During this period it has been actively developed and the range of problems tackled greatly extended. Originally it was used for studies of transient species involv ing vibrational, electronic and ESR spectroscopy. Nowadays the study of transient species forms a comparatively small part of HI work since it has been amply demonstrated that very fruitful information can be obtained of the structure and interactions of stable molecules and their aggregates. In addition to the s~ectroscopic methods mentioned above the MI technique is nowadays a standard method in research based on vibrational relaxation, luminescence, Mossbauer, magnetic circular dichroism, pulsed NMR and photoelectron spectroscopy. The matrix isolation technique affords considerable advantages over more conventional methods in most applications of spectroscopy. Areas where the technique has been widely applied, or shows great potential, include: metal atom chemistry, and its relation to surface chemistry, high temperature inorganic species, transition metal complexes, interstellar species, free radicals and unstable molecules, conformational studies, molecular com plexes, and intermolecular forces.

Book excerpt: The combination of Fourier transform infrared (FT-IR) spectroscopy with the matrix-isolation techniques has advantages compared with the use of more conventional grating spectroscopy. Furthermore, the recent commercial availability of Fourier transform spectrometers has made FT-IR a practical alternative. Some advantages of the FT-IR spectrometer over the grating spectrometer are the result of the computerized data system that is a necessary part of the FT-IR spectrometer; other advantages are a consequence of the difference in optical arrangements and these represent the inherent advantages of the FT-IR method. In most applications with the matrix-isolation technique, the use of FT-IR spectroscopy results in either an improved signal-to-noise ratio or a shorter time for data collection compared with grating infrared spectroscopy. Fourier transform infrared spectroscopy has been used in the laboratory to study several molecular species in low-temperature matrices. Some species have been produced by high-temperature vaporization from Knudsen cells and others by sputtering. By sputtering, Ar and Kr matrices have been prepared which contain U atoms, UO, UO2, UO3, PuO, PuO2, UN, or UN2, depending upon the composition of the gas used to sputter as well as the identity of the metallic cathode. Infrared spectra of matrices containing these compounds are presented and discussed. (JRD).

It is natural that as the field of biochemical dynamics has achieved new levels of refinement its students have increasingly adopted the concepts and methods of transition-state theory. Indeed, every dynamical problem of biochemistry finds its most elegant and economical statement in the terms of this theory. Enzyme catalytic power, for example, derives from the interaction of enzyme and substrate structures in the transition state, so that an understanding of this power must grow from a knowledge of these structures and interactions. Similarly, transition-state interactions, and the way in which they change as protein structure is altered, constitute the pivotal feature upon which molecular evolution must turn. The complete, coupled dynamical system of the organism, incorporating the transport of matter and energy as well as local chemical processes, will eventually have to yield to a description of its component transition-state structures and their energetic response characteristics, even if the form of the description goes beyond present-day transition-state theory.
