This auspicious designation reflects their importance in numerous biological functions. Indeed, life as we understand it relies on the full range of biological processes that are either performed by or catalyzed by porphyrin-containing proteins. Chlorophyll-containing photosynthetic reaction centers in plants, for instance, convert light energy into chemical energy while producing oxygen along the way. It is this oxygen, evolved from photosynthesis, that is transported, stored, and reduced by heme-containing proteins in many organisms, including mammals. Not surprisingly, therefore, these molecules remain of fundamental interest to chemists and biochemists. Indeed, they continue to be intensely investigated by researchers world-wide. Inspired by the importance of the porphyrins, a new research direction has emerged in recent years that is devoted to the preparation and study of non-porphyrin polypyrrole macrocycles. Here, the principal objectives have been to generate completely synthetic systems that bear some structural resemblance to naturally-occurring porphyrin derivatives while being quite different in their specific chemical makeup. Within this context, three different research directions have evolved, namely those involving the syntheses of contracted, isomeric, and expanded porphyrins, respectively. It is the chemistry of these systems that is the subject of this book. Because of the newness of the field, the emphasis of this book will be on synthesis and characterization (all work on porphyrin isomers and much of that associated with expanded porphyrins has only appeared in the last 10 years). One chapter on applications has, however, been included. Also, in the context of the preparative portions of the text, some efforts have been made to explain why various porphyrin analogue targets are of interest.

Book excerpt: Porphyrins and their analogs are a class of chemically and biologically important compounds that have found a variety of applications in different fields such as catalysis, medicine, and materials. The physical, chemical, and biological dependence of the peripheral substituents of porphyrins on their properties has prompted great effort towards the synthesis of new porphyrins with different electronic, steric, and conformational environments. To this end, porphyrins have been prepared using a modular approach from bromo- and triflate synthons. These synthons underwent palladium-catalyzed cross-coupling with chiral amines, amides, alcohols, and boronic esters to create products that were tested for biological activity. Metalloporphyrins were screened as catalysts for cyclopropanation and C-H amination, yielding excellent results. By changing the porphyrin catalystsa[rsquo] chiral groups, all four enantiomers could be produced in the cyclopropanation of styrene derivatives with ethyl diazoacetate (or t-butyl diazoacetate). Similarly, a variety of sultams were produced from benzenesulfonyl azides in high yields and high enantioselectivities using chiral cobalt porphyrins as catalysts. Porphyrins, metalloporphyrins, and the catalytic products generated were tested for activity in a variety of medicinal collaborations, namely as therapeutics for methicillin-resistant Staphylococcus aureus, Alzheimera[rsquo]s disease, malaria, viral infections that include influenza and herpes, and cancer, as well as biological studies with ferrochelatase. They were also used in materials experiments with two different polymer groups.

In her thesis, Lina Blusch not only explains the challenging synthesis and characterization of the ligand system, but also its application to the synthesis of homo- and hetero-bimetallic Ni and Cu complexes. She observes interesting metal-metal-interactions in the complexes, that lead to a non-innocent multistep redox chemistry. The ligand system and its complexes show an intriguing twisted geometry, giving rise to helical chirality and other fascinating properties. This study explores the first steps and opens up a new chemistry of expanded porphyrins with the potential to biomimetic applications.
