Book excerpt: Separating proteins in aqueous solutions through fractional precipitation by addition of salts is one of the oldest protein separation methods, and it remains the only purification step for some industrial enzymes and an initial purification step for higher-value therapeutic proteins. However, despite its widespread application, the fundamental characteristics of protein precipitation by salt are still incompletely understood. The primary focus of this thesis is the experimental exploration of the phase behavior of single and binary protein systems during protein precipitation in concentrated ammonium sulfate and sodium chloride solutions, and the effect of solution conditions on protein interactions. Protein precipitation phase behavior of single protein systems was measured as a function of solution conditions for lysozyme and ovalbumin. X-ray powder diffraction (XRD) and optical microscopy were used to characterize the structure of the protein precipitates. These studies showed that crystallization after initial precipitation is feasible even at high ionic strengths if sufficient mixing time is allowed, and such behavior may be a more general phenomenon although different crystallization pathways may be found in some systems and under different conditions; for instance, different pathways are seen in ovalbumin-ammonium sulfate systems as a function of pH. A more important development is the interpretation that emerges of precipitation as a manifestation of a metastable liquid-liquid separation, with the precipitate being a kinetically trapped dense liquid phase. Furthermore, the diversity of possible dense phases (amorphous precipitate, crystal phases or gel) associated with liquid-liquid phase separation shows that understanding the effects of protein interactions of different kinds and at different ranges could have important consequences for the systematic control of protein phase behavior. The phase behavior in selective precipitation of lysozyme-ovalbumin binary mixtures was investigated at various conditions. The best combination of selectivity and recovery of lysozyme was achieved in pH 7 NaCl solutions. At an initial concentration of 30 mg/g H 2 O of lysozyme and 30 mg/g H 2 O of ovalbumin, more than 80% of the lysozyme was collected in precipitate form with 93% purity at ionic strengths above 3 m, while ovalbumin was collected in the supernatant at 80% purity. The purities of both proteins were therefore improved by at least 30% in one step. Moreover, crystallization following initial precipitation was observed in some binary precipitation experiments and indicates that binary precipitation, like single protein precipitation, is a metastable equilibrium state. The observations of selective precipitation behavior were correlated with values of the osmotic second virial coefficient, measured by cross-interaction chromatography (CIC). The results show that interaction measures such as protein self- and cross-interactions can be used as a predictor for general trends and/or inconsistencies in the separation behavior and for optimizing working conditions. Such a mechanistic relationship can obviate the need for extensive trial-and-error methods in order to optimize conditions for protein separations.

Available in PDF, EPUB and Kindle. Book excerpt: This book is the first to be entirely devoted to the challenging art of handling membrane proteins out of their natural environment, a key process in biological and pharmaceutical research, but one plagued with difficulties and pitfalls. Written by one of the foremost experts in the field, Membrane Proteins in Aqueous Solutions is accessible to any member of a membrane biology laboratory. After presenting the structure, functions, dynamics, synthesis, natural environment and lipid interactions of membrane proteins, the author discusses the principles of extracting them with detergents, the mechanisms of detergent-induced destabilization, countermeasures, and recent progress in developing detergents with weaker denaturing properties. Non-conventional alternatives to detergents, including bicelles, nanodiscs, amphipathic peptides, fluorinated surfactants and amphipols, are described, and their relative advantages and drawbacks are compared. The synthesis and solution properties of the various types of amphipols are presented, as well as the formation and properties of membrane protein/amphipol complexes and the transfer of amphipol-trapped proteins to detergents, nanodiscs, lipidic mesophases, or living cells. The final chapters of the book deal with applications: membrane protein in vitro folding and cell-free expression, solution studies, NMR, crystallography, electron microscopy, mass spectrometry, amphipol-mediated immobilization of membrane proteins, and biomedical applications. Important features of the book include introductory sections describing foundations as well as the state-of-the-art for each of the biophysical techniques discussed, and topical tables which organize a widely dispersed literature. Boxes and annexes throughout the book explain technical aspects, and twelve detailed experimental protocols, ranging from in vitro folding of membrane proteins to single-particle electron cryomicroscopy, have been contributed by and commented on by experienced users. Membrane Proteins in Aqueous Solutions offers a concise, accessible introduction to membrane protein biochemistry and biophysics, as well as comprehensive coverage of the properties and uses of conventional and non-conventional surfactants. It will be useful both in basic and applied research laboratories and as a teaching aid for students, instructors, researchers, and professionals within the field.

