Book excerpt: Avian Influenza (AI) and Newcastle Disease (ND) are two devastating diseases of poultry, which cause losses to the poultry industry and influence the liveability of rural communities worldwide. Following the H5N1 epidemic they appear to be endemic at least in Asia, Eastern Europe, The Middle East and Africa. Particularly in case of AI outbreaks it is essential that infection is diagnosed promptly and that isolates are made available to the international scientific community. Currently, several organisations including OIE, FAO and the EC have organised training courses in affected areas. However, often these courses do not cover all aspects of AI/ND diagnosis but only certain aspects. This results in fragmented areas of knowledge and in the application of different diagnostic protocols in different parts of the world. The objective of this book is to provide a comprehensive approach to AI diagnosis ranging from the clinical elements that should trigger a suspicion in the field, to the post mortem technique, collection of samples, processing/ shipment of specimens, virological, serological and molecular diagnosis and guidelines for notification.

Book excerpt: Newcastle disease is a highly contagious viral disease affecting many domestic and wild avian species that have severe impact on poultry industry. It can also be transmitted to humans that cause conjunctivitis and flu like symptoms. Effective vaccination of flock is the means to control this disease. This book was to investigate persistence of maternal antibody as well as the comparative efficacy of Avinew and BCRDV vaccines against Newcastle disease in chicks. Birds of groups A and B were primarily and secondarily vaccinated with Avinew and BCRDV respectively at the age of day 3 and 20. Sera samples were obtained from 10 randomly selected birds on each occasion of day 1, 10, 17, 27, 30, 34 and 44 of age of birds. Birds of group C were maintained as unvaccinated control for the determination of existence of maternal antibody. Thus it was indicated that performance of Avinew in respect of elucidation of HI antibody was comparatively better than that of BCRDV and maternal antibody against NDV in chicks persisted to a minimal until the age of day 27 and none at day 30 or 34. This analysis will be useful to professionals working for poultry industry, researcher and vaccine manufacturer.

This book was released on 2009 with total page 322 pages. Available in PDF, EPUB and Kindle. Book excerpt: Since 2002, the South African poultry industry has experienced outbreaks of Newcastle disease (ND) caused by a newly introduced virus (NDV) strain belonging to lineage 5d/VIId ("goose paramyxovirus" - GPMV). Control of the disease has proved difficult with commercially available vaccines appearing ineffective. In the first of two studies, broilers chicks were vaccinated with VG-GA vaccine (lineage II), then challenged with both GPMV and a "classic" challenge virus (RCV) of lineage 3d/VIII to compare the efficacy of the vaccine against both strains. In the second study, commercial and SPF hens in lay were vaccinated with La Sota vaccine and challenged with GPMV isolate, and immunohistochemistry staining used to determine the distribution pattern of viral antigen in the oviduct of the hens. Results from the second study showed no clear difference in the protection of the oviduct from challenge with GPMV by either the cloacal and ocular routes of vaccination. Vaccinated birds were fully protected (100%) against challenge by La Sota vaccine, but not against infection and replication of the virus, as birds showed varying degrees of macropathology with numerous stained viral antigens in the oviducts demonstrated by immunohistochemistry. The susceptibility and colonisation of the oviduct of laying hens by both the lentogenic La Sota and the virulent NDV isolates was confirmed, with the uterus being more susceptible than magnum and isthmus. Necrosis and apoptosis of cells of the oviduct were not detected but cellular infiltration, gland dilatation and interstitial oedema were observed.

Written by experts from both the commercial poultry field and the companion avian field, the book provides thorough coverage of both common and less common diseases of backyard chickens, ducks, and other poultry. The book begins with introductory chapters covering general information, an overview of US laws, and basic husbandry concerns, then moves into specific disease chapters organized by system. The book takes an individual medicine perspective throughout, with photographs, radiographs, and histopathological photomicrographs to illustrate principles and diseases. Backyard Poultry Medicine and Surgery is an invaluable guide to diseases and treatments for any practitioners treating backyard poultry.

This book was released on 2006 with total page 220 pages. Available in PDF, EPUB and Kindle. Book excerpt: Newcastle disease (ND), caused by Newcastle disease virus, is an acute, contagious and pathogenic infection of pet, free living and domestic birds. ND is an epidemic disease and it is responsible for high economic losses due to up to 100 % mortality. The control of ND in the intensive commercial poultry farms is largely dependent on prophylactic immunisation using conventional vaccines. The ND V4 vaccine and its derivative ND V4-HR vaccine were selected originally for use in village chickens, due to their immunogenicity, thermostability, transmissibility and ease of administration. The efficacy of V4 and V4HR vaccines have been established in many Asian and African countries in their ability to challenge a wide range of recognised and local velogenic NDV. Therefore, ND V4 was tested for efficacy against B1172 challenge NDV isolated in south Africa in 1993. Twenty-eight one day-old replacement pullets were vaccinated by eye-drop route at 21 and 49 days old. Chickens vaccinated by eye-drop route were left to mingle with the unvaccinated in-contact chickens. At 63 days all chickens including the unvaccinated control group were individually challenged with B1172 NDV. Serological monitoring of NDV antibody response was done using HI and ELISA tests. The ND V4 vaccine induced full protection against B1172 NDV in chickens vaccinated by eye-drop vaccination and in 55 % of chickens vaccinated by the in-contact method. No association was seen between NDV antibody titer at pre-challenge and the ability to withstand B1172 challenge NDV. A fair to good agreement was seen between the HI and ELISA test in monitoring NDV antibody response during the experiment. Although, the ELISA showed a higher sensitivity and specificity than the HI test, further studies are required using this method of comparison.

Book excerpt: Glucocorticoids regulate multiple metabolic and developmental processes and play a vital role in the maintenance of basal and stress-related homeostasis. For the last 50 years, pharmacologic doses of glucocorticoids have been used in the treatment of inflammatory, autoimmune, and lymphoproliferative diseases and in the prevention of allograft rejection, while substitution doses have been employed in the management of adrenocortical insufficiency. aspects of glucocorticoid action, in particular, (i) the impact of maternal and early life stress on stress-related gene regulation in the offspring; (ii) the importance of glucocorticoids and their receptors; (iii) further understanding of the mechanisms of GR action, including its effect on chromatin modulation, its interaction with coactivators and corepressors, and the genetic dissection of GR function in mice; (iv) The interaction of hGR with other transcription factors, such as NF-kappa-B, p53, transforming growth factor beta (TGF-beta) and the chicken ovalbumin upstream promoter transcription factor II (COUP-TFII); recycling, ubiquitination and degradation of the receptor, actions of the GR-beta isoform, a novel synthetic nonsteroidal target gene-specific agonist, the importance of target tissue activity of 11-beta-hydroxysteroid dehydrogenase type 1 in glucocorticoid action in health and disease, the interaction of the receptor with the nutrient carnitine, the anthrax products protective antigen (PA) and lethal factor (LF), and the human immunodeficiency virus type-1 (HIV-1)-encoded molecules Vpr and Tat; (vi) an update on the effects of glucocorticoids on the immune system; and (vii) the clinical implications of glucocorticoid action, including glucocorticoid resistance/hypersensitivity, familial and sporadic glucocorticoid resistance, and the effects of stress and depression.
