Available in PDF, EPUB and Kindle. Book excerpt: The purpose of the first four volumes of the Handbook of Genetics is to bring together collections of relatively short, authoritative essays or an notated compilations of data on topics of significance to geneticists. Many of the essays will deal with various aspects of the biology of certain species or species groups selected because they are favorite subjects for genetic investigation in nature or the laboratory. Often there will be an encyclo pedic amount of information available on such species, with new papers appearing daily. Most of these will be written for specialists in a jargon that is bewildering to a novice, and sometimes even to a veteran geneticist working with evolutionarily distant organisms. For such readers what is needed is a written introduction to the morphology, life cycle, reproductive behavior, and culture methods for the species in question. What are its particular advantages (and disadvantages) for genetic study, and what have we learned from it? Where are the classic papers, the key bibli ographies, and how does one get stocks of wild type or mutant strains? Lists giving the symbolism and descriptions for selected mutants that have been retained and are thus available for future studies are provided whenever possible. Genetic and cytological maps, mitotic karyotypes, and haploid DNA values are also included when available. Volume 4 deals with certain vertebrate species that have been studied in considerable detail from the standpoint of genetics or molecular cytogenetics. Such data are available for only a relatively few vertebrates.

Available in PDF, EPUB and Kindle. Book excerpt: The impetus for compiling this book was the recent development of culture strains of neuroblastoma and glial cells and the immediate and enthusiastic way they have been taken up as model systems. After the first sudden rush of activity, it seems appropriate to pause, to assess progress, and to contemplate the future contributions that may be possible using these culture techniques. Long before the advent of established strains, cultures of nervous tissue had already contributed to neurobiology. Ross Harrison, in 1906, in a single experimental series, established tissue culture as a promising new technique in cell biology and settled the Golgi-Cajal controversy as to whether axonic processes originated as outgrowths from the cell body or were formed first in the intercellular spaces and were later connected to the cell body. Harrison observed process growth from nerve cells in cultures, thus settling the matter in favor of Cajal. Of great importance to neurobiology is the discovery by Rita Levi-Montalcini of nerve growth factor. Cultures of spinal ganglia played a major role in the discovery, isolation, and characterization of the factor (Levi-Montalcini et ai. , 1954). In my opinion, this discovery, although very well known, has not yet been adequately recognized for its germinal influence on neurobiology and embryology. Progress since the advent of clonal cultures has been more modest. I would like to cite two pieces of work which emphasize the technical ad vantages of these cultures.

Available in PDF, EPUB and Kindle. Book excerpt: Since its discovery in I960, the hybridization of somatic cells has evolved from a biological curiosity into an analytical method that today underlies nearly all investigations of the genetic aspects of various biological phenomena. As an eyewitness to this development from its inception forward, Boris Ephrussi here relates the history of somatic hybridization and the formation of its methodology, lie follows with a discussion of the characteristics and properties of the resultant hybrid cells. Together, these topics comprise an authoritative introduction to the principles of the technique. Dr. Ephrussi proceeds to an examination in greater detail of three specific areas of biological research to which the techniques of hybridization are currently being applied with promising consequences. Thus the major part of the book deals with applications of somatic hybridization to mammalian genetics, cell differentiation, and cancer. Originally published in 1972. The Princeton Legacy Library uses the latest print-on-demand technology to again make available previously out-of-print books from the distinguished backlist of Princeton University Press. These editions preserve the original texts of these important books while presenting them in durable paperback and hardcover editions. The goal of the Princeton Legacy Library is to vastly increase access to the rich scholarly heritage found in the thousands of books published by Princeton University Press since its founding in 1905.

Available in PDF, EPUB and Kindle. Book excerpt: There is no commonly accepted mechanism to explain differentiation of either normal or neoplastic cells. Despite this fact, the organizers of the 3 rd International Conference on Differentiation recognized that there is much emerging evidence which supports the view that both normal cells and many cancer cells share common differentiative processes. Accordingly, the organizers perceived that clinical scientists and developmental biologists would greatly benefit by together considering differentiation. In that way, developmental biologists would be apprised of recent insights in cancer cell biology and the physician scientist would be updated on events in developmental biology and both would gain new understanding of the cell biology of neoplasia. A specific example may reveal the potential value of developmental biologists interacting with cancer physicians. An example chosen at random suggests that probably any paper included in the symposium volume would serve the purpose. Dr. Stephen Subtelny reviewed recent studies by his laboratory concerning germ cell migration and replication in frog embryos. How might those results interest the cancer scientist? Dr. Subtelny showed that primordial germ cells of a fertile graft will reverse their migratory direction and move into a sterile host. Perhaps in this context it would not be inappropriate to state that the germ cells of the graft metastasized into the host. Germ cells from grafts of a different species will populate the previously sterile host gonad.

Available in PDF, EPUB and Kindle. Book excerpt: Growth, Nutrition, and Metabolism of Cells in Culture, Volume 1, summarizes the state of knowledge of the growth, nutrition, and metabolism of various types of cell cultures. The chapters are both detailed and comprehensive enough for the specialist and broad enough to provide a general background for the nonspecialist. The present volume discusses the uptake, synthesis, and degradation of biologically important compounds, particularly the major components usually present in tissue culture medium. The book begins by tracing the history of the development of tissue culture. This is followed by separate chapters on early development of cell culture nutrition; the biological effects of serum; the energy metabolism of malignant cells; the gaseous environment of the mammalian cell in culture; and the uptake and utilization of amino acids by cells in culture. Subsequent chapters cover purine and pyrimidine metabolism; lipids in cell culture; the use of cell cultures for sterol metabolism studies; the genetic expressions of human diploid fibroblast cell cultures; and structural features of mammalian complex carbohydrates.

Book excerpt: This volume covers all aspects of melanin pigmentation, providing a concise, comprehensive picture of new knowledge gained at the frontiers of research. It draws heavily on the author's 30-year activity in the field and his continuing work with specialists of widely diverse disciplines. The core of the volume deals with the structure, physicochemical properties, and biosynthesis of the major classes of melanin pigments, including neuromelanins. Further discussions include the biology of the various types of pigment-producing cells, the structure and mode of action of tyrosinase, and the chemistry of urinary melanogens and their biomedical applications as metabolic markers of melanocyte activity, especially for the follow-up of malignant melanoma. Finally, the volume considers progress in the photobiology and photochemistry of melanins, with special emphasis on the controversial role of these pigments in skin photoprotection. Melanins and Melanogenesis is ideally suited as a basic guide for newcomers, and a handy source of specific information for practitioners in academic, medical, and industrial settings.

Book excerpt: Many potential applications of synthetic and systems biology are relevant to the challenges associated with the detection, surveillance, and responses to emerging and re-emerging infectious diseases. On March 14 and 15, 2011, the Institute of Medicine's (IOM's) Forum on Microbial Threats convened a public workshop in Washington, DC, to explore the current state of the science of synthetic biology, including its dependency on systems biology; discussed the different approaches that scientists are taking to engineer, or reengineer, biological systems; and discussed how the tools and approaches of synthetic and systems biology were being applied to mitigate the risks associated with emerging infectious diseases. The Science and Applications of Synthetic and Systems Biology is organized into sections as a topic-by-topic distillation of the presentations and discussions that took place at the workshop. Its purpose is to present information from relevant experience, to delineate a range of pivotal issues and their respective challenges, and to offer differing perspectives on the topic as discussed and described by the workshop participants. This report also includes a collection of individually authored papers and commentary.
