Book excerpt: The thesis describes the first total synthesis of Eucophylline, the south fragment of the Leucophyllidine alkaloid recently isolated from Leuconotis griffithiiand L. eugenifolius. The 10-step synthesis encompasses a new straight forward method to access to the azabicyclo[3.3.1]nonane skeleton through a free-radical three-component olefinic carbo-oximation process. During these studies, we also isolated enamines exhibiting unusual stabilities resulting from their strained bicyclicconformation. The synthesis of the Eburnane part, the north fragment of Leucophyllidine, was developed relying on a new free-radical three-component Carbo-Cyanation reaction. This new reaction showed high tolerance toward different functional groups and provided an efficient mean to install the nitrile group in complex structures and to introduce quaternary centers. The biomimetic coupling between the Eburnane and Eucophylline fragments was finally studied using structurally similar models. Preliminary results of this model study showed that absence of a gemdimethyleffect in the Eburnane model probably prevents the formation of the cyclicimine, key intermediate for the desired coupling.

Book excerpt: This thesis presents our studies towards the first total synthesis of the novel anti- HIV agent lancifodilactone G, which has a highly unusual aliphatic enol. The first chapter provides a survey of architecturally diverse nortriterpenoids that were isolated from the Schisandraceae family. A proposed biosynthetic pathway for lancifodilactone G and closely related natural products provides a rationale for the formation of the consecutive 7/8/5 fused carbo cycles that are unique to Schisandra nortriterpenoids. Chapter 1 goes on to outline the reported strategies to access the core of lancifodilactone G and concludes with a retrosynthetic analysis proposed by the Evans group, which includes a biosynthetically inspired single-pot polycyclisation reaction. Chapter 2 describes the highly stereocontrolled synthesis of the eastern fragment (F-G rings) using transition metal-mediated Pauson-Khand reaction. This chapter also reviews the metal-mediated diastereoselective Pauson-Khand reaction directed by the stereogenic centre at C2, with the ample illustration to total synthesis. Attempted strategies for the assembly of the bicyclic cyclopentanone motif via a dienyl Pauson-Khand reaction of silicon- and oxygen- tethered diene-enes are presented. The failure of these strategies at different stages of the synthesis resulted in the exploration of a classical Pauson-Khand approach, which successfully furnished the eastern fragment. Finally, a second-generation synthesis is described which provided the fully functionalised eastern fragment with improved efficiency and overall yield. Chapter 3 discusses the successful synthesis of the western fragment (B-C rings) using a diastereoselective [4+3] cycloaddition strategy. Attempted strategies for the synthesis of the key 2,3-disubstituted furan derivative are presented, which was achieved via a hetero Pauson- Khand reaction. This chapter includes a brief account of the classical [4+3] cycloaddition reactions of furans using an in situ generated oxyallyl cation and also employing vinyl carbenoids in the metal-catalysed version. The review also highlights the application of the [4+ 3] cycloaddition reaction in the expeditious assembly of functionalised 7-membered rings that occur in a number of important biologically active natural products. The third chapter goes on to describe the application of these cycloaddition reactions in the synthesis of the fully functionalised western fragment of Lancifodilactone G. Chapter 4 describes a model study aimed at expediting the synthesis of the western fragment using a rhodium-catalysed allylic substitution reaction. A brief mechanistic discussion on unique aspects of the allylic alkylation reaction is illustrated. Chapter 4 concludes by outlining the coupling strategy for eastern and western fragments and the end game studies for the completion of the synthesis of lancifodilactone G.

Book excerpt: The route to the key bicyclic intermediate was streamlined to eight steps in 16% yield, using a TMSI promoted coupling of a furan and an enone. Additionally, methodology for the selective ozonolysis of the bicyclic intermediate was developed via ozone titration. Work on the dihydroxylation step led to the discovery and development of a new pH-neutral Sharpless-style asymmetric dihydroxylation. The synthesis of pentacyclic core of citreamicin μ was accomplished in 12 steps. New methodologies were developed, including: an ortho- selective bromination of a vanillin derivative and the use of 4-(Phenylazo)diphenylamine (PDA) as an internal indicator for the acetylide coupling. The usefulness of PDA led to its development as a general all-purpose indicator for the titration of strong bases, Lewis acids and reducing agents. The discovery of (n-Bu)4NOAc as a privileged additive led to the development of new methods for the synthesis of isocoumarins and new methodology for the condensation of amino acids using LiMe4 was developed.

Book excerpt: Progress towards the total synthesis of gambierol inspired by the biosynthetic hypothesis for ladder polyethers is described. Syntheses of the ABCD and FGH fragments have been completed in 29 and 3 1 steps, respectively, using water-promoted endo-selective epoxide-opening cascades to construct the poly-THP motifs. The concentration and temperature effects in the waterpromoted cascades were examined, and an operationally simple protocol was developed for the synthesis of polycyclic cascade products on multigram scale. [diagram] A sulfone tether between the ABCD and FGH fragments was used to facilitate an intramolecular olefination via the Ramberg-Backlund reaction. The highly site-selective hydrogenation of the fragment coupling alkene in the presence of H-ring alkene was developed using Noyori's hydrogenation catalyst, and several ring E cyclization precursors were subsequently prepared from C21 ketone. Investigation of a variety of methods for the C21-0 bond construction revealed a remarkably general spiroketalization pathway, which was previously unprecedented in ladder polyether synthesis and invariably led to the isolation of undesired 6,6-spiroketal products under most conditions. We have hypothesized that the presence of the FG ring junction methyl group disfavors the desired cyclization by exerting destabilizing 1,3-diaxial strain onto the newly forming ring junction of the requisite oxepane. We have therefore proposed that future studies examine the reverse direction for the synthesis of ring E via C16-0 bond formation. This approach would require installation of the tertiary alcohol on ring F prior to cyclization and would thus avoid the undesired reactivity resulting from the steric effects of the FG ring junction methyl group. Studies were conducted with the purpose of completing the first total synthesis of 14-isoazacamptothecin. During the courses of this endeavor, new insights into pyridine chemistry were uncovered. It was demonstrated for the first time that C-4 substitution of 2,5-dibromopyridine via C-4 lithiation is indeed possible. New reactions in heterocyclic chemistry, namely (a) “Halogen Dance-Lithium Migration-Halogen Dance†or “Lithium Migration-Halogen Dance†and (b) Double Halogen Dance†were discovered to have taken place as explanations for the observation of unexpected products. Synthetic methods toward the synthesis of new quinoline analogs using readily available and inexpensive starting materials were developed. Although the initial inspirational target of the project was the AB ring fragment of CPT analogs, new techniques and methods were developed that would be useful in far reaching applications such as in the synthesis of clinically useful compounds and natural products containing a quinoline ring in the parent structure.
