The effects of branched-chain amino acids on fatty acid metabolism was studied. The ketoacid of leucine (KIC) inhibited the oxidation of oleate in the perfused heart. Although the ketoacids of valine (KIV) and isoleucine (KIL) were decarboxylated in cardiac muscle, they did not affect fatty acid oxidation. This appears to be due to the fact that KIC is almost completely oxidized in the heart, whereas only 19% of KIV is completely oxidized. -- 3-Hydroxyisobutyrate, an intermediate in ketovaline metabolism, was shown to be released from heart when KIV was the substrate. 3-Hydroxyisobutyrate was shown to be a substrate for gluconeogenesis in isolated kidney cortical tubules and hepatocytes.

Book excerpt: Branched-chain amino acids (BCAAs) are essential amino acids that are crucial for skeletal muscle anabolism. Compared to the effects of these amino acids on skeletal muscle, much less is known on how impairment in BCAA catabolism affects this tissue. BCAA catabolism starts with the reversible transamination by the mitochondrial enzyme branched-chain aminotransferase 2 (BCAT2). This is followed by the irreversible decarboxylation, catalyzed by branched-chain -keto acid dehydrogenase (BCKD) complex. Our lab has previously shown that BCAT2 and BCKD catabolic activities are essential for the differentiation of skeletal myoblasts into myotubes in vitro. Here, we investigate the effect of depletion of BCAT2 or BCKD in differentiated myotubes. We demonstrate that disrupting BCAT2 negatively affects myotube survival. Furthermore, we show that BCKD depletion augments myofibrillar protein content and increases mTORC1 signaling in myotubes. Conclusively, interventions that target BCKD abundance or functions may promote muscle protein anabolism in individuals with muscle wasting conditions.

Book excerpt: Branched-chain amino acids are indispensible amino acids that are known for having anabolic effects in skeletal muscle. These effects BCAAs elicit in existing skeletal muscle are well documented in literature. However, much less is known about how they affect the process of myogensis and muscle regeneration. Data generated in our lab have shown that the intact BCAA catabolic pathway is required for proper skeletal muscle cell differentiation. In the current study, the main enzymes involved in the catabolism of BCAAs were assessed, in addition to the activity of the rate-limiting enzyme (Branched-chain -ketoacid dehydrogenase - BCKD) in the pathway. Increasing the flux of BCAAs through this pathway via siRNA mediated knockdown of a negative regulator of the pathway augmented differentiation. It is evident that BCAA catabolism is intricately involved in the process of muscle cell differentiation. Further study into the regulation of BCAA catabolism during differentiation is warranted; to aid in the development of nutritional based interventions for populations suffering from muscle wasting condition.
