Book excerpt: In a clear exposition this inspirational book shows the in situ three-dimensional morphology of cerebellar neurons, intracortical circuits and synaptic connections that underpin the functioning of cerebellar neurons in networks. It brings together in one volume a new view of the three-dimensional cytoarchitectonic arrangement of the cerebellar cortex. The book shows the cerebellar extrinsic and intrinsic intracortical circuits formed by mossy and climbing fibers as exposed by the cryofracture methods. The high degree of lateral collateralization of these fibers is also displayed providing new insights on the information processing in the cerebellar cortex. Besides, field emission high resolution electron microscopy shows its potential contribution to the study of synaptic morphology. The concluding chapter deals with the contribution of scanning electron microscopy to cerebellar neurobiology. This monograph is an authoritative survey and a must for anyone who is interested in the structure of the central nervous system. It will also appeal to an interdisciplinary audience who wants to learn more about electron microscopy and neurocytology.

Available in PDF, EPUB and Kindle. Book excerpt: The origins of this book go back to the first electron microscopic studies of the central nervous system. The cerebellar cortex was from the first an object of close study in the electron microscope, repeating in modern cytology and neuroanatomy the role it had in the hands of RAMON y CAJAL at the end of the nineteenth century. The senior author vividly remembers a day early in 1953 when GEORGE PALADE, with whom he was then working, showed him an electron micrograph of a cerebellar glomerulus, saying "That is what the synapse should look like. " It is true that the tissue was swollen and the mitochondria were exploded, but all of the essentials of synaptic structure were visible. At that time small fragments of tissue, fixed by immersion in osmium tetroxide and embedded in methacrylate, were laboriously sectioned with glass knives without any predetermined orientation and then examined in the electron microscope. After much searching, favorably preserved areas' were studied at the cytological level in order to recognize the parts of neurons and characterize them. Such procedures, dependent upon random sections and uncontrollable selection by a highly erratic technique of preservation, precluded any systematic investigation of the organization of a particular nucleus or region of the central nervous system. It was difficult enough to distinguish neurons from the neuroglia.

Available in PDF, EPUB and Kindle. Book excerpt: Research in the morphology-angioarchitecture and ultrastructure-of cerebral veins has been widely neglected in past decades; investigation was mainly focussed on the arterial side of brain circulation. This circumstance has certainly had a negative impact on the development of knowledge in clinical medicine about cerebral venous disease. Cerebra} venous pathology and its consequence is, however, a frequent problern in clinical neurosur gery, both with regard to operative techniques and conservative manage ment. Therefore, it is not surprising that the initiative to collect, for the first time, data on our present knowledge in basic research of cerebral veins, their structure and function under normal and pathological circumstances, came from clinicians. Regarding the cerebral veins the clinician has primarily in view the dysfunctions originating from embryogenetic malformations, phlebitic obstruction, tumourous shunts, or traumatic lesions. But in addition to that, particular attention should be paid to the microstructure ofthe venous vessel walls, their barrier function, and the venous vasomotor system. Studying these interrelationships has for a long time been both fascinating and of immediate interest to me.

Available in PDF, EPUB and Kindle. Book excerpt: Our knowledge of cerebellar functions and cerebellar disorders, called ataxias, is increasing considerably. Studies of the cerebellum are now a central focus in neuroscience. During the last four decades, many laboratories worldwide have dedicated their research activities to understanding the roles of the cerebellum in motor control, cognitive processes and biology of mental processes, behavioral symptoms, and emotion. It is now accepted that the cerebellum acts as a cognitive operator in learning, perception, and attention. Moreover, major improvements in our assessment of in vivo cerebellar architecture using imaging techniques have occurred. A typical example is the accurate description of cerebellar anatomy during fetal development with MRI, a progress which has direct impacts on patient care. These advances have been associated with discoveries of new clinical disorders, in particular in the field of genetic ataxias. More than 20 new genes have been identified these last 10 years. Only for dominant ataxias, more than 30 diseases have now been unravelled. The number of ataxic disorders will increase with aging, the cerebellum being the structure of the brain with the most important loss of neurons with age. More than 300 different cerebellar disorders are encountered during daily practice, but we are missing a single source of information explaining their pathogenesis. Despite the immense amount of knowledge acquired about the cerebellar circuitry these last years, a large book covering the neuroscience of the cerebellum is missing. The goal of this endeavour is to bring up to date information relevant for basic science and also for clinical activities. To reach this goal, the most renowned authors are gathered in a unique and in-depth book with a format of a handbook. We emphasize the connections between molecular findings, imaging features, behavioural/neuropsychological aspects, and clinical implications.

Following on the heels of the revolution based on the Green Fluorescent Protein, refined genetically-encoded fluorescent reporters and genetic targeting strategies now enable optical recording of synaptic transmission in defined neuronal populations at speeds approaching the enviable temporal resolution of electrophysiology. Super-resolution light microscopy permits observation of synapses and their molecular machinery at sub-diffraction resolution. At the ultrastructural level, automated forms of electron microscopy, improvements in specimen fixation methods, and recent efforts to correlate data from light and electron micrographs now make the reconstruction of functional neural circuits a reality. Finally, the use of optogenetic actuators, such as channelrhodopsins, allows precise temporal and spatial manipulation of neuronal activity and is revealing profound insights into the organization of neural circuits and their roles in behavior. This research topic highlights recent advances in both light and electron microscopy, with a specific focus on approaches that combine innovations from several different fields to obtain novel information about synapse structure and function. We are confident that this collection of articles - three original research papers, six reviews, one methods paper and one perspective article - will enable neuroscientists to achieve the next generation of experiments aimed at cracking the neural code.
