[I] showed a dependence on pressure gradient and pore diameter that, at the time of publication, did not permit an easy interpretation of the hemolytic mechanism. Acting on the assumption that thresholds of hemolysis are easier to correlate with physical forces than extents of hemolysis, we performed a series of experi ments repeating some of the conditions reported in [I] and then focusing on low L1P in order to define better the thresholds of hemolysis for several pore sizes. Employing a model of a deformed red cell shape at the pore entrance (based on micropipette observations) we related the force field in the fluid to a biaxial tension in the membrane. The threshold for lysis correlated with a membrane tension of 30 dynes/cm. This quantity is in agreement with lysis data from a number of other investigators employing a variety of mechanisms for introduc ing membrane tension. The sequence of events represented here is: a. Fluid forces and pressure gradients deform the cell into a new, elongated shape. b. Extent of deformation becomes limited by the resistance of the cell mem brane to undergo an increase in area. c. Fluid forces and pressure gradients acting on the deformed cell membrane cause an increase in biaxial tension in the membrane. d. When the strain caused by this tension causes pores to open in the membrane, the threshold for hemolysis has been reached [2].

Book excerpt: In the third Erlangen-Nuremberg symposium on experimental geronto logy we discussed the effect of aging on blood cells and rheology. Most studies with red cells have been performed on humans, rats, and rabbits. All these erythrocytes have no nucleus; the cells are therefore a model in which new synthesis of proteins is impossible. All the processes are age dependent except for exchange with the plasma. Investigations of red cells have attracted attention because of the latter's availability in large quantities from biochemists, physiologists, immunologists, and micro scopists. We now have a relative abundance of data on the biochemical and immunologic composition as well as on the ultrastructure of the cells. The second topic under discussion at the symposium was the immune system and cell receptors. One of the most apparent features of aging of the organism is progressive dysregulation of the immune system. As a result, the incidence of infection, cancer, and autoimmune and immune complex diseases increases. While we have a lot of new data on the function and differentiation of lymphoid cells, we have little informa tion on lymphocyte receptors during the aging process. Rheologic studies reflect changes in red cell deformability and blood coagulation. The erythrocyte deformability influences blood flow in all parts of the circulation. The fluidity of the red cell membranes controls whole blood viscosity. During aging, there are significant alterations in blood coagulation and hemostasis. One of the best known modifications of blood co agulation is a typical thrombophilic tendency with aging.

Book excerpt: Red blood cells (RBCs) are the most abundant cells in human blood, representing 40 to 45% of the blood volume (hematocrit). These cells have the particular ability to deform and bridge together to form aggregates under very low shear rates. The theory and mechanics behind aggregation are, however, not yet completely understood. The purpose of this work is to provide a novel method to analyze, understand and mimic blood behaviour in microcirculation. The main objective is to develop a methodology to quantify and characterize RBC aggregates and hence enhance the current understanding of the non-Newtonian behaviour of blood at the microscale. For this purpose, suspensions of porcine blood and human blood are tested in vitro in a Poly-di-methylsiloxane (PDMS) microchannel to characterize RBC aggregates within these two types of blood. These microchannels are fabricated using standard photolithography methods. Experiments are performed using a micro Particle Image Velocimetry ( PIV) system for shear rate measurements coupled with a high speed camera for the flow visualization. Corresponding numerical simulations are conducted using a research Computational Fluid Dynamic (CFD) solver, Nek5000, based on the spectral element method solution to the incompressible non-Newtonian Navier-Stokes equations. RBC aggregate sizes are quantified in controlled and measurable shear rate environments for 5, 10 and 15% hematocrit. Aggregate sizes are determined using image processing techniques. Velocity fields of the blood flow are measured experimentally and compared to numerical simulations using simple non-Newtonian models (Power law and Carreau models). This work establishes for the first time a relationship between RBC aggregate sizes and corresponding shear rates in a microfluidic environment as well as one between RBC aggregate sizes and apparent blood viscosity at body temperature in a microfluidic controlled environment. The results of the investigation can be used to help develop new numerical models for non-Newtonian blood flow, provide a better understanding of the mechanics of RBC aggregation and help determine aggregate behaviour in clinical settings such as for degenerative diseases like diabetes and heart disease.

The first such London Workshop was held on the occasion of the Second European Conference on Clinical Haemorheology in 1981 and its con clusions were published in an abbreviated form. The interest in blood filtration as a technique for assessing red cell deformability has continued to increase and there are now several hundred centres around the world using some form of filtration. It was, therefore, felt that the informal detailed discussions held by a small invited group of experts from many countries would be of more general interest and therefore worth publishing. This has been made possible by the continued interest and generous support of Hoechst. Spontaneous open discussion rather than formal presentations has been the hallmark of these workshops, with precirculation of all scientific data to the participants. We have attempted to preserve this flavour in editing the transcript of the proceedings. This, together with our anxiety for rapid publication, may have resulted in a less than finely polished text which may contain some in accuracies. The topics discussed were loosely structured into eight sessions, outlined in the accompanying programme.