Book excerpt: Fundamental studies on aqueous two-phase systems and differential ammonium sulfate precipitation have been performed in this dissertation. The significant findings in this dissertation are: (1) A semi empirical correlation relating the partition coefficient of phase-forming polymers in aqueous two-phase systems to the differences in phase-forming polymer concentrations between the top and bottom phases has been developed. Good agreement between the correlation and the experimental results was obtained. (2) The partitioning behavior of PEG-protein conjugates in PEG/potassium phosphate and PEG/dextran systems has been investigated. A correlation that relates the partition coefficient of PEG protein conjugates to the partition coefficient of PEG and protein, the number of PEG conjugating to the protein, and the hydrophobicity changes due to a covalent bond and the protein's structural change is derived and tested with the experimental results. (3) Countercurrent distribution in aqueous two-phase systems and differential ammonium sulfate precipitation have been successfully applied to fractionate PEG-protein conjugates. (4) Regarding centrifugal precipitation chromatography, a mathematical model of ammonium sulfate gradient formation in the separation channel of centrifugal precipitation chromatography has been developed and solved numerically. To demonstrate the ability of the technique to purify proteins, mixtures of proteins were subjected to the technique. From the experiment, the resolution was a function of the degree of saturation of ammonium sulfate solution fed into the salt channel. (5) A mathematical model of protein retention in centrifugal precipitation chromatography has been developed and solved numerically. Using lysozyme as a protein model, the numerical results are compared with the experimental result and good agreements between the numerical and experimental results are achieved. (6) The effect of centrifugal precipitation chromatography on enzymatic activities of proteins has been investigated. Using alpha-chymotrypsin as a protein model, it is observed that the effect of fluid mixing in the separation channel of centrifugal precipitation chromatography on the enzymatic activity of alpha-chymotrypsin is minimal. Therefore, most proteins should remain active after being subjected to centrifugal precipitation chromatography. A better channel design and operating centrifugal precipitation chromatography at low temperature should reduce the losses in enzymatic activity of proteins further.

Book excerpt: Low-Abundance Proteome Discovery addresses the most critical challenge in biomarker discovery and progress: the identification of low-abundance proteins. The book describes an original strategy developed by the authors that permits the detection of protein species typically found in very low abundance and that may yield valuable clues to future discoveries. Known as combinatorial peptide ligand libraries, these new methodologies are one of the hottest topics related to the study of proteomics and have applications in medical diagnostics, food quality, and plant analysis. The book is written for university and industry scientists starting proteomic studies of complex matrices (e.g., biological fluids, biopsies, recalcitrant plant tissues, foodstuff, and beverage analysis), researchers doing wet chemistry, and graduate-level students in the areas of analytical and biochemistry, biology, and genetics. Covers methodologies for enhancing the visibility of low-abundance proteins which, until now, has been the biggest challenge in biomarker progress Includes detailed protocols that address real-life needs in laboratory practice Addresses all applications, including human disease, food and beverage safety, and the discovery of new proteins/peptides of importance in nutraceutics Compiles the research and analytic protocols of the two scientists who are credited with the discovery of these landmark methodologies, also known as combinatorial peptide ligand libraries, for the identification of low-abundance proteins

Book excerpt: A UNIQUE BOOK ON THE PRESENT STATUS OF SOLVENTS AND SOLUTIONS WITH IMPORTANT PROBLEMS RELATED TO THEIR STRUCTURE AND PROPERTIES The literature on the properties of solvents and solutions used in academic research and in a wide range of industries has grown enormously during the last four decades, and is scattered in different specialized journals. Solvents and Solutions is a groundbreaking text that offers a systematic compilation of important problems related to selected properties of solvents and solutions based on the literature published so far. The author places emphasis on explaining the basic concepts involved in understanding the properties and behavior of various solvents and solutions of electrolytes and nonelectolytes in a consistent manner. After a description of the general characteristics of structure of solvents and solutions and the solubility of electrolytes and nonelectrolytes under normal temperature and pressure conditions, the book first deals with different aspects of the density and the refractive index of solvents and dilute as well as concentrated solutions, and finally with the transport (i.e. viscosity and electric conductivity) and thermal properties of solvents and solutions. Solvents and solutions is the first text devoted to the description and discussion of their properties since the publication of a monograph on the physical properties of aqueous electrolyte solutions more than three decades ago. The main features of this book are: Reflects developments in the investigation of solvents and solutions during the last three decades. Outlines basic concepts involved in understanding the properties and behavior of solvents and solutions. Describes and discusses different properties of ionic liquids as solvents and the behavior of their mixtures with other commonly used solvents. Contents of different chapters are not only self-contained but the contents are practically independent of each other. Written as a practical guide for researchers who are looking for an uptodate overview of the physical and transport properties of solvents and solutions, and as a reference source for workers in chemical industries and related fields and for graduate students of chemical engineering and physical chemistry.
