Book excerpt: an attempt to rationalize these terminological and conceptual difficulties we have considered the origins of mammalian heat production from two different points of view. The scheme depicted in Fig. 1. 1 illustrates the fate of energy in the body as seen by the nutritionist. After allowing for losses of energy in faeces and urine, the metabolizable energy obtained from food is utilized for main taining and increasing body energy content (maintenance, external work, growth and production). The transformation of metabolizable energy into these forms of net energy also involves inevitable energy losses in the form of heat - thermic energy. Similarly, maintaining homeothermy in cold en vironments involves shivering and non-shivering thermogenesis (NST) and the energy costs of assimilating nutrients and retaining net energy results in obligatory heat losses due to diet-induced thermogenesis (DIT). This obligatory DIT is mainly due to the energy cost of protein and fat synthesis but, in addition to this, there is an adaptive component of DIT that helps maintain body energy content (i. e. body weight) by dissipating the metabolizable energy consumed in excess of the requirements for maintenance, growth and production. These are now reproduced in this book in exactly the same form and order in which they were presented at the Symposium, in the hope of providing a bird's-eye view of the various facets of thermogenesis research. A "mini-review", summarizing the current knowledge in each domain, therefore precedes the corresponding papers. The editors are deeply indebted to the contributors of these introductory lectures for so competently selecting from the mass of available information in each field, only the most essential elements. Book excerpt: Adaptation to altitude hypoxia is characterized by a variety offunctional changes which collectively facilitate oxygen trans port from the ambient medium to the cells of the body. All of these changes can be seen at one time or another in the course of hypoxic exposure. Yet, as already stressed (Hannon and Vogel, 1977), an examination of the literature gives only a sketchy and often conflicting picture of the exact nature of these changes and how they interact as a function of exposure duration. This is partly because of the limited number of variables explored in a given study, but it is also attributable to differences in experimental design, differences among species in susceptibility to hypoxia, nonstandardized experimental conditions, lack of proper control of physical (e. g. , temperature) and physiological variables (e. g. , body mass), failure to take measurements at key periods of exposure, and gaps in knowledge about some fundamental mechanisms. Furthermore the available data on animals native to high altitude are meager and/or inconclusive. Extensive further work under well-controlled experimental conditions is required before a detailed picture can be made. Nevertheless, it has been a guiding principle in the prepara tion of this monograph rather to summarize the vastly dis persed material that constitutes the comparative physiology of adaptation to high altitude into a coherent picture, than to provide a comprehensive survey of the field.

Book excerpt: Cyclic nucleotides are intimately involved in the consequences of either stimulation or blockade of receptors; therefore, an understanding of the biochemistry of cyclic nucleotides ought to be important for pharmacologists. Pharmacology is a science that among other things investigates chemical compounds that affect the physiology of cells, tissues and organs. Frequently pharmacologists account for the effect of low concentrations of a drug upon a tissue by invoking the presence of a receptor upon the surface of the cell. Traditional pharmacologists excelled at identifying and classifying the properties of receptors. A. J. CLARK'S monograph in the earlier series of the Handbook of Experimental Pharmacology (CLARK 1937) summarized the mathematics underlying the traditional pharmacological approach towards receptors. By its nature, however, classic pharmacology provided little useful information about the intracellular events occurring as a consequence of occupying a receptor; for example, ALQUIST (1948) identified the beta-adrenocep tor, but he did not provide any insight into how stimulation of the receptor produces tissue-specific physiological responses. The discovery of cyclic AMP by RALL and SUTHERLAND (see RALL, Vol. I) led to biochemical investigations of many different receptors (including ALQUIST'S beta-adrenoceptor) that share a cyclic nucleotide as a common factor in the biochemical mechanisms that translate the occupancy of receptors into physiological effects. Ten years ago, in the introduction to their monograph on cyclic nucleotides, ROBISON et al. (1971) commented on the rapid growth of interest in cyclic nucleotides over the preceding years.
