NO)2}^10 DNIC to the NHC-DNIC, while maintaining the Fe(NO)2 unit intact. Subsequent alkylation led to the isolation of the NHC-DNIC product; signifying the integrity of the Fe(NO)2 unit and further establishing that NHCs are 'better' ligands to Fe(NO)2 than imidazoles. Making use of an NHC-stabilized DNIC [(NHC)(RS)Fe(NO)2], a RS−/RS[derivative dot] oxidation process in the presence of added CO was discovered, wherein the oxidized {Fe(NO)2}^9 is reduced to {Fe(NO)2}^10 through carbon monoxide (CO)/RS[deritvative dot] ligand substitution. Kinetic studies established a bimolecular rate law and activation parameters suggested an associative mechanism. Computational results indicated a unique role for the delocalized frontier molecular orbitals of the Fe(NO)2 unit, involving an initial side-on approach of CO to the electron-rich N-Fe-N site. Attempts to further test the aforementioned computational proposal employed classical Hammett correlations for the elucidation of structure-activity relationships. Hammett correlations were used to monitor the changes in electron density at the {Fe(NO)2} unit and effects on the rate of reaction with CO of a series of (NHC)(SPhX)Fe(NO)2 derivatives with varying para substituents at the aryl thiolate. In accordance with the computational mechanistic proposal, rate enhancement was observed with increasing electron donating ability of the substituents. Although DNICs appear to be promising candidates in the context of nitric oxide releasing therapeutics, their lack of biocompatibility has hindered the progress of such applicability. The first examples of 'sugar appended' monomeric and dimeric DNICs were synthesized, making use of the tuning sites provided by the NHC-DNIC architecture. Book excerpt: Herein we describe the synthesis, characterization, and role several iron complexes in the coupling of pyrrole and phenylboronic acid to form 2-phenylpyrrole. The oxidation state and spin state of the iron complexes were characterized using X-ray crystallography, UV-vis absorbance spectroscopy, cyclic voltammetry, and in some cases electron paramagnetic reasonance spectroscopy. Furthermore, the results indicate that the iron(III) complexes are essential for catalytic and regioselective production of the 2-phenylpyrrole product. The complexes were compared to evaluate the effect of five properties on catalyst reaction yields: the coordination requirements of the catalyst, half-potential, topological constraint/rigidity, N-atom modification(s), increasing oxidative stability of the complex, and geometric parameters. The need for two labile cis-coordination sites was confirmed based on a 42% decrease in catalytic reaction yield observed when complexes containing penta-dentate ligands were used in place of complexes with tetra-dentate ligands. A strong correlation between iron(III/II) redox potential and catalytic reaction yields was also observed, with [Fe2+L6(Cl)2] providing the highest yield (81%, -405 mV). A Lorentzian fitting of redox potential versus yields predicts that these catalysts can undergo more fine tuning to further increase yields. Interestingly, the remaining properties explored did not show a direct, strong relationship to catalytic reaction yields. Furthermore, the role of the sacrificial oxidant, the lack of radical participation, and the formation of μ-oxodiiron species was established leading to a more detailed mechanistic cycle. Finally, a library of five pyclen-based ligands have been developed for use as iron and europium chelators. A preliminary investigation into iron PCTA complexes indicate that the iron center can bind to all seven of the ligand donors, however, the iron complex does not afford a CEST signal. Establishing that heptadentate ligands containing carboxylic acid pendent arms are not ideal for the developments of iron PARACEST agents.
