Book excerpt: Over the last decade, the use of highly branched macromolecules, called dendrimers, is emerging. Polyamidoamine (PAMAM) dendrimers, in particular, have attracted high interest due to their unique properties, such as well-defined size, shape, and solubility in water. Our group, as part of an interdisciplinary team, is currently working on developing a nanoextraction technology for high-yield dendrimer syntheses. Impurities may occur during the synthesis of these dendrimers, and therefore, to achieve higher purity products, chromatographic separations of dendrimers are an important part toward this goal. This thesis describes approaches toward this objective, focusing in two areas: at first, development of materials and devices that will enable separations of dendrimers in confines of capillary and chromatographic columns, and second, evaluation of the feasibility of transferring the separation techniques previously developed to the channels of microfluidic devices. The results showed that PAMAM dendrimers of various generations were successfully analyzed using capillary zone electrophoresis (CZE), capillary electrochromatography (CEC), and high-performance liquid chromatography (HPLC). Both CZE and HPLC have allowed for fast generational separations up to the fifth generation. Moreover, CZE can also assess the homogeneity and presence of side products in the synthesized dendrimers. RP-HPLC, on the other hand, allowed for separations of dendrimers with terminal amino and carboxyl groups. PAMAM dendrimers can also be used as template molecules to prepare monolithic capillary columns for liquid chromatography and capillary electrochromatography. This method has provided two advantages: 1) dendrimers can be used as template molecules for controlling pore structures, 2) selective sorbents can be produced with surface sites that are complementary in chemical functionality to the dendrimer template molecule. These highly selective sorbents were able to provide separation of dendrimers from various generations, particularly of high generations, in the CEC mode. As the first step toward the development of a nanoextraction technology for high yield dendrimer syntheses, the feasibility of extending the separation techniques developed on-column into channels of a microfluidic chip was examined. In this study, electrophoresis was employed to separate different generations of dendrimers in glass microfluidic chips using laser-induced fluorescence detection. The use of microfluidic system for dendrimer application offers great potential due to its rapid analysis times, small sample volume requirements, and the ability to integrate several steps of analyses in one chip.

Book excerpt: The interior physical and chemical properties of amine-terminated poly(amidoamine), PAMAM, and poly(propyleneimine), PPI, dendrimers were investigated using the solvatochromic probes ET-30 and phenol blue. Dendrimers are polymers that are produced using a highly-controlled, stepwise synthetic scheme, which results in very regularly and highly branched, three-dimensional, monodisperse polymers. Their synthetic controllability and unique structural properties has led to exciting application possibilities. Though some characterization of these dendrimers has been published, an improved understanding of the chemical and physical properties of the interior regions of these dendrimers is still needed and is vital to their successful use in future applications. In this research, ET-30 and phenol blue are used as extrinsic probe molecules to elucidate the unique properties of the PAMAM and PPI dendrimers, in part because efforts to provide detailed conformational/structure information via direct methods have had little success. These molecules exhibit different behavior depending upon the properties of their surrounding environment; in particular, both have well characterized responses to microenvironmental polarity. Methods used were UV/Vis absorption spectroscopy and fluorescence emission, anisotropy and lifetime measurements. For every aqueous PAMAM and PPI dendrimer generation studied, two discrete dye populations were clearly observed. Model compounds that mimic the surface groups and branching moieties of both dendrimer families were used to better define the associated dye's location. The mimics confirmed that these probes associated inside the dendrimer, and in the case of phenol blue, it did not interact with the dendrimer surface groups. Both families of dendrimers were shown to form nonpolar associations with the two dyes beyond the surface of the dendrimer. While ET-30 proved too large to be completely encapsulated by the PAMAM dendrimers, phenol blue was found to reside within the PAMAM and PPI dendrimer interior, near their cores. In general, the interior microenvironments of these dendrimers were observed to decrease in polarity for successively larger generation dendrimers. However, the interior polarity of the largest generations of each family remained relatively constant. Important differences among the generations within a dendrimer family and between the two were elucidated. Trends in polarity, rigidity, binding affinity and shape were all observed. In addition, it was found that phenol blue fit better inside the PPI dendrimers, compared to PAMAM. This finding was not a total surprise considering their structures.

I have also studied the potential applications of methoxy poly(ethylene glycol)-block-poly(ether sulfide) [or mPEG-block-poly(ether sulfide)] dendrimers in drug delivery by examining the solubility of pyrene in water in the presence of the various concentrations of the dendrimers. It was found that generation 3 of mPEG-block-poly(ether sulfide) dendrimer modestly increased the solubility of pyrene in water. Applications of these dendrimers as phase transfer catalysts have also been investigated. It was found that generation 3 of mPEG-block-poly(ether sulfide) dendrimer acted as an excellent phase transfer catalyst in the dehydrohalogenation of 2-bromoethyl benzene. The effect of mPEG-block-poly(ether sulfide) dendrimer on calf-thymus DNA has also been investigated by circular dichroism (CD). Finally, the effects of mPEG and mPEG-block-poly(amido amine) dendrimers on Dickerson-Drew Dodecamer DNA have been studied by NMR spectroscopy and CD and the results are presented and discussed.

Book excerpt: The convergent strategy towards dendrimer synthesis is well-suited to generate macromolecules with a diverse periphery, at the expense of time and effort, while the divergent strategy has historically been effective at yielding higher generation dendrimers, although they are often plagued by impurities. Both the convergent and divergent routes were applied to the synthesis of melamine-based dendrimers, offering a comparison of the routes within a system. Generation-1 dendrons heterogeneously functionalized with Boc-protected amines and hydrazones were synthesized convergently and coupled to a generation-1 tris(piperazine) core to yield a generation-2 dendrimer bearing 18 Boc-amines and three hydrazones. Although the yield for the final coupling step was rather low (56%), the yields for all intermediate steps were quite high. Attempts toward obtaining a generation-3 dendrimer through this route were unsuccessful due presumably to steric hindrance. The materials obtained showed no impurities in their 1H and 13C NMR and mass spectra, although several chromatographic purifications were necessary throughout the synthesis. A divergent strategy based on addition of a dichlorotriazine monomer to polyamine cores was used to synthesize dendrimers of generations 1-5. All intermediates and dendrimers were either purified by precipitation, or did not need purification. 1H NMR spectroscopy indicated that reactions were complete up to G4-NH2 by integration, and mass spectroscopy confirmed that assignment. HPLC and GPC of Gn-Cl dendrimers showed sharp peaks for G1-G3, but G4-Cl appeared to have a small amount of impurities that are similar in size and polarity to the fully-substituted dendrimer. The G1-G3 dendrimers were confidently assigned as pure by conventional organic chemistry standards, but the assignment of purity to higher generations remained tentative. A G1-Cl dendrimer was functionalized with imidazole, and then deprotected and PEGylated with PEG5000 to yield a water soluble dendrimer. The imidazole-capped, Boc-protected dendrimer and the deprotected dendrimer were characterized by 1H and 13C NMR spectroscopy and mass spectrometry. The degree of PEGylation on the PEGylated material could not be definitively ascertained; however, the material is capable of solubilizing very hydrophobic Zn-phthalocyanines in water.
