Book excerpt: AIM: The main objective of this clinical trial was to evaluate early plaque formation on non-resorbable polytetrafluoroethylene (PTFE) membranes having either a dense (d-PTFE) or an expanded (e-PTFE) microstructure and exposed to the oral cavity of healthy volunteers. MATERIAL & METHODS: Twelve healthy individuals were enrolled in this study. Five test membranes, non-resorbable expanded polytetrafluoroethylene (e-PTFE) with a dual-layer configuration and five control non-resorbable dense (d-PTFE) membranes, were bonded on the buccal surfaces of teeth of each subject, who refrain from toothbrushing during the study period. At two time points, 4h and 24h, the specimens were detached from the teeth and transferred to individual tubes and well plates for the respective microbiological analyses. Plaque samples were also harvested from neighboring teeth as negative control. Quantification of the total aerobic and anaerobic bacteria and Staphylococcus sps. was performed by means of viability counting (colony-forming units per sample). Biofilm quantification of live/dead bacteria was performed by confocal laser scanning microscopy (CLSM) and descriptive assessment of plaque by scanning electron microscopy. Wilcoxon matched-pairs signed rank tast was applied for statististics. RESULTS: All 120 applied membranes on the tooth surfaces of 12 volunteers were available for analyses after the respective accumulation periods.Between the early (4h) and later (24h) time-points viable bacterial counts (CFU) increased on all membranes, with no difference between test and control. While the number of viable aerobes and anaerobes was greater on the tooth surfaces compared to the membranes at both time points, it was observed that the number of Staphylococcus sps. had a different colonization pattern: they decreased on the tooth surfaces and increased on both membranes over time, with a statistically significant difference compared to the tooth surfaces at 4h for both membranes and at 24h for the test membranes. Quantitative analysis of biofilm CLSM images showed that the amount of live and dead biofilm biomass and thickness was greater on the control than on the test membranes at 4h while no statistically significant difference could be observed at 24h between the two membranes.CONCLUSIONS:This study demonstrated that d-PTFE membrane was associated with greater biofilm biomass and thickness during the initial exposure compared to e-PTFE membrane. The current experimental setup could provide a valuable toolbox to study the in vivo behavior of different membranes or antimicrobial strategies on bacterial adhesion and biofilm formation.
