The FAD2 genes belong to such family and play a vital role in converting monounsaturated oleic acid to polyunsaturated linoleic acid. Oleic acid has the health benefits for humans, such as reduction in cholesterol level, antioxidation property, and industrial benefits like longer shelf life. The development of genotypes with high oleic acid content in seeds has become one of the primary goals in breeding oilseed plants. The identification and characterization of the FAD2 genes in plants have been an important step to better manipulate gene expression to improve the seed oil quality. The induction of mutations in FAD2 genes to reduce FAD2 enzyme activity has been an integral approach to generate genotypes with high oleic acid. This chapter will describe the FAD2 gene family in the model organism soybean and the correction of mutations in FAD2 genes with the increase of oleic acid content. Leveraging advanced research of FAD2 gene family in soybean promotes the study of FAD2 genes in other legume species, including peanut. The future perspectives and challenges associated with mutations in FAD2 genes will be discussed.

Soybean has spread to many Asian countries two to three thousand years ago, but was not known in the West until the 18th century. Among the various constraints responsible for decrease in soybean yields are the biotic and abiotic stresses which have recently increased as a result of changing climatic scenarios at global level. A lot of work has been done for cultivar development and germplasm enhancement through conventional plant breeding. This has resulted in development of numerous high yielding and climate resilient soybean varieties. Despite of this development, plant breeding is long-term by nature, resource dependent and climate dependent. Due to the advancement in genomics and phenomics, significant insights have been gained in the identification of genes for yield improvement, tolerance to biotic and abiotic stress and increased quality parameters in soybean. Molecular breeding has become routine and with the advent of next generation sequencing technologies resulting in SNP based molecular markers, soybean improvement has taken a new dimension and resulted in mapping of genes for various traits that include disease resistance, insect resistance, high oil content and improved yield. This book includes chapters from renowned potential soybean scientists to discuss the latest updates on soybean molecular and genetic perspectives to elucidate the complex mechanisms to develop biotic and abiotic stress resilience in soybean. As an oilseed, soybean provides high quality protein for livestock and poultry feed, and the oil is used extensively in the cooking and food manufacturing industries across the world. Soybean producers have targeted improved oil traits as a priority area of research to enhance the market share of U.S. soybean. Soybean seed is approximately 18% oil and standard commodity soybean oil is a mixture of five fatty acids; palmitic (11%), stearic (4%), oleic (22%), linoleic (53%), and linolenic (8%). Oils high in monounsaturated fatty acids possess increased oxidative stability which negates the need for hydrogenation and eliminates the production of trans-fats. Food products containing trans-fatty acids are currently a major health concern and as such, soybean breeders and molecular geneticists have been challenged to efficiently alter the fatty acid composition to meet the specific needs of the various industries. Fatty acid desaturases are enzymes responsible for the insertion of double bonds (normally in the Z or cis conformation) into alkyl chains, following the abstraction of two hydrogen atoms. The physical properties and nutritional value of many animal and plant storage lipids are determined by desaturases. The quality of soybean oil is of paramount importance both economically and from a nutritional stand point and depends to a large extent on the ratio of polyunsaturated to monounsaturated fatty acid. Temperature also plays an important role in the final lipid content of oil seed plants. In this study, we designed and used gene-specific primers to the following fatty acid desaturases; stearoyl-ACP desaturase (SACPD), omega-6 fatty acid desaturase (FAD2-1), and omega-3 fatty acid desaturase (FAD3), to characterize soybean varieties. We further determined the effect of temperature on the expression of these desaturase genes by quantifying transcript accumulation at various.

In contrast, transformation approaches for sunflower, the fourth most important oil crop, have only been occasionally used in basic research and no commercial transgenic varieties have been released. In part, this situation is due to the lack of reliable and efficient regeneration and transformation methods for sunflower. Using a well-characterized soybean transformation system, a family of soybean promoters associated with early somatic embryo induction was characterized following introduction and tracking of GFP expression in embryogenic tissues and transgenic soybean plants. These promoters displayed tissue specific expression and were active in newly induced soybean somatic embryos. In sunflower, regeneration and transformation methods were studied and improved. Germination of sunflower seeds on a cytokinin-containing medium led to a 6-fold increase in shoot induction from leaf tissues. Use of a short pulse treatment of cotyledon tissue on a shoot induction medium gave more than a 14-fold increase in the numbers of well-developed shoots. A micrografting technique was developed for plant recovery from shoots with over 50% graft survival. Using sunflower cotyledons, a new Agrobacterium-mediated transformation procedure using low inoculum/long co-culture yielded 30x more transgenic shoots than the standard protocol using high inoculum/short co-culture. Expression analysis via qRT-PCR showed lower expression levels of genes related to plant defense response in the explants treated with low inoculum, suggesting that the low inoculum reduced the induction of plant defense, which impacts plant transformation and regeneration. The use of low inoculum/long co-culture has great potential for improving Agrobacterium-mediated transformation of other plant species.
