Available in PDF, EPUB and Kindle. Book excerpt: This new text highlights the value of this biological system as a research and teaching tool. The book is a sequel to the 1983 edition and is organized into 6 major sections: DNA metabolism, regulation of gene expression, morphogenesis, structure of selected proteins, host–phage interactions, and laboratory experiments in T4 molecular genetics. Since T4 has played a central role in the development of molecular biology as an academic discipline, the themes presented in this book provide a framework for designing graduate and undergraduate courses in prokaryotic genetics and biochemistry.

Book excerpt: The motivation for us to conceive this series of volumes on regulation was mainly our belief that it would be fun, and at the same time productive, to approach the subject in a way that differs from that of other treatises. We thought it might be interesting and instructive for both author and reader-to examine a particular area of investigation in a framework of many different problems. Cutting across the traditional boundaries that have separated the subjects in past volumes on regulation is not an easy thing to do-not because it is difficult to think of what interesting topics should replace the old ones, but because it is difficult to find authors who are willing to write about areas outside those pursued in their own laboratories. Anyone who takes on the task of reviewing a broad area of interest must weave together its various parts by picking up the threads from many different laboratories, and attempt to produce a fabric with a meaningful design. Finding persons who are likely to succeed in such a task was the most difficult part of our job. In the first volume of this treatise, most of the chapters dealt with the mechanisms of The second volume involved a somewhat regulation of gene expression in microorganisms. broader area, spanning the prokaryotic-eukaryotic border. Topics ranged from phage mor phogenesis to the role of gradients in development. The last volume-Volume 3A-con cerned hormones, as does this volume-Volume 3B.

Book excerpt: Many potential applications of synthetic and systems biology are relevant to the challenges associated with the detection, surveillance, and responses to emerging and re-emerging infectious diseases. On March 14 and 15, 2011, the Institute of Medicine's (IOM's) Forum on Microbial Threats convened a public workshop in Washington, DC, to explore the current state of the science of synthetic biology, including its dependency on systems biology; discussed the different approaches that scientists are taking to engineer, or reengineer, biological systems; and discussed how the tools and approaches of synthetic and systems biology were being applied to mitigate the risks associated with emerging infectious diseases. The Science and Applications of Synthetic and Systems Biology is organized into sections as a topic-by-topic distillation of the presentations and discussions that took place at the workshop. Its purpose is to present information from relevant experience, to delineate a range of pivotal issues and their respective challenges, and to offer differing perspectives on the topic as discussed and described by the workshop participants. This report also includes a collection of individually authored papers and commentary.

Available in PDF, EPUB and Kindle. Book excerpt: Molecular Action of Toxins and Viruses investigates the molecular action of bacterial toxins and viruses and its striking similarity to the mechanisms by which many neural and hormonal stimuli control normal cell functions. Topics covered include the biological activity of diphteria toxin; the role of cholera toxin in the regulation of hormone-sensitive adenylate cyclase; toxic lectins and related proteins; and bacterial cytolysins (membrane-damaging toxins). Comprised of nine chapters, this volume begins with a discussion on the biosynthesis and biological activity of diphtheria toxin, toxin-resistant mutant cells, and the entry of toxin into cells and fragment A in the cytoplasm of living cells. The reader is then systematically introduced to the use of cholera toxin as a probe to study the regulation and interaction of adenylate cyclase components; the toxic action of lectins and related proteins such as abrin, ricin, and modeccin; and the ability of bacterial cytolysins to damage cell membranes. The remaining chapters focus on the mechanism of action of colicin E2, colicin E3, and cloacin DF13; similarities in the action of different bacterial toxins; the role of cell membranes in infection with bacterial viruses and colicins; and the life cycle and infection mechanisms of bacteriophage T4. The book concludes with an analysis of the involvement of protein kinases in viral transformation. This monograph will be of interest to students, practitioners, and researchers in fields ranging from molecular biology and biochemistry to cell biology, bacteriology, and physiology.

Available in PDF, EPUB and Kindle. Book excerpt: It has been 10 years since Plenum included a series of reviews on bacte riophages, in Comprehensive Virology. Chapters in that series contained physical-genetic maps but very little DNA sequence information. Now the complete DNA sequence is known for some phages, and the se quences for others will soon follow. During the past 10 years two phages have come into common use as reagents: A phage for cloning single copies of genes, and Ml3 for cloning and DNA sequencing by the dideoxy termi nation method. Also during that period the use of alternative sigma fac tors by RNA polymerase has become established for SPOl and T4. This seems to be a widely used mechanism in bacteria, since it has been implicated in sporulation, heat shock response, and regulation of nitro gen metabolism. The control of transcription by the binding of A phage CII protein to the -35 region of the promoter is a recent finding, and it is not known how widespread this mechanism may be. This rapid progress made me eager to solicit a new series of reviews. These contributions are of two types. Each of the first type deals with an issue that is exemplified by many kinds of phages; chapters of this type should be useful in teaching advanced courses. Chapters of the second type provide comprehensive pictures of individual phage families and should provide valuable information for use in planning experiments.

Available in PDF, EPUB and Kindle. Book excerpt: The time seems ripe for a critical compendium of that segment of the biological universe we call viruses. Virology, as a science, having passed only recently through its descriptive phase of naming and num bering, has probably reached that stage at which relatively few new~ truly new~viruses will be discovered. Triggered by the intellectual probes and techniques of molecular biology, genetics, biochemical cytology, and high-resolution microscopy and spectroscopy, the field has experienced a genuine information explosion. Few serious attempts have been made to chronicle these events. This comprehensive series, which will comprise some 6000 pages in a total of about 22 volumes, represents a commitment by a large group of active investigators to analyze, digest, and expostulate on the great mass of data relating to viruses, much of which is now amorphous and disjointed, and scattered throughout a wide literature. In this way, we hope to place the entire field in perspective, and to develop an invalu able reference and sourcebook for researchers and students at all levels. This series is designed as a continuum that can be entered anywhere, but which also provides a logical progression of developing facts and integrated concepts.

Available in PDF, EPUB and Kindle. Book excerpt: This authoritative, timely, and comprehensively referenced compendium on the bacteriophages explores current views of how viruses infect bacteria. In combination with classical phage molecular genetics, new structural, genomic, and single-molecule technologies have rendered an explosion in our knowledge of phages. Bacteriophages, the most abundant and genetically diverse type of organism in the biosphere, were discovered at the beginning of the 20th century and enjoyed decades of used as anti-bacterial agents before being eclipsed by the antibiotic era. Since 1988, phages have come back into the spotlight as major factors in pathogenesis, bacterial evolution, and ecology. This book reveals their compelling elegence of function and their almost inconceivable diversity. Much of the founding work in molecular biology and structural biology was done on bacteriophages. These are widely used in molecular biology research and in biotechnology, as probes and markers, and in the popular method of assesing gene expression.
